Establishment of European eel (Anguilla anguilla) hatchery production will rely on selectively bred individuals that produce progeny with the best traits in successive generations. As such, this study used a quantitative genetic breeding design, between four females and nine males (four wild‐caught and five cultured), to investigate the effect of paternal origin (wild‐caught vs. cultured) and quantify the relative importance of parental effects, including genetic compatibility, on early life history (ELH) performance traits (i.e. fertilization success, embryonic survival at 32 hr post‐fertilization, hatch success and larval deformities at 2 days post‐hatch) of European eel. Wild‐caught males had higher (56%) spermatocrit values than cultured males (45%), while fertilization success, embryonic survival, hatch success and larval deformities were not significantly impacted by paternal origin. This demonstrates that short‐term domestication of male eels does not negatively affect offspring quality and enables the consideration of cultured male broodstock in future breeding programmes. Moreover, paternity significantly explained 9.5% of the variability in embryonic survival, providing further evidence that paternal effects need to be taken into consideration in assisted reproduction protocols. Furthermore, maternity significantly explained 54.8% of the variation for fertilization success, 61.7% for embryonic survival, 88.1% for hatching success and 62.8% for larval deformities, validating that maternity is a major factor influencing these “critical” ELH traits. At last, the parental interaction explained 12.8% of the variation for fertilization success, 8.3% for embryonic survival, 4.5% for hatch success and 20.5% for larval deformities. Thus, we conclude that eggs of one female can develop more successfully when crossed with a compatible male, highlighting the importance of mate choice for successful propagation of high‐quality offspring. Together, this knowledge will improve early offspring performance, leading to future breeding programmes for this critically endangered and economically important species.  相似文献

Aquaculture production relies on controlled management of gametogenesis, especially in species where assisted reproduction is needed for obtaining gametes in captivity. The present study used human chorionic gonadotropin (hCG) treatments to induce and sustain spermatogenesis in European eel (Anguilla anguilla). The aim was to evaluate effects of strip-spawning timing (12 vs. 24 hr) after weekly administration of hCG and the necessity of a primer dose (in addition to weekly hormonal treatment) prior to strip spawning (primer vs. no-primer) on sperm quality parameters. Sperm parameters included milt production (weight), density and sperm kinematics at Week 9, 11 and 13 after onset of treatment. Spermiation commenced in 11.5% of males in Week 5 and by Week 9, and all males produced milt. Male weight, milt production, sperm density and spermatocrit did not differ among hormonal treatments during the experimental period. Overall, male weight decreased from 106.3 to 93.0 g, milt weight increased from 3.5 to 5.4 g, sperm density counts decreased from 11.7 × 109 to 10.5 × 109 cells/ml, and spermatocrit decreased from 46.5% to 40.5%. Furthermore, spermatocrit was positively related to haemocytometer counts (R2 = .86, p < .001), providing a reliable indicator of sperm density. Differences in sperm kinematics were observed depending on strip-spawning timing after hormonal injection (12 vs. 24 hr) but with no consistent pattern. These sperm quality parameters also did not consistently differ between the no-primer and primer treatments. Considering that each male may be stripped 4–5 times over the 2–3 months spawning season, omitting the primer would reduce animal handling, material costs and labour intensity, while sustaining high-quality sperm production.  相似文献

Photoelectrolytic decomposition of water with visible irradiation is demonstrated in a cell made of p-type gallium phosphide as the cathode and platinum as the anode. A maximum energy conversion efficiency of 0.1 percent is achieved with an external bias of 1.3 volts. Outbreaks of morbidity and mortality in double-crested cormorants (Phalacrocorax auritus) along Florida's Gulf Coast have occurred sporadically for at least 30 yr. During these outbreaks, the Clinic for the Rehabilitation of Wildlife, located on Sanibel Island in Florida, has admitted a substantial number of cormorants with consistent presentation of primarily neurologic clinical signs. In order to investigate the association of these outbreaks in cormorants with exposure to brevetoxin, we compared the timing of admittance of cormorants with outbreak-specific clinical signs to blooms of the brevetoxin-producing marine algae, Karenia brevis (formerly Gymnodinium breve), around Sanibel Island from 1995 through 1999. The clinic admitted 360 out of 613 cormorants with the common clinical sign of severe cerebellar ataxia in six outbreaks occurring during this period. The ataxia was characterized by a broad-based stance, truncal incoordination, hypermetric gait, and intention tremors of the head. The histopathologic findings in 10 cormorants euthanized in 1997 were mild and nonspecific. An immunohistochemical staining technique for the detection of brevetoxin in cormorants documented the uptake of brevetoxin in tissues from four cormorants admitted during an outbreak in 1997, but a modified technique used on samples from 11 cormorants admitted during a K. brevis bloom in 2000 produced indeterminate results. Admittance of cormorants with outbreak-specific clinical signs was positively correlated (P < 0.05) with concurrent concentrations of K. brevis in local water. The cross-correlation coefficient was also significant when increased K. brevis levels preceded cormorant admittances by 2, 4, 6, and 8 wk. This delay in time between K. brevis blooms and cormorant admittance and our clinical finding of neurologic abnormalities in cormorants without overt histopathologic features suggest an association between K. brevis blooms and local cormorant morbidity.  相似文献

The use of wood ash in forestry has been questioned because of the potential risk associated with its cadmium (Cd) content (1–30 mg kg–1). In agriculture, wood ash is only allowed for use as a fertilizer when its Cd content is below 3 mg kg–1. This restriction has not been applied to forest soils and there is a lack of knowledge about the potentially harmful effects of the Cd in wood ash on forest ecosystems. This paper summarizes our recent studies on the microbial communities of boreal coniferous forest humus exposed to Cd-containing wood ash treatment. The main objectives of our studies were to test if the Cd in wood ash has the potential to affect the humus layer microflora of coniferous upland forests and if it has the potential to enter the human food chain. These objectives were tested both in laboratory and field experiments with ash and ash spiked with Cd (in laboratory 400 or 1,000 mg Cd kg–1 as CdO or CdCl2; in field 400 mg Cd kg–1 as CdO). In one study the dissolution of ash was accelerated by irrigating it with simulated acid rain (SAR). Wood ash increased humus layer pH and microbial activities (respiration or thymidine incorporation rates) and changed its microfloral community structure (Biolog, PLFA, 16S or 18S rDNA PCR-DGGE) in both laboratory and field experiments. Spiking ash with Cd induced no further changes in the above-mentioned variables compared to ash alone. The Cd added with wood ash did not become bioavailable as detected with a bacterial biosensor Bacillus subtilis BR151(pTOO24). The form and level of Cd added in the ash had no further effect on the microbiological variables studied. Irrigation of ash with SAR did not increase the amount of bioavailable Cd, although the dissolution rate of the ash was increased. The concentration of Cd in soil water and in the berries of Vaccinium uliginosum and V. vitis-idaea, and the amount of humus bioavailable Cd did not increase with applied ash or ash spiked with Cd although the ash spiked with Cd increased the amount of humus total and extractable Cd in the 4-year field study. Only the ash spiked with Cd and not the unspiked normal wood ash resulted in significantly higher Cd concentrations in the mushroom Lactarius rufus and a slight increase in the berries of Empetrum nigrum (first year only). In conclusion, the Cd in wood ash did not become bioavailable and harmful to forest soil microbes, or leach into the humus layer even when treated with simulated acid rain. It is thus safe to use wood ash as a fertilizer in forests. However, since wood ash adds Cd to the environment, it is recommended that the same sites should not be fertilized with wood ash more than once. The effects of wood ash (3 t ha–1) on forest soil humus layer microbes are long-term, lasting at least 20 years, and probably longer if higher application dose and/or hardened ash is used.  相似文献

The concept of Woodland Key Habitats (WKH, small-scaled presumed hotspots of biodiversity) has become an essential component of biodiversity conservation in Fennoscandian and Baltic forests. There have been debates over the importance of WKHs in relation to the conservation of biodiversity in production forests. We applied a systematic review protocol and meta-analysis to summarize knowledge on comparisons of biodiversity qualities, such as dead wood and species richness, between WKHs and production forests in relevant countries. We also summarized the knowledge on the impact of edge effects by comparing WKHs surrounded by production forests to WKHs surrounded by clear cuts. Studies had been conducted in Finland, Norway and Sweden. Based on our meta-analysis, WKHs seem to be relative hotspots for dead wood volume, diversity of dead wood, number of species and number of red-listed species. There were some differences also between countries in these biodiversity qualities. Only two studies compared WKHs surrounded by production forests and clear cuts, respectively. Hence, the capability of WKHs to maintain their original species composition and support species persistence over time remains to be addressed, as well as their role in relation to other conservation tools.  相似文献

OBJECTIVE: To describe cardiac lesions and identify risk factors associated with myocarditis and dilated cardiomyopathy (DCM) in beach-cast southern sea otters. ANIMALS: Free-ranging southern sea otters. PROCEDURE: Sea otters were necropsied at the Marine Wildlife Veterinary Care and Research Center from 1998 through 2001. Microscopic and gross necropsy findings were used to classify sea otters as myocarditis or DCM case otters or control otters. Univariate, multivariate, and spatial analytical techniques were used to evaluate associations among myocarditis; DCM; common sea otter pathogens; and potential infectious, toxic, and nutritional causes. RESULTS: Clusters of sea otters with myocarditis and DCM were identified in the southern aspect of the sea otter range from May to November 2000. Risk factors for myocarditis included age, good body condition, and exposure to domoic acid and Sarcocystis neurona. Myocarditis associated with domoic acid occurred predominantly in the southern part of the range, whereas myocarditis associated with S. neurona occurred in the northern part of the range. Age and suspected previous exposure to domoic acid were identified as major risk factors for DCM. A sample of otters with DCM had significantly lower concentrations of myocardial L-carnitine than control and myocarditis case otters. CONCLUSIONS AND CLINICAL RELEVANCE: Cardiac disease is an important cause of death in southern sea otters. Domoic acid toxicosis and infection with S. neurona are likely to be 2 important causes of myocarditis in sea otters. Domoic acid-induced myocarditis appears to progress to DCM, and depletion of myocardial L-carnitine may play a key role in this pathogenesis.  相似文献

Due to economical and ecological reasons it is important to provide the necessary flexibility in fertilizer management to respond to differences in plant nutrients requirements. Plant-based sensors have potential to provide more accurate and on-line information regarding crop bio-responses to environmental stress and could overcome limitations of traditional methods which focus only on monitoring parameters of soil. Current research regarding on-line plant stress sensing techniques concentrates on spectroscopic and image processing methods. These techniques have many limitations connected with their sensitivity to environmental interferences. In recent years, impedance spectroscopy has become a well-known non-invasive tool for describing the electrical properties of many systems. The research hypothesis tested was that information provided by Electrical Impedance Spectroscopy is correlated with tomato plant stress caused by lack of mineral nutrients in the growth medium. The experiment was conducted with two sets of hydroponically-grown tomato plants (Lycopersicon esculentum Mill., cv. ‘Maliniak’). During the experiment the tomato plants were fed alternately with flow of necessary nutrients and with distilled water. The impedance spectra were measured by scanning frequencies from 100 Hz to 50 kHz to determine the most sensitive frequency. A Nutrition Index was proposed for indicating variability of mineral nutrition within plants, and its correlation with experimental plant data was tested. Data showed that the relation between the Nutrition Index and the stress caused by lack of mineral nutrients in the growing medium was a monotonic function in the case of study. The results presented in the paper support the concept that the electrical impedance spectroscopy is a non-destructive, economical and reliable measurement method, which can be utilised for plant nutrition stress monitoring.  相似文献

European eel, Anguilla anguilla, is a target species for future captive breeding, yet best methodology to estimate sperm density for application in in vitro fertilization is not established. Thus, our objectives were to evaluate methods to estimate European eel sperm density including spermatocrit, computer‐assisted sperm analysis (CASA) and flow cytometry (FCM), using Neubauer Improved haemocytometer as benchmark. Initially, relationships between spermatocrit, haemocytometer counts and sperm motility were analysed, as well as the effect of sperm dilution on haemocytometer counts. Furthermore, accuracy and precision of spermatocrit, applying a range of G‐forces, were tested and the best G‐force used in method comparisons. We found no effect of dilution on haemocytometer sperm density estimates, whereas motility associated positively with haemocytometer counts, but not with spermatocrit. Results from all techniques, spermatocrit, CASA and FCM, showed significant positive correlations with haemocytometer counts. The best correlation between spermatocrit and haemocytometer counts was obtained at 6000 ×  g (= 0.68). Of two CASA variants, one or three photographic fields (CASA‐1 and CASA‐2), CASA‐2 showed a very high accuracy to haemocytometer counts ( =  0.93), but low precision (CV: CASA‐2 = 28.4%). FCM was tested with and without microfluorospheres (FCM‐1 and FCM‐2), and relationships to haemocytometer counts were highly accurate (FCM‐1: =  0.94; FCM‐2: =  0.88) and precise (CV: FCM‐1 = 2.5; FCM‐2 = 2.7%). Overall, CASA‐2 and FCM‐1 feature reliable methods for quantification of European eel sperm, but FCM‐1 has a clear advantage featuring highest precision and accuracy. Together, these results provide a useful basis for gamete management in fertilization protocols.  相似文献
