Group B Streptococcus (GBS) is a multiserotype bacterial pathogen representing a major cause of life-threatening infections in newborns. To develop a broadly protective vaccine, we analyzed the genome sequences of eight GBS isolates and cloned and tested 312 surface proteins as vaccines. Four proteins elicited protection in mice, and their combination proved highly protective against a large panel of strains, including all circulating serotypes. Protection also correlated with antigen accessibility on the bacterial surface and with the induction of opsonophagocytic antibodies. Multigenome analysis and screening described here represent a powerful strategy for identifying potential vaccine candidates against highly variable pathogens.  相似文献

Many reports indicate that dietary selenium, potentially increasing the activity of glutathione peroxidase, could offer protection against free-radical-induced damage. The effects of diets moderately enriched in selenium, as sodium selenite or as a lyophilized selenium-rich food, were studied in rats. Adriamycin, an anticancer drug causing a free-radical-mediated cardiotoxicity, was administered intraperitoneally to some rats. The onset of an oxidative damage was indicated by the increase in the plasma level of reactive oxygen metabolites coupled to a decrease in the total antioxidant activity but without modification of glutathione peroxidase activity, which were observed in all rats, independent of the dietary treatment. On the contrary, in the heart, selenium supplementation caused an increase in the total antioxidant activity, glutathione concentration, and glutathione peroxidase and catalase activities leading to a decreased generation of reactive oxygen metabolites. These results clearly indicate that a moderate Se dietary supplementation counteracts adriamycin-induced cardiotoxicity by preservation of endogenous antioxidants.  相似文献

OBJECTIVE: To evaluate the use of a dermal biopsy punch for accurate and quick removal of ectopic cilia in dogs. ANIMALS STUDIED: This study reports the treatment of ectopic cilia detected in 12 dogs during the period from June 2002 to December 2004. Five cases were affected with ectopic cilia in the right eye, and five in the left eye, with two cases affected bilaterally. Nineteen ectopic cilia were detected in 12 dogs. PROCEDURE: The technique described in this study consists of using a 2-3-mm skin biopsy punch with the aid of an operating microscope. RESULTS: No recurrence was noted in any of the cases. The presurgical blepharospasm due to the presence of the cilia resolved as well as uneventful corneal healing of any associated corneal erosions or ulcers. CONCLUSIONS: This method offers a simple, practical alternative to the traditional scalpel procedure as described in the veterinary ophthalmic literature.  相似文献

Pituitary dependent hyperadrenocorticism (PDH) shows a high morbidity and blindness is one of its complications. Compression of the optic chiasm (OC) by the hypophysis adenoma is one of the causes. Another cause could be due to vascular and metabolic alterations of the PDH. Out of a total of 70 dogs with confirmed diagnosis of PDH, 12/70 showed blindness. In only 2/12 the OC was compromised. Electroretinography in dogs without the OC being compromised showed altered A and B wave patterns. Ophthalmological Doppler showed an alteration of the blood flow only in blind dogs without OC compression. Cortisol concentrations (Co), triglycerides (Tg) and glycaemia (G) were greater in 10 dogs with non-compressive blindness vs. dogs with conserved vision. Loss of vision correlated with the increase in these variables. Blindness in dogs with PDH would be related to changes in retinal blood flow, associated to higher Co, Tg and G concentrations.  相似文献

A field method for determining the soil hydraulic properties using a parameter estimation technique is presented. Input data for the inverse problem are soil-water potentials and soil-water contents measured at different soil depths and different times during a field transient drainage experiment. For the water retention function the parametric relation suggested by Van Genuchten was adopted. For the hydraulic conductivity function the relation proposed by Van Genuchten and the exponential relation were adopted.

With the proposed method soil hydraulic properties along a transect of a volcanic Vesuvian soil were determined using as boundary condition the unit gradient of total potential at the bottom of the soil profile. Geostatistics were used to describe the spatial variability of hydraulic conductivity characteristics of the soil here considered.

Finally, results obtained using this method were compared with those of the simplified method suggested by Sisson and Van Genuchten based on a unit gradient water flow model.  相似文献

Pepino mosaic virus (PepMV) is a highly infectious potexvirus that causes a severe disease in tomato (Solanum lycopersicum) crops worldwide. In Sicily, the first outbreak was detected in a single greenhouse in 2005 and it was promptly eradicated. However, in 2008, a large number of greenhouses were simultaneously affected, and it was impossible to eradicate or control the virus. This study addressed the dispersion and the genetic diversity of PepMV isolates obtained from the outbreak in Sicily, in comparison with worldwide PepMV isolates, to gain insight into the factors determining the evolution and epidemiology of the virus. A total of 1800 samples from plants with and without symptoms were collected in the Sicilian provinces of Agrigento, Caltanissetta, Palermo, Ragusa, Siracusa and Trapani. Three isolates collected at different times were biologically characterized. The incidence of the virus increased rapidly from 13% in 2011 to 63% in 2013, and phylogenetic analysis showed that all Sicilian isolates of PepMV belonged to the CH2 strain, one of the six strains previously described. Nucleotide diversity of the Sicilian isolates was low, thus suggesting rapid spread and genetic stability.  相似文献
