Porcine epidemic diarrhea virus (PEDV) is the causative agent of neonatal diarrhea in piglets, which causes high mortality rates. In this study, the immunoprophylactic effects of chicken egg yolk immunoglobulin (Ig Y) against PEDV were investigated in neonatal pigs. Ig Y was found to reduce the mortality in piglets after challenge exposures. The field application of Ig Y also revealed significant differences in survival rates of piglets given Ig Y, as compared with placebo or control. The results in this study indicated that Ig Y against PEDV could be an alternative way of supplementing prophylactic measures like colostral antibodies from sows.  相似文献

This study was performed to evaluate the effect of beta-tricalcium phosphate and poly L-lactide-co-glycolide-co-epsilon-caprolactone (TCP/PLGC) membrane in the repair of partial bone defects in canine proximal humerus. Three adult mixed-breed dogs were used during the experimental period. The length of the defect was quarter of the full length of humerus, and width of the defect was quarter of middle diameter of the lateral aspect of humerus. The humeri of each dog were divided into treatment (TCP/PLGC) and control groups. The defect was covered with TCP/PLGC membrane in treatment group. To evaluate regeneration of the bone, computerized tomography (CT) and histopathologic examination were performed. The radiopaque lines were appeared at the original defect sites in TCP/PLGC group but below the original site in control at 4th week. Radiopacity and thickness of the defect sites, and radiopaque lines were more increased at 8th week than those of 4th week. Histopathologic findings revealed fibrous connective tissue migration into the defect and the migration inhibited the structure of new cortex to be placed in the original level in control whereas new cortex growth was found in the level of original line in TCP/PLGC group. However, the new cortical bone in the TCP/PLGC group was thinner and less organized than the adjacent intact cortex, and the amount of new cancellous bones were also scanty. The result suggested that TCP/PLGC membrane is a good guided bone regeneration material to restore the original morphology of humerus in partial defect.  相似文献

Thirty-four dogs with no deep pain perception due to acute thoracolumbar intervertebral disc disease underwent decompression surgery within 1 week of diagnosis. All dogs underwent hemilaminectomy. Adipose derived mesenchymal stem cells (AD-MSCs) were transplanted into the injured spinal cord parenchyma for the AD-MSCs transplant dogs. Long-term outcome was evaluated at the end of the follow-up period (> 6 months). AD-MSCs combination treatment showed better recovery outcomes compared to decompression surgery alone. These results indicate that this stem cell therapy is a potential therapeutic strategy to overcome the limitations of treatment for spinal cord injury in clinical medicine.  相似文献

Alternative sources of mesenchymal stem cells (MSCs) for replacing bone marrow (BM) have been extensively investigated in the field of bone tissue engineering. The purpose of this study was to compare the osteogenic potential of canine MSCs derived from adipose tissue (AT), BM, umbilical cord blood (UCB), and Wharton''s jelly (WJ) using in vitro culture techniques and in vivo orthotopic implantation assays. After canine MSCs were isolated from various tissues, the proliferation and osteogenic potential along with vascular endothelial growth factor (VEGF) production were measured and compared in vitro. For the in vivo assay, MSCs derived from each type of tissue were mixed with β-tricalcium phosphate and implanted into segmental bone defects in dogs. Among the different types of MSCs, AT-MSCs had a higher proliferation potential and BM-MSCs produced the most VEGF. AT-MSCs and UCB-MSCs showed greater in vitro osteogenic potential compared to the other cells. Radiographic and histological analyses showed that all tested MSCs had similar osteogenic capacities, and the level of new bone formation was much higher with implants containing MSCs than cell-free implants. These results indicate that AT-MSCs, UCB-MSCs, and WJ-MSCs can potentially be used in place of BM-MSCs for clinical bone engineering procedures.  相似文献

Three factors (extent of chlorination, milling extraction rate, and particle‐size reduction) in cake‐baking functionality of Croplan 594W flour were explored using a Rapid Visco‐Analyser (RVA) and time‐lapse photography. The extent of chlorination and milling extraction rate showed dramatic effects, but postmilling to reduce flour particle size was a less significant factor. RVA results showed that starch pasting was accelerated, and both peak and set‐back viscosities were enhanced, with increasing extent of chlorination. These effects were exaggerated by the high sugar concentration relevant to cake baking, compared to the same effects in water. Cake baking with chlorinated flours, in a formulation with 50% sugar (%S) and 275 parts total solvent (TS), showed that, as the extent of chlorination increased, cake moisture content and edge height decreased. Cake center height and shape factor were curvilinear, with maxima near flour with pH 4.6. Dramatic collapse occurred for cakes baked with unchlorinated flour samples, due to delayed starch pasting, as documented by time‐lapse photography and comparison to the geometry of the final cooled cakes. Starch pasting and egg white setting occurred too early for the cakes baked with excessively chlorinated flour (pH ≤4.0), but too late for the cakes baked with unchlorinated or insufficiently chlorinated flours (pH ≥4.9), compared to the ideal starch pasting and egg white setting behavior with appropriately chlorinated flours (pH >4.0 and <4.9). Informal sensory texture evaluation showed that cake mouthfeel was related to both moisture content per se and the relationship between moisture content and cake relative humidity (%RH). Excessive flour chlorination resulted in unacceptably dry cake mouthfeel.  相似文献

A benchtop baking method has been developed to predict the contribution of gluten functionality to overall flour performance for chemically leavened crackers. To identify a diagnostic cracker formula, the effects of leavening system (sodium bicarbonate, monocalcium phosphate, and ammonium bicarbonate), sugar concentration (%S), and total solvent (TS) on cracker‐baking performance were explored. From preliminary experiments to establish a production procedure, 10 min of dough‐mixing time, a cord‐weave baking mesh, and a 500°F oven temperature were selected. For the leavening system, increasing ammonium bicarbonate (ABC) level at constant sodium bicarbonate (soda) and monocalcium phosphate (MCP) levels resulted in increased cracker height. For the diagnostic formula, 1.25 g of soda, 1.25 g of MCP, and 1.25 g of ABC were selected, based on 100 g of flour. As the sugar concentration in the cracker formula, at constant total solvent (38 TS), decreased to <20%, the resulting cracker dough became softer, and the baked cracker exhibited an increased blistering tendency because of a too‐high formula water level. In contrast, a cracker dough formulated with >40% sugar concentration was too crumbly to handle and sheet. As the total solvent in the cracker formula increased at constant sugar concentration (≈23.7%S), the resulting dough became softer. A dough with 34 TS was too crumbly to handle, while doughs with 42 and 46 TS were too soft to handle and resulted in blistering. Therefore, 38 TS and 23.7%S were identified for the diagnostic formula. Crackers baked with a hard wheat flour, a soft wheat flour, and blends validated the utility of the developed method.  相似文献

Recent global warming trends may have a significant impact on vector-borne viral diseases, possibly affecting vector population dynamics and disease transmission. This study measured levels of hemagglutination-inhibition (HI) antibodies against Japanese encephalitis virus (JEV) and neutralizing antibodies against Akabane virus (AKAV) and Aino virus (AINV) for Thoroughbred horses in Korea. Blood samples were collected from 989 racehorses in several provinces, between October 2005 and March 2007. Sera were tested using either an HI assay or a virus neutralization test. Approximately half (49.7%; 492/989) of the horses tested were antibody-positive for JEV. The HI titer against JEV was significantly correlated with racehorse age (p < 0.05). Horses with an HI antibody titer of 1:160 or higher accounted for 3.9% of the animals tested, indicating that vectors transmitting arthropod-borne viruses bit relatively few horses. In contrast, 3.8% (19/497) and 19.5% (97/497) of horse sera collected in March 2007 were positive against AKAV and AINV, respectively. The presence of antibodies against AKAV and AINV may indicate the multiplication of AKAV and AINV in these horses.  相似文献
