AIM: To compare, in cows treated with an internal teat sealant, the effect of short-acting and long-acting cloxacillin-based dry-cow therapy on somatic cell counts (SCC) after calving.

METHODS: Cows from a spring-calving, pasture-based dairy farm in the Manawatu-Whanganui region of New Zealand were randomly allocated to receive either a short-acting cloxacillin and ampicillin dry-cow therapy and internal teat sealant (n=291) or a long-acting cloxacillin and ampicillin dry-cow therapy and internal teat sealant (n=288) at the end of lactation. Cows were managed on-farm with routine husbandry procedures through the dry period and following calving. A multivariable logistic regression model was used to determine the association between length of action of dry-cow therapy and the proportion of cows with a SCC >150,000?cells/mL at the first herd test after calving.

RESULTS: Age of cow, mean SCC for the preceding season and interval from calving to the first post-calving herd test were all associated with the proportion of cows with an individual SCC >150,000?cells/mL at the first herd test (p<0.001) Treatment with the short-acting dry-cow therapy was not associated with decreased odds of cows having a SCC >150,000?cells/mL at the first herd test compared with treatment with long-acting dry-cow therapy (OR=0.724; 95% CI=0.40–1.30).

CONCLUSIONS AND CLINICAL RELEVANCE: In this herd, which routinely used internal teat sealants, the use of short-acting cloxacillin-based dry-cow therapy did not result in an increased proportion of cows with elevated SSC post-calving. This was a single farm, single year study but indicates that in this herd, changing from a long-acting to a short-acting antimicrobial may have no impact on the prevalence of subclinical mastitis.  相似文献

Crossbred pigs (n = 216) were used to test the interaction, if any, of ractopamine (RAC) and dietary fat source on the characteristics of fresh pork bellies. Pigs were blocked by BW (77.6 +/- 6.5 kg) and allotted randomly to pens (6 pigs/pen). After receiving a common diet devoid of RAC for 2 wk, pens within blocks were assigned randomly to 1 of 4 treatments arranged in a 2 x 2 factorial design, with 5% fat (beef tallow vs. soybean oil) and RAC (0 vs. 10 mg/kg). At the conclusion of the 35-d feeding period, pigs were slaughtered at a commercial pork packing plant (average BW of 108.8 +/- 0.6 kg), and fresh bellies were captured during carcass fabrication. Neither RAC (P = 0.362) nor fat source (P = 0.247) affected belly thickness. Subjective (bar-suspension) or objective (compression test) measures of belly firmness were not (P > or = 0.148) affected by the inclusion of RAC in the diet; however, bellies from pigs fed soybean oil (SBO) were softer than those from pigs fed beef tallow (BT), as indicated by perpendicular (P < or = 0.005) and parallel (P < 0.001) suspensions. Moreover, bellies from BT-fed pigs required more (P = 0.096) force to compress 50% of their thickness than bellies from SBO-fed pigs (52.29 vs. 43.51 kg). Color (L*, a*, and b* values) of the belly lean and fat was not (P > or = 0.131) affected by RAC, and lean color was similar (P > or = 0.262) between fat sources; however, belly fat from BT-fed pigs was lighter (P = 0.030) and redder (P = 0.013) in color than belly fat from SBO-fed pigs. Bellies of SBO-fed pigs had greater (P < 0.001) proportions of PUFA and lower (P < 0.001) proportions of SFA and MUFA than belly fat from pigs fed BT. Regardless of the RAC inclusion level, PUFA:SFA and iodine values were lower in belly fat from pigs fed BT than SBO; however, within SBO-fed pigs, PUFA:SFA and iodine values were further increased by feeding RAC (RAC x fat source, P < 0.001). As expected, dietary fat source altered the fatty acid composition of fresh pork bellies, which subsequently impacted fresh belly firmness. Interestingly, including RAC in swine finishing diets exacerbated the effect of feeding SBO on pork fat polyunsaturation.  相似文献

Crossbred pigs (n = 185) were used to test the effects of dietary Fe supplementation on performance and carcass characteristics of growing-finishing swine. Pigs were blocked by BW, allotted to pens (5 to 6 pigs/pen), and pens (5 pens/block) were allotted randomly to either negative control (NC) corn-soybean meal grower and finisher diets devoid of Fe in the mineral premix, positive control (PC) corn-soybean meal grower and finisher diets with Fe included in the mineral premix, or the PC diets supplemented with 50, 100, or 150 ppm Fe from Availa-Fe (an Fe-AA complex). When the lightest block averaged 118.2 kg, the pigs were slaughtered, and bone-in pork loins were collected during fabrication for pork quality data. During the grower-I phase, there was a tendency for supplemental Fe to reduce ADG linearly (P = 0.10), whereas in the grower-II phase, supplemental Fe tended to increase ADG linearly (P = 0.10). Even though pigs fed NC had greater G:F during the finisher-I phase (P < 0.05) and across the entire trial (P = 0.07), live performance did not (P > or = 0.13) differ among dietary treatments. There were linear increases in 10th-rib fat depth (P = 0.08) and calculated fat-free lean yield (P = 0.06); otherwise, dietary Fe did not (P > 0.19) affect pork carcass muscling or fatness. Moreover, LM concentrations of total, heme, and nonheme Fe were similar (P > 0.23) among treatments. A randomly selected subset of loins from each treatment was further fabricated into 2.5-cm-thick LM chops, placed on styrofoam trays, overwrapped with polyvinyl chloride film, and placed in coffin-chest display cases (2.6 degrees C) under continuous fluorescent lighting (1,600 lx) for 7 d. During display, chops from NC-fed pigs and pigs fed the diets supplemented with 100 ppm Fe tended to have a more vivid (higher chroma value; P = 0.07), redder (higher a* value; P = 0.09) color than LM chops of pigs fed 50 ppm of supplemental Fe. Moreover, greater (P < 0.01) redness:yellowness ratios in chops from pigs supplemented with 100 ppm Fe indicated a more red color than chops from PC-fed pigs or pigs fed diets supplemented with 50 ppm Fe. In conclusion, however, increasing dietary Fe had no appreciable effects on performance, carcass, or LM characteristics, suggesting that current dietary Fe recommendations are sufficient for optimal growth performance, pork carcass composition, and pork quality.  相似文献

Optical experiments were conducted on a series of organic linear chain conductors with different values of the interchain single-electron transfer integral tb, which quantifies the degree of anisotropy. Electron-electron interactions together with Umklapp scattering resulted in a correlation gap and an insulating state for small tb. An insulator-to-metal transition was observed when tb exceeded a critical value, on the order of the correlation gap Egap. The absence of a plasma edge on the insulator side of the transition for polarization perpendicular to the chains suggests that the electrons are confined to the chains. The optical features of the metallic state, when contrasted with the magnetic properties, are suggestive of spin-charge separation.  相似文献

High nitrogen fixing, phosphate solubilizing, phytohormones producing isolates of Azotobacter, Azospirillum, Acetobacter and Pseudomonas were used as inoculants for cotton. Important cultures were selected on the basis of their effect on root/shoot length and chemotactic behaviour. Selected bioinoculants were earlier tested for their beneficial properties like nitrogen fixation (ARA), ammonia excretion, IAA production etc. These bio-inoculants were further tested for phosphate solubilization property. Various chosen strains were tested with Desi (HD 123) and American (H 1098) cotton under field conditions (as for wheat). Plant height and boll weight were determined at the time of harvesting whereas survival rate of inoculated bacteria was determined after 30, 80 and 130 days respectively. In the year 2000–01, on the basis of boll number and boll weight plant?1 AVK 51 (36; 76.2?g plant?1), HT 57 (27; 56.9?g plant?1), AC18 (33; 61.5?g plant?1), Ala 27 (36; 61.4?g plant?1) and Pseudomonas (34; 71.3?g plant?1) were identified as significant both for American and desi cotton varieties. Highest survival rate was observed with Mac 68 (33.4 × 105) followed by HT54 (31.5 × 105) after 30 days of sowing, which decreased after 80 days and remained constant up to 130 days. This trend was observed with all the cultures. Similar results were observed in 2001–02. 25?kg ha?1 N saving was observed with A. chroococcum (AVK51) bioinoculant for cotton crop.  相似文献

Crossbred pigs, heterozygous for the halothane gene, were used to determine the effects of long-term dietary supplementation of magnesium mica (MM) and short-duration transportation stress on performance, stress response, postmortem metabolism, and pork quality. Pigs were blocked by weight, penned in groups (six pigs per pen), and pens (three pens per diet) were assigned randomly either to a control corn-soybean meal diet or the control diet supplemented with 2.5% MM (as-fed basis; supplemented at the expense of corn). Diets were fed during the early-finisher (0.95% lysine, as-fed basis; 43.7 to 68 kg) and late-finisher (0.85% lysine, as-fed basis; 68 to 103 kg) periods. At the conclusion of the 71-d feeding trial, 12 pigs from each dietary treatment were selected randomly and subjected either to no stress (NS) or 3 h of transportation stress (TS). Dietary MM had no effect (P > or = 0.40) on ADG or ADFI; however, G:F was improved (P < 0.05) during the early-finisher period when pigs were fed MM-supplemented diets. Plasma glucose concentrations were increased in TS pigs fed the control diet, but transportation did not affect plasma glucose in pigs fed 2.5% MM (diet x transportation stress; P = 0.02). Dietary MM did not affect blood lactate, cortisol, insulin, NEFA, Ca, or Mg concentrations in response to TS (diet x transportation stress; P > or = 0.13); however, circulating lactate, cortisol, and glucose concentrations increased in TS pigs (transportation stress x time; P < 0.01). The LM from TS pigs fed MM had higher initial (0-min) and 45-min pH values than the LM from NS pigs fed the control diet (diet x transportation stress x time; P = 0.07). Lactic acid concentration and glycolytic potential were greater in the LM of TS pigs fed MM than TS pigs fed control diets (diet x transportation stress; P < or = 0.01). Although some trends were identified, neither MM (P > or = 0.15) nor TS (P > or = 0.11) altered the color or water-holding capacity of the LM and semimembranosus. The transportation model elicited the expected changes in endocrine and blood metabolites, but dietary MM did not alter the stress response in pigs. Conversely, although pork quality traits were not improved by dietary MM, delaying postmortem glycolysis and elevating 0- and 45-min muscle pH by feeding finishing diets fortified with MM may benefit the pork industry by decreasing the incidence of PSE pork in pigs subjected to short-duration, routine stressors.  相似文献

Samples of serum and urine were obtained simultaneously from 56 healthy lactating cows to determine ranges of fractional excretion (FE) of calcium (Ca), phosphate (PO4), magnesium (Mg), sodium (Na), potassium (K), and chloride (Cl). Samples were obtained at 3 stages of lactation: period 1 = 1 to 7 days, 2 = 83 to 112 days, and 3 = 175 to 197 days. The FE of electrolytes were significantly different among periods 1, 2, and 3 for Ca (P less than 0.001), PO4 (P less than 0.025) and Mg (P less than 0.025), but were not significantly different for Na, K, and Cl. Least squares mean FE of Ca was lowest in period 1 and not significantly different for periods 2 and 3, whereas mean FE values for PO4 and Mg were highest in period 2 and not significantly different for periods 1 and 3. The mean FE values of Na, K, and Cl did not change with stage of lactation. Age and category of milk production (high, medium, and low) did not influence the FE values of the electrolytes.  相似文献

The purpose of this study was to compare the echocardiographic Doppler blood pressure and heart rate effects of 1:1 thiopental/propofol with thiopental and propofol, when used as anesthesia‐induction agents. Seven healthy dogs (six Beagles and one Pembroke Welsh Corgi), ranging in age from 1 to 9 years and weighing 14.2 ± 2.4 kg (mean ± SD), were used during the study. In a cross‐over study design with a minimum drug interval of 3 days, each dog received propofol, thiopental, or a mixture of propofol–thiopental IV until each dog received all the three anesthetic agents. An initial dose (propofol 4.9 ± 0.8 mg kg?1; thiopental 12.9 ± 2.4 mg kg?1; propofol–thiopental 2.3 ± 0.3 mg kg?1 (P)?5.7 ± 0.8 mg kg?1 (T)) of each anesthetic agent was titrated IV until intubation was accomplished. Echocardiographic Doppler blood pressure and heart rate variables were recorded prior to anesthesia and at 1, 5, and 10 minutes after induction of anesthesia. anova and the Bonferroni's t‐test were used to evaluate the groups for differences. Alpha was <0.05. There was no significant effect of treatment on systolic or diastolic ventricular wall thickness, septal thickness, left atrial diameter, or systolic left ventricular diameter. There was a tendency for diastolic left ventricular diameter to decrease over time. There was a tendency for heart rate to increase with a significant difference at the 10‐minute time period between propofol (109 ± 26 beats minute?1) and thiopental (129 ± 23 beats minute?1). At the 10‐minute recording period, heart rate following the propofol/thiopental mixture (110 ± 34 beats minute?1) was closer to that following propofol than to that following thiopental. With all induction agents, indirect blood pressure tended to decrease over time (p = 0.005); however, there was no difference between the groups. The changes observed were not considered to be of clinical significance. The propofol/thiopental mixture produces similar changes in echocardiographic variables when compared to propofol or thiopental, and could be substituted for propofol for induction of anesthesia in dogs.  相似文献

Thirty Holstein fetuses from 100 to 251 d after conception were utilized to study prenatal anatomical development of the epithelium of the rumen and reticulum. Four calves at birth were included in the study for comparison. Tissue sections were frozen and stained to locate specific enzymes. At 100 d, the epithelial layer of the rumen was differentiated into a thin basal zone and a thickened superficial zone of undifferentiated cells. The basement membrane was straight, and in both zones cells were perpendicular to it. At 120 to 141 d, low, primary undulations were detected in the basal zone, basement membrane and underlying lamina propria. At 150 to 166 d, secondary undulations and incipient papillae began to resemble the papillae of mature mucosa. In rumen papillae of 192-d to 215-d fetuses, shallow furrows began to separate papillae apexes from the mass of epithelium. In fetuses 244 to 251 d, the papillae began to be a separate entity. At birth, the basal position of the papillae still remained fused. An incipient separation between the papillae was seen. Several dehydrogenase enzymes, including those associated with the Krebs cycle and reductase associated with energy transformation, were observed in both ruminal and reticular tissue. Alkaline phosphatase activity was localized in the stratum corneum and in blood vessels. Development of the honeycomb configuration of reticular epithelium was evident in the 100-d fetus and progressed rapidly with age.  相似文献
