A study was conducted to standardize a protocol for cryopreservation of spermatozoa of the endangered mahseer, Tor khudree (Sykes) and T. putitora (Hamilton). The suitability of the cryoprotectants, dimethyl sulphoxide (DMSO) and glycerol, and the combination of the two were tested. Four equilibration periods and four freezing rates were also tested for their standardization. A combination of 9% DMSO and 11% glycerol gave significantly higher mean percentage of hatching in both T. khudree (45.59±1.86%) (control 71.08±0.59%) and T. putitora (45.00±1.25%) (control 73.48±1.19%) among the eight different treatments. Among the four different equilibration periods tested, the equilibration period of 30 min?1 yielded the highest mean hatching percentage in T. khudree (39.46±1.94%) (control 71.70±0.61%) and T. putitora (38.28±1.06%) (control 73.11±0.82%). Freezing straws at a height of 8 cm above LN2 surface for 10 min?1 gave higher hatching percentages for both T. khudree (41.75±1.72%) (control 73.99±1.17%) and T. putitora (41.34±2.04%) (control 72.48±1.51%). The study reports the superior performance of the combination of DMSO and glycerol for the first time. 相似文献

Pangasianodon aquaculture is the fastest growing food producing sector of the world. Recent national production of Pangasianodon hypophthalmus in India accounts for 0.7 million tonnes at present. Being influenced by trade and markets, P. hypophthalmus has been introduced to many countries including India. There has been a steady increase in the production of P. hypophthalmus by small and marginal farmers of the country. However, concern has arisen about the long-term sustainability due to diseases and its negative impacts on socio-economic aspects and biodiversity. Unregulated culture of P. hypophthalmus has been causing concern to environmental safety and warranting a very cautious and regulated approach to its culture. The standards, protocols and guidelines have been developed to address the issues of sustainability of Pangasianodon catfish production. 相似文献

Tor tor is an important game and food fish of India with a distribution throughout Asia from the trans-Himalayan region to the Mekong River basin to Malaysia, Pakistan, Bangladesh and Indonesia. A new cell line named TTCF was developed from the caudal fin of T. tor for the first time. The cell line was optimally maintained at 28°C in Leibovitz-15 (L-15) medium supplemented with 20% fetal bovine serum (FBS). The propagation of TTCF cells showed a high plating efficiency of 63.00%. The cytogenetic analysis revealed a diploid count of 100 chromosomes at passage 15, 30, 45 and 60 passages. The viability of the TTCF cell line was found to be 72% after 6 months of cryopreservation in liquid nitrogen (-196°C). The origin of the cell lines was confirmed by the amplification of 578- and 655-bp sequences of 16S rRNA and cytochrome oxidase subunit I (COI) genes of mitochondrial DNA (mtDNA) respectively. TTCF cells were successfully transfected with green fluorescent protein (GFP) reporter plasmids. Further, immunocytochemistry studies confirm its fibroblastic morphology of cells. Genotoxicity assessment of H?O? in TTCF cell line revealed the utility of TTCF cell line as in vitro model for aquatic toxicological studies. 相似文献

Observations with the High Resolution Imager on the Rontgensatellit reveal x-ray emissions from Jupiter's equatorial latitudes. The observed emissions probably result from the precipitation of energetic (>300 kiloelectron volts per atomic mass unit) sulfur and oxygen ions out of Jupiter's inner radiation belt. Model calculations of the energy deposition by such heavy ion precipitation and of the resulting atmospheric heating rates indicate that this energy source can contribute to the high exospheric temperatures(>800 kelvin at 0.01 microbar) measured by the Galileo probe's Atmospheric Structure Instrument. Low-latitude energetic particle precipitation must therefore be considered, in addition to other proposed mechanisms such as gravity waves and soft electron precipitation, as an important source of heat for Jupiter's thermosphere. 相似文献

Artemia nauplii cultured for 24 h in lipid‐enriched media contained 0, 0.38, 1.67, 0.79, 5.71 and 5.14 ng 17α‐methyl testosterone (MT) mg?1 dry weight for the control, 5, 15, 30, 50 and 100 mg of 17α‐MT L?1 of media respectively. Giant fresh water prawn Macrobrachium rosenbergii larvae were fed hormone‐enriched Artemia nauplii for 50 days. A significant difference in sex ratios (0.92, 0.69 and 0.93) was obtained in relation to the control group. These results suggest that further research is needed with regard to standardization of doses and duration of treatments for complete hormonal control of sex differentiation in M. rosenbergii. 相似文献

Two new cell culture systems namely epitheloid cells of Lates (LCE) and fibroblastic cells of Lates (LCF) have been developed from fry and fingerling of the economically important brackishwater fish Lates calcarifer. Primary cultures were initiated by explant technique using caudal fin of fingerling and whole body tissue of the fry. The nutritional requirements and the growth pattern in response to different culture environment were similar for the two cell cultures. The culture medium used was Leibovitz‐15 supplemented with 20% fetal bovine serum (FBS) and 1% fish serum. The LCE comprised of epithelioid cells and LCF cells were fibroblastic. With a split ratio of 1:2, the confluency of cells was attained in 8–10 days at an incubation temperature of 28°C. The cells were found to grow well in a wide range of temperature (24–32°C) and stable at 20 and 36°C. The growth rate of LCF and LCE cells increased proportionately with the concentration of FBS from 5% to 20%. A decrease of serum level to 10% after eight subcultures produced no apparent change in cell morphology and growth rate. The viability of cells was found to be 70% when revived after a month of storage in liquid nitrogen (?196°C). 相似文献

A field grazing trial was undertaken to monitor the health and production of crossbred sheep grazing pasture where Echium plantagineum constituted a considerable proportion of the available forage. The trial, conducted for 19 months over successive grazing seasons, demonstrated a significant difference in production, with sheep on the E. plantagineum pasture being lighter and growing less wool compared with sheep on Echium-free pasture. No mortalities involving pyrrolizidine alkaloid poisoning were recorded in sheep grazing E. plantagineum, although there was histological evidence of moderately severe liver damage associated with high liver copper concentrations in at least one sheep following the grazing of large quantities of the plant. 相似文献

The effects of different concentrations of cadmium chloride on the extent of lipid peroxidation (LPO) and alterations in the
antioxidant enzyme activities were studied in liver, kidney and gill tissues of freshwater murrel, Channa punctatus. The fish specimens were exposed to 6.7, 13.4 and 20.1 mg l−1 sublethal concentrations of cadmium chloride and the oxidative stress was assessed after 24, 48, 72 and 96 h post-exposure.
The biomarkers selected for the study were thiobarbituric acid reactive substances for assessing the extent of lipid peroxidation
and antioxidant defense system such as reduced glutathione (GSH), glutathione reductase (GR), glutathione peroxidase (GPX), glutathione-S-transferase (GST), catalase (CAT) and superoxide dismutase (SOD) activities. In general, the cadmium exposure
elevated the LPO in subject tissues of treated group and modulated the activities of GPx, GST, SOD, CAT, GR and level of GSH
after given exposure as compared to the control. All enzymes activities, except CAT (in kidney and gills), and amount of LPO
elevated significantly (P < 0.05) in treated group with respect to control in all tissues, while significant difference was not observed between the
exposed concentrations and within exposure duration. The results indicated that increase in LPO level and the fluctuation
in antioxidant defense system in fish could be due to cadmium-induced increase in the production of reactive oxygen species
(ROS). The potential role of these parameters as biomarkers of heavy metal pollution in aquatic system is discussed. 相似文献

Cell lines provide an important biological tool for carrying out investigations into physiology, virology, toxicology, carcinogenesis
and transgenics. Teleost fish cell lines have been developed from a broad range of tissues such as ovary, fin, swim bladder,
heart, spleen, liver, eye muscle, vertebrae, brain, skin. One hundred and twenty-four new fish cell lines from different fish
species ranging from grouper to eel have been reported since the last review by Fryer and Lannan (J Tissue Culture Methods
16: 87–94, 1994). Among the cell lines listed, more than 60% were established from species from Asia, which contributes more than 80% of
total fish production. This includes 59 cell lines from 19 freshwater, 54 from 22 marine and 11 from 3 brackish water fishes.
Presently, about 283 cell lines have been established from finfish around the world. In addition to the listing and a scientific
update on new cell lines, the importance of authentication, applications, cross-contamination and implications of overpassaged
cell lines has also been discussed in this comprehensive review. The authors feel that the review will serve an updated database
for beginners and established researchers in the field of fish cell line research and development. 相似文献
