Efficient control of Xanthomonas axonopodis pv. dieffenbachiae, the causal agent of anthurium bacterial blight, requires sensitive and reliable diagnostic tools. The European standard EN ISO 16140:2003 has been followed to compare a nested PCR assay (N‐PCR) to a reference method (isolation and serological identification of bacterial colonies) and to other alternative serological detection methods. The evaluation was performed in two steps: a comparative study and a collaborative study involving 15 European laboratories. Although inclusivity was maximal (100%) for all methods, a maximal exclusivity was obtained only with N‐PCR followed by an enzymatic restriction digestion of the amplicons. Exclusivity indices of 90·6, 88·7 and 47·2% were found for indirect ELISA, immunofluorescence and double antibody sandwich ELISA, respectively. An exclusivity of 92·5% was obtained with the reference method, further increased to 100% if pathogenicity tests were performed as a supplemental assay. The best level of sensitivity (relative detection level) was obtained with the reference method followed by the N‐PCR assay. The N‐PCR performance in terms of relative accuracy, accordance and concordance was very similar to that of the reference method. Moreover, N‐PCR had undeniable advantages compared to the reference method (less labour‐intensive and less time‐consuming). In addition, post‐test probabilities of infection were calculated to select the most appropriate detection scheme related to the prevalence of the pathogen. The N‐PCR assay has since been included in a revised version of the EPPO detection protocol.  相似文献

Through genetic selection and improvement of environment, litter size of sows increases. Increased energy requirement during lactation, increases the risk of excessive mobilization from body stores, with detrimental effects on reproductive performance. Feed intake capacity tends to decrease due to selection towards leaner pigs with a lower feed conversion ratio. However, to facilitate sows to wean large litters extra feed intake, or even better, a higher feed efficiency during lactation would be favourable. The objective of the present study was to describe the dynamics of body composition of sows and piglets during lactation, and to relate these traits to a newly introduced trait called “lactation efficiency”. Energy metabolism of lactating sows was described, based on on-farm observations of weight and backfat of sows before parturition and at weaning, weight of piglets at birth and at weaning and feed intake of sows during lactation. “Lactation efficiency” was defined as energy efficiency of sows, and calculated for individual sows at two different farms. The average lactation efficiency was 68% and 65% for both farms; meaning that 68 and 65% of the metabolisable energy through feed intake or mobilization from body stores, above maintenance of the sow (input), was used for piglet growth and piglet maintenance (output). The association between lactation efficiency and other reproductive traits was studied by estimating the correlations within farms. Sows with a higher lactation efficiency showed lower feed intake (r = − 0.27 and r = − 0.35 for both farms respectively) and smaller fat losses (r = − 0.34 and r = − 0.29, respectively). The energy output of efficient sows was slightly higher (r = 0.23 and r = 0.30). The more efficient sows were the better mothers, as mortality of their piglets was lower (r = − 0.12 and r = − 0.16), piglet growth rate was higher (r = 0.16 and r = 0.23), and at weaning their litters were less variable (r = − 0.08; only available at one farm). Results were remarkably similar for the two farms, despite different feeding strategies. Extra input, by means of feed intake or mobilization from body stores generated extra output by means of litter weight at weaning. This experiment demonstrated that an accurate recording of energy metabolism and relevant reproduction traits with little intervention is possible on commercial farms.  相似文献

This paper describes a comparison study of test methods and supports the use of real‐time polymerase chain reaction (PCR) for the detection of Clavibacter michiganensis subsp. sepedonicus and Ralstonia solanacearum in potato tubers in routine testing. These 2 bacteria are quarantine organisms under European Union (EU) regulatory control and testing for (latent) infections of these bacteria in seed potatoes is mandatory. Real‐time PCR tests were performed on 276 routine potato tuber samples, including samples infected with either C. michiganensis subsp. sepedonicus or R. solanacearum, and the performance of these real‐time PCR tests was compared with that of immunofluorescence (IF). Real‐time PCR tests, using different primer sets and extraction and PCR protocols, proved to be sensitive and specific for the detection of C. michiganensis subsp. sepedonicus and R. solanacearum in potato tubers in routine testing, and performed at least as well as IF. Real‐time PCR is a good addition to the detection protocols as laid down in EU regulations (EU Council Directives 2006/56/EC and 2006/63/EC).  相似文献

The effects of birth weight (BiW; low BiW [LBW] vs. high BiW [HBW]) and estimated breeding value (EBV) for protein deposition (low EBV [LBV] vs. high EBV [HBV]) on N retention, N efficiency, and concentrations of metabolites in plasma and urine related to N efficiency in growing pigs were studied. At an age of 14 wk, 10 LBW–LBV (BiW: 1.07 ± 0.09 [SD] kg; EBV: −2.52 ± 3.97 g/d, compared with an average crossbred pig with a protein deposition of 165 g/d), 10 LBW–HBV (BiW: 1.02 ± 0.13 kg; EBV: 10.47 ± 4.26 g/d), 10 HBW–LBV (BiW: 1.80 ± 0.13 kg; EBV: −2.15 ± 2.28 g/d), and 10 HBW–HBV (BiW: 1.80 ± 0.15 kg; EBV: 11.18 ± 3.68 g/d) male growing pigs were allotted to the experiment. The pigs were individually housed in metabolism cages and were subjected to an N balance study in two sequential periods of 5 d, after an 11-d dietary adaptation period. Pigs were assigned to a protein adequate (A) or protein restricted (R, 70% of A) regime in a change-over design. Pigs were fed 2.8 times the energy requirements for maintenance. Nontargeted metabolomics analyses were performed in urine and blood plasma samples. The N retention (in g/d) was higher in the HBW than in the LBW pigs (P < 0.001). The N retention (in g/[kg metabolic body weight (BW0.75) · d]) and N efficiency, however, were not affected by the BiW of the pigs. The N retention (P = 0.04) and N efficiency (P = 0.04) were higher in HBV than in LVB pigs on the A regime but were not affected by EBV in pigs on the R regime. Restricting the dietary protein supply with 30% decreased the N retention (P < 0.001) but increased the N efficiency (P = 0.003). Nontargeted metabolomics showed that a hexose, free amino acids (AA), and lysophosphatidylcholines were the most important metabolites in plasma for the discrimination between HBV and LBV pigs, whereas metabolites of microbial origin contributed to the discrimination between HBV and LBV pigs in urine. This study shows that BiW does not affect N efficiency in the later life of pigs. Nitrogen efficiency and N retention were higher in HBV than in LBV pigs on the A regime but similar in HBV and LBV pigs on the R regime. In precision feeding concepts aiming to further optimize protein and AA efficiency in pigs, the variation in EBV for protein deposition of pigs should be considered as a factor determining N retention, growth performance, and N efficiency.  相似文献

Microbiota play an important role in total tract nutrient digestion, especially when fibrous diets are fed to pigs. This study aimed to use metagenomics to predict faecal nutrient digestibility in grower-finisher pigs. The study design consisted of 160 three-way crossbreed grower-finisher pigs (80 female and 80 male) which were either fed a diet based on corn/soybean meal or a more fibrous diet based on wheat/barley/by-products. On the day before slaughter, faecal samples were collected and used to determine faecal digestibility of dry matter, ash, organic matter, crude protein, crude fat, crude fibre and non-starch polysaccharides. The faecal samples were also sequenced for the 16S hypervariable region of bacteria (V3/V4) to profile the faecal microbiome. With these data, we calculated the between-animal variation in faecal nutrient digestibility associated with variation in the faecal microbiome, that is the “microbiability”. The microbiability values were significantly greater than zero for dry matter, organic matter, crude protein, crude fibre and non-starch polysaccharides, ranging from 0.58 to 0.93, as well as for crude fat with a value of 0.37, but not significantly different from zero for ash. Using leave-one-out cross-validation, we estimated the accuracy of predicting digestibility values of individual pigs based on their faecal microbiota composition. The accuracies of prediction for crude fat and ash digestibility were virtually 0, and for the other nutrients, the accuracies ranged from 0.42 to 0.63. In conclusion, the faecal microbiota composition gave high microbiability values for faecal digestibility of dry matter, organic matter, crude protein, crude fibre and non-starch polysaccharides. The accuracies of prediction are relatively low if the interest is in precisely predicting faecal nutrient digestibility of individual pigs, but are promising from the perspective of ranking animals in a genetic selection context.  相似文献

A study was performed in order to assess the presence of Xylella fastidiosa in imported ornamental plants, among them Olea europaea, Coffea arabica and Nerium oleander. Positive results were only obtained from C. arabica, where 15 plant samples tested positive for X. fastidiosa by PCR, nine from Costa Rica and six from Honduras. Transmission electron microscopy observations indicated that rod‐shaped bacterial cells exhibiting the characteristics of X. fastidiosa cells were present in the xylem vessels of leaf petioles obtained from the infected C. arabica plants. Diversity of X. fastidiosa in C. arabica plants was assessed through a multilocus sequence typing (MLST) analysis of seven housekeeping genes (leuA, petC, lacF, cysG, holC, nuoL and gltT) and compared with X. fastidiosa infecting different host plants worldwide. Based on this MLST analysis, the prevalence of different sequence types (STs) of X. fastidiosa in the C. arabica ornamental plants was demonstrated and related to different X. fastidiosa subspecies, underlining the risk of introducing additional genetic diversity for X. fastidiosa to Europe. ST53, related to X. fastidiosa subsp. pauca, was frequently found in these C. arabica samples. A second ST related to X. fastidiosa subsp. pauca, ST73, has been assessed in coinfection with ST53 in one individual plant. Additionally, ST72 and ST76, related to X. fastidiosa subsp. fastidiosa, have been recorded. Next to these previously described STs, a novel ST, namely ST77 has been revealed, related to X. fastidiosa subsp. fastidiosa. Isolation of X. fastidiosa from leaf petioles and midribs of infected C. arabica plants was successfully performed only after the application of an additional ultrasonication step during the extraction procedure. Based on this approach, a number of X. fastidiosa isolates were obtained and further characterized.  相似文献

The aims of this study were, first, to evaluate the effects of climatic variables on daily feed intake of lactating sows and, second, to establish whether the response of sows to variation in temperature on feed intake during lactation was heritable. A total of 82,614 records for daily feed intake during lactation were available for 848 sows with 3,369 litters farrowing from January 2000 to December 2007. Climatic parameters available from the nearest weather station were maximum 24 h outside temperature, day length changes, and humidity. Although ambient room temperature was modified at the animal level in the farrowing shed, these climatic variables still had a significant effect on feed intake during lactation. Regression coefficients temperature and humidity were 0.01385 ± 0.00300 (temperature) - 0.00031 ± 0.00009 (temperature(2)) and 0.01443 ± 0.00620 (humidity) - 0.00009 ± 0.00004 (humidity(2)). There was an interaction between temperature and humidity, partly due to the climate control in the farrowing shed. At low temperature, feed intake increased considerably with greater humidity, in contrast to a small reduction in feed intake with greater humidity at high temperature. Day length change was modeled with a cosine function. At the start of autumn (September 21), sows ate 0.36 ± 0.056 kg/d less feed than at the start of spring (March 21). Daily feed intake during lactation was described as a function of days in lactation and as a function of both days in lactation and temperature using random regression models. The average heritability and repeatability summarized over the day in lactation at the mean temperature were 0.21 and 0.69, respectively. Genetic variance of temperature response on feed intake was less than 20% of the day effect. The permanent environmental variance was 2-fold (day) and 4-fold (temperature) greater than the corresponding additive genetic variance. Heritabilities of daily feed intake were greater during the first week of lactation compared with the rest of lactation. The genetic correlation between days decreased as time increased down to about 0.2 between the first and last day in lactation. The genetic correlation between feed intake records at the extreme temperatures decreased to about -0.35. It was concluded that random regression models are useful for research and results may be used to develop simpler models that can be implemented in practical breeding programs. An effect of temperature on lactation feed intake was found even in this climate-controlled environment located in a temperate climate zone. Larger effects are expected in more extreme climatic conditions with less temperature-controlled farrowing sheds.  相似文献

The relationship between placental efficiency (PLEFF, i.e., the ratio of birth weight [BWB] to placental weight [PLW]) and neonatal pig vitality as measured by the probability of preweaning death of live born piglets was examined for 1,036 live born piglets of 118 litters. The data were first analyzed to establish whether the relationships between PLEFF, PLW, and BWB were affected by parity (first vs. higher). Furthermore, the data collected were used to establish whether PLEFF is a better predictor of the risk of neonatal pig mortality before weaning than BWB and PLW. The relationships of BWB to PLW and PLEFF to PLW differed (P <0.01 and P <0.05, respectively) between piglets from gilts and sows. This difference appeared to be due mainly to an additional population of piglets with very large placentas in sows that were not present in gilts. Despite being significant, the courses of the relationships were essentially similar for piglets in gilts and in sows. For the curvilinear relationship of BWB to PLW, up to a certain threshold value, an increase of PLW resulted in an increase in BWB, and thereafter BWB did not change. A consequence of this is that PLEFF at relatively high PLW does not give the same information as PLEFF at relatively low PLW. For the second-order relationship of PLEFF to BWB, PLEFF increased with an increase in BWB, until BWB = 1,657 g, and decreased thereafter. The PLEFF decreased linearly with PLW. A change in PLW had a much larger impact on the value of PLEFF than a change in BWB. Although BWB and PLW were negatively associated with the chance of dying before weaning (P <0.001 and P <0.01, respectively), only PLEFF tended to be negatively associated with the chance of dying only before weaning (P = 0.08). Its underlying trait, BWB, played a greater role on the effect of PLEFF on the chance of preweaning death than PLW. In conclusion, PLEFF in swine is a complicated trait that should be treated with care. It is merely a mathematical derivative of BWB and PLW, whereby the extent to which BWB depends on PLW depends on the value of PLW. Placental functioning and fetal growth capacity, however, also have their effects on the value of BWB. It is concluded that, of the three traits (BWB, PLW, and PLEFF), the best predictor for the chance of preweaning mortality, which also happens to be easiest to measure, remains BWB.  相似文献

The increased productivity of sows increases the risk of a more pronounced negative energy balance during lactation. One possibility to prevent this is to increase the lactation efficiency (LE) genetically and thereby increase milk output for a given feed intake and mobilization of body tissue. The benefits of selection for LE depend on its heritability and the relationships with other traits of interest. The objectives of this study were to estimate genetic parameters for LE, its underlying traits, and to predict the consequences of current selection strategies in dam lines. Data from 4 farms were available to estimate genetic parameters. Heritabilities were estimated by using a univariate repeatability model, and genetic correlations were estimated bivariately. Selection index theory was used to predict the genetic progress by 3 alternative breeding programs: 1) a breeding program that aimed at balanced progress in the total number of piglets born, piglet mortality, and percent prolonged interval from weaning to estrus; 2) extension of this breeding goal with LE; and 3) a breeding goal that included only one selection criterion, litter weight gain, to demonstrate the effect of indirect selection for milk production. The heritability for LE was low (0.12). Body fat mass (0.52) and BW (0.45) of sows at the beginning of lactation showed the greatest heritabilities. Protein mass at the beginning of lactation, protein loss, weight loss, and ad libitum feed intake during lactation showed moderate heritabilities (0.39, 0.21, 0.20, and 0.30, respectively). Low to moderate heritabilities were found for litter weight at birth, within-litter SD in the birth weight of piglets, litter weight gain, fat loss, and restricted feed intake during lactation (0.19, 0.09, 0.18, 0.05, and 0.14, respectively). Within-litter SD in the weaning weight of piglets showed no genetic variability. It was predicted that a breeding goal for dam lines with an emphasis on the total number of piglets born, piglet mortality, and percent prolonged interval from weaning to estrus would not dramatically change BW or body composition at the beginning of lactation, or mobilization of body tissue and feed intake during lactation. Inclusion of LE in the breeding goal will improve stayability, as defined by the first-litter survival of sows and LE itself, without negative consequences for other economically important traits. Nevertheless, it might be worthwhile to design a breeding goal in which LE increases and feed intake remains unchanged.  相似文献
