A total of 105 nonboar-exposed, F2 ([Landrace x Yorkshire] x Duroc) gilts were used in two replicates of a randomized complete block experiment to evaluate the effect of dietary feed intake on pubertal onset and subsequent body composition. Feed intakes were established at 50% of ad libitum (AL-50), 75% of ad libitum (AL-75), or at ad libitum (AL-100) levels from 4.5 to 9 mo of age. A corn-soybean meal diet fed to all gilts was formulated to meet or exceed nutrient requirements except for energy. Puberty was measured by two methods: 1) monitored once daily by back pressure applied by the herdsman or 2) from elevated plasma progesterone concentrations. Body composition was evaluated by the deuterium oxide method after plasma progesterone concentrations were elevated. Daily feed intake for the experimental period averaged 1.6, 2.3, and 3.2 kg, and the BW of gilts at 8 mo of age were 111, 131, and 154 kg for the AL-50, AL-75, and AL-100 groups, respectively. Body weight, backfat thickness, and body fat content increased linearly (P < .01) as feed intake increased, but age at puberty was not severely influenced. A minimum body fat content or percentage did not seem to initiate pubertal onset. There was a trend for a lower percentage of the AL-50 gilts to ovulate (P = .08) than those fed the AL-75 and AL-100 intakes. Late blight, caused by the oomycete Phytophthora infestans (Mont.) de Bary, is a devastating disease in potato and tomato and causes yield and quality losses worldwide. The disease first emerged in central America and has since spread in North America including the United States and Canada. Several new genotypes of P. infestans have recently emerged, including US-22, US-23 and US-24. Due to significant economic and environmental impacts, there has been an increasing interest in the rapid identification of P. infestans genotypes. In addition to providing details regarding the various phenotypic characteristics such as fungicide resistance, host preference, and pathogenicity associated with various P. infestans genotypes, information related to pathogen movement and potential recombination may also be determined from the genetic analyses. Restriction fragment length polymorphism (RFLP) analysis with the RG57 loci is one of the most reliable procedures used to genotype P. infestans. However, the RFLP procedure requires propagation and isolation of the pathogen and relatively large amounts of DNA. Isolation of the late blight pathogen is sometimes impossible due to the poor condition of the infected tissues or the presence of fungicide residues. In this study, we describe a procedure to identify P. infestans at the molecular level in planta using terminal restriction fragment length polymorphism (T-RFLP) of the RG57 loci. This T-RFLP assay is sufficiently sensitive to detect and differentiate P. infestans genotypes directly in planta without propagation and isolation of the pathogen, to facilitate the timely implementation of best management practices.  相似文献

Sugarcane management systems affect soil attributes such as the carbon cycle. This fact has stimulated the sugar and alcohol industry to refine the sugarcane production systems by replacing the pre-harvest burning (PB) and manual harvest with mechanized harvesting followed by residue deposition. The aim of this study was to evaluate different management systems with respect to C cycling carbon dioxide and soil parameters (chemical, physical and biological) which were determined over the season. Three sugarcane cultivation systems were evaluated at the following periods: (a) PB, (b) 5-year green harvest and (c) 10-year green harvest. The results indicated that CO2 emission was 36% greater in the 10-year sugarcane green harvest system than in the PB system. The bulk density and macroporosity were the factors that were most affected by the different sugarcane management systems and that significantly influenced soil CO2 emissions. The principal component analysis showed that soil CO2 emission was 18% influenced by base saturation (V%) and 14% by pH, especially in the PB area. Additionally, 19% was affected by carbon and macroporosity in the 5- and 10-year green harvest areas, respectively. From our results, it can be concluded that the most CO2 emissions are in the areas of sugarcane green, this is due to the higher carbon concentration when compared with the area of burning sugarcane. The parameters that most influenced the CO2 emissions were bulk density, porosity, macroporosity, pH and V%.  相似文献

REASONS FOR PERFORMING STUDY: Surveillance of equine influenza viruses has suggested that strains included in currently licensed vaccines are a poor match for those predominantly circulating in the field. OBJECTIVE: To assess the ability of Duvaxyn IE-T Plus to provide cross protection against the newly evolved South Africa/4/03 (H3N8) strain of equine influenza virus. METHODS: The vaccine efficacy was evaluated by challenge infection with influenza strain A/eq/South Africa/4/03 (H3N8) 2 weeks after a primary course of 2 vaccinations with Duvaxyn IE-T Plus given at a 4-week interval. The outcome of challenge in vaccinated ponies was compared with that in unvaccinated animals. RESULTS: At the time of challenge, all vaccinated ponies had high levels of antibody to Newmarket/1/93, Newmarket/2/93 and South Africa/4/03 strains measured by single radial haemolysis. After challenge infection, there were statistically significantly decreased clinical scores and virus shedding was significantly lower in the vaccinated ponies compared to unvaccinated controls. CONCLUSION: Two doses of Duvaxyn IE-T Plus provides good clinical and virological protection against challenge with a variant virus 2 weeks after the 2 doses of vaccine. POTENTIAL RELEVANCE: When variant strains of equine influenza virus first emerge, booster immunisations with currently available vaccines may limit infection provided sufficiently high antibody levels are achieved, suggesting that vaccination in the face of an outbreak may be beneficial.  相似文献

The effect of glyphosate on leaves and basal bulbs of yellow nutsedge (Cyperus esculentus L.) plants was studied. Treatment with glyphosate did not affect leaf anatomy, although light necrotic spots were seen on the veins. In contrast, basal bulb organization was greatly affected with large necrotic zones and disorganization in the vascular cylinder due to herbicide accumulation. Depletion or disappearance of vacuolar phenols was observed in bulbs of plants treated with the highest doses of glyphosate. The most remarkable effect of glyphosate was the appearance of one layer of sclerenchymatic cells between root and rhizome primordia and the cortical tissues which could play an important role in the inhibition of rhizome and root emergence by the herbicide.  相似文献
