Malignant catarrhal fever (MCF) is a fatal herpesvirus infection of domestic and wild ruminants, with a short and dramatic clinical course characterized primarily by high fever, severe depression, swollen lymph nodes, salivation, diarrhea, dermatitis, neurological disorders, and ocular lesions often leading to blindness. In the present study, fatal clinical cases of sheep associated malignant catarrhal fever (SA-MCF) were identified in cattle in the state of Karnataka. These cases were initially presented with symptoms of diarrhea, respiratory distress, conjunctivitis, and nasal discharges. Laboratory diagnosis confirmed the detection of ovine herpesvirus-2 (OvHV-2) genome in the peripheral blood samples of two ailing animals. The blood samples collected subsequently from sheep of the neighboring areas also showed presence of OvHV-2 genome indicating a nidus of infection in the region. The positive test results were further confirmed by nucleotide sequencing of the OIE approved portion of tegument gene as well as complete ORF8 region of the OvHV-2 genome. Phylogenetic analysis based on the sequence of the latter region indicated close genetic relationship with other OvHV-2 reported elsewhere in the world.  相似文献

Artemisia pallens Bess. is a low volume and high value essential oil plant used in perfumery, cosmetic and flavouring industries. On account of the failure of conventional procedures to induce variability in species, mutation techniques have been tried in our experiments. Dry and viable seeds (moisture content 8%) of homozygous pure breeding lines were subjected to 150–500 Gy doses of gamma rays and 0.01–0.1% ethyl methane sulphonate (EMS) for 8 h. Desirable qualitative mutants were recovered from segregating M2 generation (4,283 plants scored) raised as single plant progenies. The spectrum of morphological mutants included late and early flowering types; bushy and high yielding types; tall and more capitula-producing types and high oil and high davanone yielding types. These were raised through M3 families to evaluate stability and transmission of mutant characters. As such out of 15 different types selected in M2, only 11 types bred true to their characteristic variability. Based on their performance, the mutants were characterised depending upon their distinguishing features. Davanone, the main component of oil showed the maximum increase (64.22% against 54.64% in control) in mutant ‘S–5’ recovered from exposure with 250 Gy γ-rays. Mutant ‘E-6’ was economically most viable having increased oil biosynthesis (0.36% against 0.22% in control) and hence yields higher oil per unit area than the parental control (isolated from 0.05% EMS treatment).  相似文献

Moderately thermophilic bacterial strain CC-HSB-11(T) (Muricauda lutaonensis), which was described recently from a coastal hot spring of Green Island, Taiwan, has been identified to produce zeaxanthin as a predominant xanthophyll by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Cell culture in bioreactor produced 3.12 ± 0.18 mg zeaxanthin L(-1) of culture. Micronization of zeaxanthin was achieved through supercritical carbon dioxide antisolvent precipitation method. Yield of zeaxanthin after the process was 53.4%. Dynamic light scattering assay determined the polydisperse existence of micronized particles of size 3 nm to 2 μm. Field emission scanning electron microscopy revealed distinct morphology and size distribution heterogeneity of particles. Integrity of zeaxanthin after the antisolvent process was assessed by LC-MS/MS. The technique capitalizes on the inherent ability of CC-HSB-11(T) to synthesize zeaxanthin and the work demonstrated feasibility of antisolvent precipitation method to produce microparticles exploiting a bacterial strain.  相似文献

Root-nodulating bacteria were isolated and characterized from seven native shrubby legumes growing in Taiwan. Phenotypic characteristics measured included growth rates in various media, colony morphology, and tolerances to extremes of temperature, salt and pH. The isolates were very diverse phenotypically. Among the 83 isolates that were screened, the majority were fast-growing rhizobia. Twenty eight strains tolerated high concentration of salt (4.5% NaCl) and grew well between temperatures of 37 and 45 °C. The majority of the strains also tolerated extreme pH in their medium from 3.5 to 12. All strains formed nitrogen fixing nodules, and the highest activity was detected in the legume Hedysarum crinita L. PCR restriction fragment length polymorphism (PCR-RFLP) and sequencing of the small subunit ribosomal RNAs revealed that the majority of the isolates belonged to the genera Rhizobium, Bradyrhizobium and Agrobacterium. Only a single strain represented the genus Sinorhizobium. In addition, a strain related to Burkholderia from the β-class of the Proteobacteria (CC-CC-5) was found within nodules of the legume Catenaria caudatum. The study contributes to the understanding of symbiotic nitrogen fixation in selected wild legumes that are native to Taiwan and provides insights into the distribution of nodulating and nitrogen-fixing bacteria from other distinct lineages.  相似文献

Wood polymer nanocomposites (WPNC) based on nano-SiO2 were prepared by impregnation of styrene acrylonitrile copolymer (SAN), SiO2 nanoparticles modified with γ-trimethoxy silyl propyl methacrylate (MSMA), and nanoclay into wood. The structure of modified SiO2 nanoparticles and WPNC was characterized by Fourier transform infrared spectroscopy (FTIR). XRD analysis showed the delaminated structure of SAN/SiO2/clay-treated wood composites. The synergistic effect of nano-SiO2 and nanoclay was investigated. Thermal stability of SiO2 nanoparticles decreased after modification, while that of wood treated with SAN, SiO2, and nanoclay improved. Morphological characteristics were examined by scanning electron microscopy (SEM). Mechanical properties, water uptake (%), dimensional stability, hardness, and flammability were found to improve due to incorporation of SiO2 and nanoclay into wood polymer composites. Maximum improvement in properties was observed in the wood polymer composites containing SiO2 and nanoclay at the ratio of 1:1.  相似文献

Supply of 0.1 mM sodium selenite (Na2SeO3 to excised maize leaf segments during greening reduced total chorophylls and inhibited in vivo nitrate reductase activity (NRA). Inhibition of chlorophyll formation by selenite was affected by ammonium but remained unaltered by nitrate. Selenite also inhibited NADH-glutamate dehydrogenase (NADH-GDH) and NADH-glutamate synthase (NADH-GOGAT) activities. Inclusion of ammonium enhanced the inhibition of NADH-GDH activity by selenite but decreased the inhibition of NADH-GOGAT activity by selenite. The results suggest the involvement of the glutamine synthetase/glutamate synthase (GS/GOGAT) pathway of ammonia assimilation in selenite toxicity during chlorophyll formation in greening maize leaf segments.  相似文献

The objective of the present study was to investigate the effect of fermented apple diet (FAD) supplementation on the growth performance and meat quality in finishing Berkshires. The FAD was made from dropped apple mixed with rice bran and barley bran. Until 81 ± 1 kg live weight at 133 ± 1 days, the animals were fed a growing diet, after which experimental samples were fixed at 0, 2, 4 and 6% FAD as C, T1, T2 and T3 in the finishing diets. Growth performance, ADG, ADFI and feed efficiency were improved in T1 than other groups. In carcass parameters, carcass weight was higher ( P  < 0.05) in T1 than in other groups. In meat quality, moisture and crude protein contents decreased ( P  < 0.05) by addition of FAD. pH24 and WHC were higher ( P  < 0.05) in T1 than other groups. In sensory evaluation, marbling of fresh meat and tenderness, juiciness, flavor and overall acceptability of cooked meat were improved by the addition of FAD. According to the results of our experiment, FAD can be used for improvement of meat quality parameters.  相似文献

Northeast India is rich in Citrus genetic diversity representing several wild and cultivated species. Besides commercially cultivated species, several wild, semi-wild and domesticated species namely Citrus indica, C. macroptera, C. ichangensis, C. latipes, C. megaloxycarpa and C. assamensis are found to grow in Northeast India. These species have great potential in improvement of Indian Citrus industry being source of genes for combating biotic and abiotic stresses. The natural diversity of these genetic resources is shrinking at alarming rate due to large scale deforestation, shifting cultivation practiced in these areas and climate change scenario in this important hotspot of biodiversity. Socio-economic importance of each species have been studied during surveys and interesting domestication trends were observed based on their cultural and economic significance, which led to “in situ on farm” conservation of C. indica, C. macroptera and threat to C. megaloxycarpa and C. ichangensis. Consequently, genetic resources of most of these species are facing severe threat of extinction necessitating policy interventions and adoption of dynamic conservation and management strategies. Species specific conservation strategy especially through “in situ on farm” conservation has been proposed with suitable sites in Northeastern Indian states based on population structure and suitability of habitat. Farmers’ support through “The Biological Diversity Act, 2002” and “The Protection of Plant Varieties and Farmers’ Rights Act, 2001” of India would bring the required impact on management of these complex genetic resources of Citrus. In the present study, we have documented the current status, socio-economic potential, domestication trends and associated traditional knowledge of these wild and semi-wild Citrus species occurring in Northeast India and enumerated suitable conservation strategies and desired policy interventions for their dynamic conservation and sustainable utilization.  相似文献
