A kernel screening assay (KSA) was used to assess the genetic and environmental effects on the vulnerability of maize to aflatoxin accumulation. Kernels of 26 inbred lines that had been grown in seven environments, and 190 lines of the Intermated B73xMo17 (IBM) population grown in one location in the United States, were inoculated with a toxigenic strain of A. flavus and incubated in the dark at 30°C for 6 days. Percent kernel colonization (PKC), sporulation and aflatoxin were influenced by the maize genotypes (G), the location (“ear environment” or E) and the GxE interactions. Overall, low broad‐sense heritabilities were observed for PKC, sporulation and aflatoxin. PKC was significantly correlated with sporulation in all environments. Aflatoxin was positively correlated with colonization for two and with sporulation for all ear environments. Higher grain sulphur or magnesium in IBM was associated with less colonization or aflatoxin. Postharvest susceptibility of maize to aflatoxin is thus influenced by factors that are modulated by the ear environment. In a KSA, sporulation could be a proxy test for aflatoxin accumulation.  相似文献

Objectives

(1) To collect the perceptions of veterinarians performing equine castrations in Australia on techniques, preferences and outcomes, (2) to investigate veterinarian use and experience with the Henderson castrating instrument and (3) to investigate potential associations between demographics, castration methods and techniques, and complications.

Design

Online survey of members of the Australian Veterinary Association’s Special Interest Group, Equine Veterinarians Australia (EVA).

Methods

A link to the survey was included in the EVA e‐newsletter and practices on the EVA website were contacted by telephone and follow‐up email. Fisher’s exact test was used to determine associations between ligation and complications. A generalised linear model with a negative binomial family was used to determine associations between count response variables and categorical independent variables.

Results

Responses were obtained from 138 veterinarians (response rate, 13.1%) who performed 5330 castrations over 12 months. Castrations were most commonly performed in the field, on anaesthetised horses, using emasculators, via an open approach and without ligation of the spermatic cord. Estimated complications after use of emasculators were swelling (25%), haemorrhage (5%) and infection (5%). The Henderson instrument was used by approximately 10% of respondents and its use for castration was associated with fewer reports of postoperative swelling compared with emasculators (P = 0.002). Rates of evisceration with the Henderson and emasculator methods were comparable (0.43% and 0.9%, respectively).

Conclusion

Castration preferences varied widely among survey participants. Reported complication types and rates were comparable to those reported previously in other countries. Perceptions that the Henderson instrument was associated with less swelling should be investigated further via a prospective controlled investigation.  相似文献

Fasciolosis is an endemic zoonotic parasitic disease with significant impacts on human health and both animal health and production. Early post-infection impacts on the host remain unclear. The objective of this study was to determine the changes, if any, to levels of endotoxin in cattle plasma in response to early-stage infection with Fasciola hepatica. Thirty-six (36) commercial bred cattle were experimentally infected with approximately 400 viable metacercariae. Plasma lipopolysaccharide (endotoxin) levels were examined on 24 occasions from 0 h before infection to 336 h after infection using the Limulus Amoebocyte Lysate chromogenic end point assay and compared with that of six (6) uninfected control animals. Peak lipopolysaccharide levels in infected animals were reached at 52 h after infection and returned to pre-infection levels at time 144 h after infection. Infected animals had significantly elevated lipopolysaccharide levels between 24 and 120 h after infection when compared to uninfected animals. The mean change in endotoxin units (EU)/mL over time after infection was statistically significant in infected animals. Elevations of lipopolysaccharide occurred in all infected animals suggesting a possible repeatable and titratable endotoxemia conducive to therapeutic agent model development.  相似文献

To assess the effects of feeding high-oil corn on carcass characteristics and meat quality, 60 yearling steers were fed high concentrate diets containing either control corn (82% of diet), high-oil corn (82% of diet), or high-oil corn at a concentration that was isocaloric with the control diet (74% of diet). After being fed for 84 d, steers were slaughtered. At 72 h postmortem, carcass data were collected and rib sections from five steers grading U.S. Choice and five steers grading U.S. Select from each treatment were collected, vacuum packaged, and aged for 14 d. Three steaks (2.54 cm thick) were removed from each rib for Warner-Bratzler shear force measurement, sensory appraisal, and fatty acid composition analyses. Data were analyzed with treatment as the main effect for the carcass data and treatment, quality grade, and two-way interaction in the model for the longissimus data. The two-way interaction was nonsignificant (P > 0.05) for all variables tested. No differences were detected (P > 0.05) in carcass measurements except for marbling scores and quality grades, both of which were greater (P < 0.05) for carcasses from steers fed the high-oil corn. Overall, 78% of steers fed the high-oil corn graded U.S. Choice compared with 47% for the control and 67% for isocaloric group. Shear force and sensory properties of the longissimus were not different (P > 0.05) among treatments. Steaks from U.S. Choice carcasses rated higher (P < 0.05) for tenderness and tended to rate higher (P < 0.10) for juiciness. Feeding the isocaloric and high-oil diets increased (P < 0.05) linoleic acid, arachidonic acid, and the total PUFA content of lipid extracted from the longissimus. Saturated fatty acid percentage was lowest (P < 0.05) for high-oil corn and highest (P < 0.05) for control, with isocaloric being intermediate. Feeding high-oil corn increased (P < 0.05) pentadecyclic acid, margaric acid, and total odd-chain fatty acid content. Feeding high-oil corn in finishing beef cattle diets enhanced intramuscular lipid deposition and increased unsaturation of fatty acids of the longissimus.  相似文献

Three Angus steers (410 kg) cannulated in the proximal duodenum were used in a replicated 3 x 3 Latin square to evaluate the effects of dietary lipid level and oil source on ruminal biohydrogenation and conjugated linoleic acid (CLA) outflow. Dietary treatments included: 1) typical corn (TC; 79.2% typical corn), 2) high-oil corn (HOC; 79.2% high-oil corn), and 3) the TC diet with corn oil added to supply an amount of lipid equal to the HOC diet (OIL; 76.9% TC + 2.4% corn oil). Duodenal samples were collected for 4 d following 10-d diet adaptation periods. Data were analyzed with animal, square, period, and treatment in the model and planned, nonorthogonal contrasts were used to test the effects of dietary lipid content (TC vs HOC and OIL) and oil source (HOC vs OIL) on ruminal biohydrogenation. Intake and duodenal flow of total long-chain fatty acids were increased (P < 0.05) by over 63% for diets containing more lipid regardless of oil source. Apparent ruminal dry matter and long chain fatty acid digestibilities were not altered (P > 0.05) by dietary lipid level or oil source. Ruminal biohydrogenation of total and individual 18-carbon unsaturated fatty acids was greater (P < 0.05) for diets with higher lipid content. Biohydrogenation of oleic acid was greater (P < 0.05) for HOC than OIL, but biohydrogenation of linoleic acid was lower (P < 0.05) for HOC than OIL. Duodenal flows of palmitic, stearic, oleic, linoleic, and arachidic acids were more than 30% greater (P < 0.05) for diets containing more lipid. Flow of all trans-octadecenoic acids was greater (P < 0.05) for diets containing more lipid. Corn oil addition increased (P < 0.05) the flow of trans-10 octadecenoic acid and the trans-10, cis-12 isomer of CLA by threefold compared to feeding high-oil corn. Feeding high-oil corn or adding corn oil to typical corn rations increased intake, biohydrogenation, and duodenal flow of unsaturated long-chain fatty acids. Compared with high-oil corn diets, addition of corn oil increased duodenal flow of trans-10, trans-12 and cis-12 isomers of octadecenoic acid and the trans-10, cis-12 isomer of CLA. The amount of cis-9, trans-11 isomer of conjugated linoleic acid flowing to the duodenum was less than 260 mg/d, a value over 20 times lower than flow of trans-11 vaccenic acid indicating the importance of tissue desaturation for enhanced conjugated linoleic acid content of beef.  相似文献

One in vitro and one in vivo metabolism experiment were conducted to examine the effects of supplemental Zn on ruminal parameters, digestion, and DMI by heifers fed low-quality prairie hay supplemented with urea. In Exp. 1, prairie hay was incubated in vitro for 24 h with five different concentrations of supplemental Zn (0, 5, 10, 15, and 20 ppm) and two concentrations of supplemental Mn (0 and 100 ppm), both provided as chloride salts. Added Mn increased (P < 0.02) IVDMD, but added Zn linearly decreased (P < 0.03) IVDMD. Added Zn tended to increase the amount of residual urea linearly (P < 0.06) at 120 min and quadratically (P < 0.02) at 180 min of incubation, although added Mn counteracted these effects of added Zn. Six 363-kg heifers in two simultaneous 3 x 3 Latin squares were fed prairie hay and dosed once daily via ruminal cannulas with urea (45 or 90 g/d) and with Zn chloride to provide the equivalent of an additional 30 (the dietary requirement), 250, or 470 ppm of dietary Zn. After a 7-d adaptation period, ruminal contents were sampled 2, 4, 6, 12, 18, 21, and 24 h after the supplement was dosed. Supplemental Zn did not alter prairie hay DMI (mean = 4.9 kg/d) or digestibility, although 470 ppm added Zn tended to decrease (P < 0.06) intake of digestible DM, primarily due to a trend for reduced digestibility with 470 ppm supplemental Zn. Zinc x time interactions were detected for both pH (P = 0.06) and NH3 (P = 0.06). At 2 h after dosing, ruminal pH and ruminal ammonia were linearly decreased (P < 0.05; P < 0.01) by added Zn. At 5 h after feeding, ruminal pH was linearly increased (P < 0.05) by added Zn, suggesting that added Zn delayed ammonia release from urea. The molar proportion of propionate in ruminal fluid was linearly and quadratically increased (P < 0.02; P < 0.01) whereas the acetate:propionate ratio was linearly and quadratically decreased (P = 0.02; P < 0.05) by added Zn. Through retarding ammonia release from urea and increasing the proportion of propionate in ruminal VFA, Zn supplementation at a concentration of 250 ppm may decrease the likelihood of urea toxicity and increase energetic efficiency of ruminal fermentation.  相似文献

Bluetongue virus serotypes 1 and 3 infection in Poll Dorset sheep   总被引:1,自引:0,他引:1  
Objective To study the clinical signs following bluetongue virus serotypes 1 and 3 infection in Poll Dorset sheep.
Design A clinical and pathological study.
Procedure Twenty Poll Dorset sheep were inoculated with bluetongue virus serotypes 1 or 3, each inoculum having a different passage history. The sheep were examined daily and their clinical appearance and rectal temperatures recorded. Heparinised and non-heparinised blood samples were taken at intervals for virological and serological study. Gross pathological findings were recorded for several sheep at necropsy and tissue samples were collected from three sheep for virological studies.
Results All inoculated sheep developed clinical disease. The clinical signs and gross pathological changes varied considerably but were consistent with damage to the vascular endothelial system. There was a decline in the titres of infectious bluetongue virus and of antigen in tissues collected between 7 and 12 days after infection.
Conclusions The severity of disease was related to the speed of onset and duration of pyrexia and not the development or titre of viraemia. Generally, those animals with sensitive mouths, depression, coronitis, recumbency and reluctance to move were the most debilitated. Whole blood was the most reliable source of infectious virus from acutely and chronically infected and convalescent animals. However, tissue samples particularly spleen, collected from dead or killed animals suffering from either peracute or acute forms of disease were most appropriate for the rapid confirmation of a clinical diagnosis.  相似文献

A serum viral inhibitor (SVI) was isolated from goats and partially characterized. The inhibitor prevented the cytopathic effects of vesicular stomatitis virus, encephalomyocarditis virus, and a caprine herpesvirus, indicating broad antiviral activity. The SVI was distinct from interferon because SVI did not induce an antiviral state in cells (ie, lack of protection of SVI-treated cells from virus challenge). The SVI had activity on heterologous cells, including human, bovine, and ovine cells. The lack of antiviral activity in mouse cells indicated that SVI was not an antibody. Like fibroblast interferon, however, SVI was sensitive to trypsin, was acid stable at pH 2 and 4 C for 4 days, was heat stable at 56 C for 1 hour, and could not be sedimented by centrifugation at 100,000 X g for 4 hours.  相似文献

Small Shorea stenoptera Burck. (Dipterocarpaceae) trees of reproductive age growing in an arboretum in west Java were studied to determine the pattern of vegetative shoot development, the time and method of floral initiation and the effect of paclobutrazol on floral enhancement. Vegetative buds were enclosed by two stipules between which was a leaf primordium, a small axillary vegetative bud and another pair of stipules. This sequence was reiterated five to seven times before the vegetative apex was visible. At the time of floral initiation, axillary buds developed into floral spikes and compound inflorescences formed at the end of drooping branches. A compound inflorescence might bear many floral spikes and each floral spike bore many flowers. The compound inflorescence was a modification of the reiterative developmental pattern observed in vegetative shoots. The time of floral initiation began in late June or early July and continued until about November. Floral enhancement using paclobutrazol as a soil drench was attempted in mid-July, but this was later found to be after the onset of floral initiation, and the treatment failed to enhance flowering; however, it appeared to enhance the rate of floral and fruit development. The similarity in vegetative bud development among dipterocarp genera suggests that the time of floral initiation may be easily determined in many species based on simple dissection techniques.  相似文献
