Improvements in sow productivity have raised questions regarding dietary vitamin D recommendations. The present study aimed to evaluate the effects of the housing system with access to sunlight exposure and supplementation of 25-hydroxicholecalciferol on performance and serum levels of 25(OH)D3 in sows during gestation and lactation. Sows were distributed in an experimental design with two housing systems: gestation crates or gestation free-range system with external area for sunlight exposure; and two diets: 0 or 50 μg of 25-hydroxicholecalciferol kg−1. The use of 25-hydroxicholecalciferol tended (P = 0.052) to improve total born and influenced (P = 0.046) on number of born alive. Litter weight at birth was also increased (P = 0.01) by 25-hydroxicholecalciferol supplementation; 25-hydroxicholecalciferol supplementation and housing system (free-range with sunlight exposure) tended to increase weaning weight (P = 0.07) and litter daily gain (P = 0.051) during lactation. Exposure to sunlight and 25-hydroxicholecalciferol supplementation increased 25(OH)D3 serum levels when compared with control treatment during gestation (136.95 vs. 113.92 ng mL−1; P = 0.035) and lactation (120.29 vs. 88.93 ng mL−1; P = 0.026). In conclusion, the association of 25-hydroxicholecalciferol supplementation with exposure to sunlight during gestation improved significantly 25(OH)D3 serum levels and consequently performance traits in gestation and lactation.  相似文献

The continuous use of plowing for grain production has been the principal cause of soil degradation. This project was formulated on the hypothesis that the intensification of cropping systems by increasing biomass‐C input and its biodiversity under no‐till (NT) drives soil restoration of degraded agro‐ecosystem. The present study conducted at subtropical [Ponta Grossa (PG) site] and tropical regions [Lucas do Rio Verde, MT (LRV) site] in Brazil aimed to (i) assess the impact of the continuous plow‐based conventional tillage (CT) on soil organic carbon (SOC) stock vis‐à‐vis native vegetation (NV) as baseline; (ii) compare SOC balance among CT, NT cropping systems, and NV; and (iii) evaluate the redistribution of SOC stock in soil profile in relation to soil resilience. The continuous CT decreased the SOC stock by 0·58 and 0·67 Mg C ha−1 y−1 in the 0‐ to 20‐cm depth at the PG and LRV sites, respectively, and the rate of SOC sequestration was 0·59 for the PG site and ranged from 0·48 to 1·30 Mg C ha−1 y−1 for the LRV site. The fraction of C input by crop residues converted into SOC stock was ~14·2% at the PG site and ~20·5% at the LRV site. The SOC resilience index ranged from 0·29 to 0·79, and it increased with the increase in the C input among the NT systems and the SOC sequestration rates at the LRV site. These data support the hypothesis that NT cropping systems with high C input have a large potential to reverse the process of soil degradation and SOC decline. A genome of a virus preliminarily named avian gyrovirus 2 (AGV2), a close relative to chicken anemia virus, was recently discovered in a chicken in the state of Rio Grande do Sul, Southern Brazil. To study the occurrence of AGV2 in Rio Grande do Sul and the neighboring state Santa Catarina, a number of adult chickens (n=108 and n=48, respectively) were tested for the presence of AGV2 DNA. An AGV2-specific PCR was developed, optimized and used to analyze DNA extracted from clinical samples. AGV2 DNA was detected in 98/108 (90.7%) of samples collected in the state of Rio Grande do Sul and 29/48 (60.4%) of the samples collected in the state of Santa Catarina. In order to check whether AGV2 DNA would be detected in samples from a geographically distant region, DNA from brain samples of 21 diseased chickens from the Netherlands were tested independently, by the same method. In such specimens, 9/21 (42.9%) brain tissue samples were found to contain AVG2 DNA. Sequence analysis of some of the PCR products demonstrated that the amplified AGV2 sequences could vary up to 15.8% and could preliminarily be divided in three groups. This indicated the occurrence of variants of AGV2, which may reflect differences in geographical origin and/or in biological properties. The data presented here provides evidence that AGV2 seems fairly distributed in chickens in Southern Brazil and that AGV2 also circulates in the Netherlands. Besides, circulating viruses display genetic variants whose significance should be further examined, particularly to determine whether AGV2 would play any role in chicken diseases.  相似文献

Mouse inoculation test (MIT) is a highly sensitive test for rabies diagnosis but slow and expensive. To detect rabies virus an in vitro technique using Neuro 2a cell culture (CC) was compared with MIT in two laboratories.

In one laboratory, CC appeared to be on the whole more sensitive than MIT, nevertheless MIT was the only one to detect some positive samples. In the other laboratory, MIT was more sensitive. These results justify the use of CC for epidemiological diagnosis but emphasize the interest of MIT (the reference technique) for special cases.  相似文献

Control of canine visceral leishmaniasis (VL) remains a difficult and serious problem mostly because there is no reliable and effective vaccine available to prevent this disease. A mixture of three recombinant leishmanial antigens (TSA, LeIF and LmSTI1) encoded by three genes highly conserved in the Leishmania genus have been shown to induce excellent protection against infection in both murine and simian models of cutaneous leishmaniasis. A human clinical trial with these antigens is currently underway. Because of the high degree of conservation, these antigens might be useful vaccine candidates for VL as well. In the present study, using the dog model of the visceral disease, we evaluated the immunogenicity of these three antigens formulated with two different adjuvants, MPL-SE and AdjuPrime. The results were compared with a whole parasite vaccine formulated with BCG as the adjuvant. In order to investigate if sensitization with the recombinant antigens would result in recognition of the corresponding native parasite antigens upon infection, the animals were exposed for four weeks after the termination of the immunization protocol with the recombinant antigens to a low number of L. chagasi promastigotes, an etiological agent of VL. Immune response was evaluated by quantitative ELISA in the animal sera before and after exposure to the viable parasites. Both antigen specific IgG1 and IgG2 antibody levels were measured. Immunization of dogs with the recombinant antigens formulated in either MPL-SE or AdjuPrime resulted in high antibody levels particularly to LmSTI1. In addition, this immunization although to low levels, resulted in the development of antibody response to the whole parasite lysate. Importantly, experimental exposure with low numbers of culture forms of L. chagasi promastigotes caused a clear boost in the immune response to both the recombinant antigens and the corresponding native molecules. The boost response was predominantly of the IgG2 isotype in animals primed with the recombinant antigens plus MPL-SE. In contrast, animals primed with the recombinant antigens formulated in AdjuPrime as well as animals vaccinated with crude antigen preparation responded with mixed IgG1/IgG2 isotypes. These results point to the possible use of this antigen cocktail formulated with the adjuvant MPL-SE in efficacy field trials against canine VL.  相似文献

EU annual serosurveillance programs show that domestic duck flocks have the highest seroprevalence of H5 antibodies, demonstrating the circulation of notifiable avian influenza virus (AIV) according to OIE, likely low pathogenic (LP). Therefore, transmission characteristics of LPAIV within these flocks can help to understand virus circulation and possible risk of propagation. This study aimed at estimating transmission parameters of four H5 LPAIV (three field strains from French poultry and decoy ducks, and one clonal reverse-genetics strain derived from one of the former), using a SIR model to analyze data from experimental infections in SPF Muscovy ducks. The design was set up to accommodate rearing on wood shavings with a low density of 1.6 ducks/m2: 10 inoculated ducks were housed together with 15 contact-exposed ducks. Infection was monitored by RNA detection on oropharyngeal and cloacal swabs using real-time RT-PCR with a cutoff corresponding to 2–7 EID50. Depending on the strain, the basic reproduction number (R0) varied from 5.5 to 42.7, confirming LPAIV could easily be transmitted to susceptible Muscovy ducks. The lowest R0 estimate was obtained for a H5N3 field strain, due to lower values of transmission rate and duration of infectious period, whereas reverse-genetics derived H5N1 strain had the highest R0. Frequency and intensity of clinical signs were also variable between strains, but apparently not associated with longer infectious periods. Further comparisons of quantitative transmission parameters may help to identify relevant viral genetic markers for early detection of potentially more virulent strains during surveillance of LPAIV.  相似文献

Combining deep tillage (DT) with lime application at greater depths may improve sugarcane yield. The objective of this study was to evaluate the impact of conventional tillage (CT) and DT systems and liming on sugarcane productivity and soil physical attributes. The experiment was conducted in a clayey-textured Rhodic Hapludox soil cultivated with sugarcane for two growing seasons (first and third ratoons) using a randomized block design with four treatments and four repetitions. The treatments consisted of DT without liming (DT0), DT with liming (DT2), CT without liming (CT0) and CT with liming (CT2). In addition to sugarcane stalk and sugar yields, macroporosity (MA), microporosity (MI), total soil porosity (TP), weighted average diameter (WAD), aggregate stability index (ASI), soil penetration resistance (SPR), pH, potential acidity (H + Al), total soil organic carbon (SOC), particulate organic carbon (POC), mineral-associated organic carbon (MOC), calcium (Ca) and magnesium (Mg) were analysed. DT2 promoted the best soil conservation effect on sugarcane. In general, DT, regardless of lime application, reduced SOC and POC in soil surface layer. Nevertheless, in the long term, localized lime addition resulted in significant reductions in compaction, reaching values below 2 MPa. The soil fertility improvement provided by DT2 promoted increased sucrose concentrations and stalks yield. Considering that there was an improvement in the physical quality and fertility of soil for better plant development, the deep tillage with localized lime addition can be considered an effective alternative for sugarcane cultivation.  相似文献
