One hundred and sixteen isolates of Fusarium oxysporum f. sp. lactucae obtained from 85 fields in three crisphead lettuce-producing areas in Nagano Prefecture, Japan were typed for races using
differential cultivars Patriot, Banchu Red Fire and Costa Rica No. 4. They were also grouped into vegetative compatibility
groups (VCGs) using complementation tests with nitrate non-utilizing (nit) mutants. Two California strains reported as F. oxysporum f. sp. lactucum, a type culture of F. oxysporum f. sp. lactucae, and 28 avirulent isolates of F. oxysporum obtained from crisphead lettuce were included for comparison. Among Nagano isolates, 66 isolates were identified as race 1,
and 50 as race 2. Race 1 strains derived from Shiojiri and Komoro cities and race 2 from Kawakami village and Komoro city.
All isolates of race 2 were biotin auxotrophs, and the race could be distinguished based on its requirement for biotin on
minimal nitrate agar medium (MM). Pathogenic isolates were classified into two VCGs and three heterokaryon self-incompatible
isolates. Strong correlations were found between race and VCG. All the race 1 strains were assigned to VCG 1 except self-incompatible
isolates, and all the race 2 strains to VCG 2. The 28 avirulent isolates of F. oxysporum were incompatible with VCG 1 and VCG 2. California strains was vegetatively compatible with VCG 1, and they were assigned
to race 1. Based on vegetative compatibility, these two races of F. oxysporum f. sp. lactucae may be genetically distinct, and F. oxysporum f. sp. lactucae race 1 is identical to F. oxysporum f. sp. lactucum.

The rumen microbiota comprises a vast range of bacterial taxa, which may affect the production of high-quality meat in Japanese Black cattle. The aim of this study was to identify core rumen microbiota in rumen fluid samples collected from 74 Japanese Black cattle raised under different dietary conditions using 16S rRNA gene amplicon sequencing. In the rumen of fattening Japanese Black cattle, 10 bacterial taxa, showing >1% average relative abundance and >95% prevalence, irrespective of the dietary conditions and the fattening periods, were identified as the core rumen bacterial taxa, which accounted for approximately 80% of the rumen microbiota in Japanese Black cattle. Additionally, population dynamics of the core rumen bacterial taxa revealed two distinct patterns: Prevotella spp. and unclassified Bacteroidales decreased in the mid-fattening period, whereas unclassified Clostridiales, unclassified Ruminococcaceae, Ruminococcus spp., and unclassified Christensenellaceae increased during the same period. Therefore, the present study reports the wide distribution of the core rumen bacterial taxa in Japanese Black cattle, and the complementary nature of the population dynamics of these core taxa, which may ensure stable rumen fermentation during the fattening period. 相似文献

The carcass composition of crossbred Japanese Black × Limousin F2 cattle was examined in order to find an accurate carcass composition equation. The test animals included 17 steers and 17 heifers. The 28 image measurements from the area encircling the vertical line to the thoracic vertebra and the line from the thoracic vertebra between the sixth and seventh rib‐bones were measured by computer image analysis. The relationships between the 29 parameters that added the carcass left side weight of the animal and the carcass composition were suggested. The carcass composition included muscle weight, muscle ratio, fat weight and fat ratio. The carcass composition from steers was estimated by an equation composed of these three or four parameters (R2 = 90.80%, 79.30%, 90.75% and 73.70%, respectively). The selected parameters were measured without cutting the thoracic vertebra. The carcass composition from heifers was estimated by an equation composed of two to four parameters (R2 = 96.15%, 90.98%, 93.60% and 88.22%, respectively). The parameters for the estimation of the muscle and fat weight, and muscle and fat ratio are very similar. Six ruminally cannulated Wagyu (Japanese Black) steers (average initial bodyweight (BW) 387 ± 29 kg) were used in a split‐plot design experiment, comprising a 3 × 3 Latin square design (whole plot) and a randomized block design (subplot). The whole plot treatments were three different feeding levels of supplemental diet, fed at 0.2, 0.4 and 0.6% of BW, on a dry matter (DM) basis. Subplot treatments were two different supplemental diets: a potato pulp silage‐based diet (PPS) and a grain‐based diet (GRAIN). Chopped, medium‐quality cool‐season grass hay (predominately Timothy, Phleum pratense L) was fed daily at 0.7% BW (on a DM basis) as the basal diet. Each period consisted of 21 d, which included 11 d of adaptation to the diets and 10 d of the collection period. Chromium oxide was used as an indigestible marker. In situ forage degradation was measured using the nylon bag technique. The dry matter intake increased (linear; P < 0.01) as the feeding level increased and was not affected by the diet. Digestibility was not affected by any treatments. The GRAIN diet tended to decrease the rate of in situ forage degradation as the feeding level increased, but this trend was not found in the steers fed the PPS diet. Steers fed the GRAIN diet had a lower (P < 0.1) ruminal pH compared with steers fed the PPS diet. Ruminal pH was not significantly affected by feeding level; however, it was numerically higher for steers supplemented at 0.2% per BW than that for the steers supplemented above 0.4% per BW due probably to the higher starch intake. The total volatile fatty acids concentration numerically increased as the feeding level increased and was not affected by the diet. Increasing the feeding level decreased (linear, P < 0.01) the proportion of acetate. Neither diet nor the feeding level had any effects on the proportion of ruminal propionate. The results suggested that, for steers fed the PPS diet, there are not adverse effects on forage digestion in the rumen that occur as the feeding level is increased. 相似文献

We conducted field studies to evaluate the impact that curculio weevil attacks on Pasania edulis (Fagacea) acorns have on the regeneration of this tree. The germination ratio of weevil-attacked acorns was significantly lower than that of sound acorns. The number and position of the attacks on acorns affected the germination ratio. The seedlings from weevil-attacked acorns were shorter and had a smaller leaf area than those from sound acorns. The negative effects on seedling height lasted for at least three years after germination. We also studied the handling of the two types of acorns by granivorous Apodemus mice in a broad-leaved forest dominated by P. edulis. The mice picked up, transported, hoarded, and recovered sound and weevil-attacked acorns similarly. In the study site, the seedlings from weevil-attacked acorns were estimated to account for 1.5–20.4 % of the total seedlings for cohorts of mast years and 0–3.7 % for those of poor or medium acorn production. From these results, we conclude that acorns with the lower part of their cotyledons slightly damaged by weevil larvae might still be able to contribute to the regeneration of P. edulis in the field but that their contribution would be negligible or small even in mast years in this study site. 相似文献

ABSTRACT: The effect of salt concentration on the thermal denaturation profile of myosin in walleye pollack and carp myofibrils was compared by studying the subfragment-1 (S-1) and rod denaturation rates upon heating. Species-specific denaturation mode observed at 0.1 M KCl was no longer detected when samples were heated above 0.5 M KCl, where S-1 and rod denaturation rates were identical to each other. As the heating of the chymotryptic digest of myofibril formed practically no rod aggregates, S-1 denaturation in a form of myosin was the rate limiting step for rod aggregate formation. As the aggregate formation by rod was remarkably suppressed by lowering the temperature, the free movement of myosin tail upon heating was suggested to play an important role in the rod aggregate formation in a high salt medium. 相似文献

Digestion of carp myofibrils at 30 degrees C in 0.5 M KCl medium with calcium ion generated unique 135 kDa heavy meromyosin (HMM). The HMM was not produced when digested at 10 degrees C. A further digestion of the 135 kD HMM isolated in the absence of calcium ion generated uniquely short subfragment-2 (S-2) with a size of 40 kDa (40 kDa S-2) together with subfragment-1 (S-1). The 40 kDa S-2 was identified by N- and C-end sequencing, and demonstrated to locate the amino end of the rod portion. The unfolding temperature for the 40 kDa S-2 was around 52 degrees C as studied by circular dichroism measurement. The same unfolding peak was also detected with the intact rod together with a large unfolding peak at around 36 degrees C coming from the rest of the rod portion, light meromyosin. The unfolding peak for the 40 kDa S-2 in myosin was a little lower (48 degrees C) than that in free form, suggesting the involvement of the head portion in the stability of the 40 kDa S-2 in the structure. 相似文献

Three strains—Nocardioides sp. MFC-A, Rhodococcus rhodochrous MFC-B, and Stenotrophomonas sp. MFC-C—were newly isolated from rice soil or bark compost as microorganisms capable of cometabolically degrading mefenacet (2-benzothiazol-2-yloxy-N-methyl-acetanilide). Several other species belonging to the genera Nocardioides, Rhodococcus, and Stenotrophomonas were also capable of degrading mefenacet, suggesting that the ability is not specific to these isolates, but common at least in these genera. N-methylaniline and 2-benzothiazoloxy acetic acid were identified as the metabolites by liquid chromatography-mass spectrometry. Besides mefenacet, the isolates could degrade one or more other amide pesticides, such as flutolanil, mepronil, metolachlor, and pretilachlor. These results indicate that the strains Nocardioides sp. MFC-A, Rhodococcus rhodochrous MFC-B, and Stenotrophomonas sp. MFC-C degrade mefenacet via hydrolysis of the amide bond, but their substrate specificities differ. 相似文献
