The effectiveness of the morantel sustained release trilaminate (MSRT) in controlling gastrointestinal nematodes through a grazing season was evaluated using 60 yearling beef stocker calves randomly divided into 2 groups of 30 animals each. In April 1985, the calves comprising the treatment group each received an MSRT designed to release morantel tartrate continuously for 90 days while those of the control group remained unmedicated. All animals were weighed and samples of rectal feces were taken at 14-day intervals, beginning on Day 0, until trial termination (Day 168). At trial termination, 10 control and 10 treated calves were necropsied for recovery of gastrointestinal nematodes. Three sets of parasite-na?ve tracer calves were utilized to evaluate the initial, interim and final levels of pasture contamination by nematode larvae. Overall, the use of the MSRT resulted in a 75.5% reduction (P less than 0.001) in output of nematode eggs from the principals, an 81.8% reduction (P less than 0.001) in numbers of gastrointestinal nematodes in principals (at trial termination), and a 96.9% reduction (P less than 0.05) of pasture larval nematode contamination (as indirectly indicated by parasite burdens in tracer calves). The effect of ivermectin delivered from a sustained-release bolus (I-SRB) on the weight gain of beef cattle through a grazing season was evaluated using 20 yearling beef steer calves randomly divided into two groups of ten animals each. Calves in the control group each received a placebo bolus, while those in the treatment group each received an I-SRB designed to release 12 mg ivermectin day-1 for approximately 90 days. All animals were weighted and samples of feces were collected from the rectum at monthly intervals, beginning on Day 0, until trial termination (Day 148). Pasture larval counts were also conducted on herbage collected on each sampling date. On Day 119, two control and two treated calves were removed from pasture, housed in isolation for 3 weeks, then necropsied for recovery of gastrointestinal nematodes. Three sets of parasite-naive tracer calves were utilized to evaluate the initial, interim and final levels of pasture contamination by nematode larvae. A fourth set was used to evaluate the level of pasture contamination the following spring. The use of the I-SRB resulted in a greater than 99% reduction in fecal egg counts of trichostrongyles and numbers of gastrointestinal nematodes in the treated principals, as well as an average daily gain advantage of 0.114 kg over the 148 day period. A 67-98% reduction in pasture larval nematode contamination occurred on pastures grazed by the treated animals, as indicated by the parasite burden in tracer calves and pasture larval counts. The treatment effect was eventually lost by the following spring since tracer calves on the treated pasture had only 33% fewer nematodes than those on the control pasture.  相似文献

Six Swedish and one Canadian single spore isolate of Drechslera teres f. teres were used to screen 109 barley lines for disease resistance and to select a differential set of barley lines for use in assessing pathogen virulence. A large variation for net blotch resistance was found among the 109 barley lines which were classified into four groups, those showing: 1) only resistant reactions; 2) differential reactions; 3) only intermediate reactions and 4) only susceptible reactions. The European commercial varieties included, showed susceptibility to all Swedish isolates, but a few were resistant to the Canadian isolate. The 18- member differential set separated 25 Swedish and two Canadian isolates of D. teres into 14 pathotypes, three of which made up 59% of the isolates. Only one barley differential (CI 9776) was resistant to all net form isolates. Host selection on the pathogen seems to be present as all six isolates obtained from cv. Golf belonged to the same pathotype and 4 of 5 isolates from cv. Karin shared the same virulence pattern. The net form of net blotch (D. teres f. teres) predominated in the sampled regions and only one of 26 Swedish isolates was of the spot form (D. teres f. maculata). Phototransduction in rod cells is likely to involve an intracellular messenger system that links the absorption of light by rhodopsin to a change in membrane conductance. The direct effect of guanosine 3',5'-monophosphate (cGMP) on excised patches of rod outer segment membrane strongly supports a role for cGMP as an intracellular messenger in phototransduction. It is reported here that magnesium and calcium directly affect the conductance of excised patches of rod membrane in the absence of cGMP and that magnesium, applied to intact rod cells, blocks a component of the cellular light response. The divalent cation-suppressed conductance in excised patches showed outward rectification and cation-selective permeability resembling those of the light-suppressed conductance measured from the intact rod cell. The divalent cation-suppressed conductance was partly blocked by a concentration of the pharmacological agent L-cis-diltiazem that blocked all of the cGMP-activated conductance. Divalent cations may act, together with cGMP, as an intracellular messenger system that mediates the light response of the rod photoreceptor cell.  相似文献

Serum antibody responses in feral or domesticated dogs naturally infected with Echinococcus granulosus or/and other common helminths were examined in an enzyme-linked immunosorbent assay (ELISA) using antigens prepared from E. granulosus protoscoleces or oncospheres. The ELISA using the protoscolex antigen was optimised with serums from experimental dogs monospecifically infected with E. granulosus or other helminth parasites, and helminth-free dogs. Anti-protoscolex antibody was detected in 16 of 22 (72.7%) serums from feral dogs with E. granulosus burdens ranging from 300 to 302,600 worms per dog. Seven serums from feral dogs which did not harbour E. granulosus at autopsy but which originated from an endemic hydatid region were tested using protoscolex antigen, and 1 serum gave a positive reaction. One hundred and two serums from dogs known never to have been infected with E. granulosus all gave negative reactions to protoscolex antigen. The sensitivity of the ELISA test proved to be superior to that which has been achieved by arecoline purging as a method of diagnosis for E. granulosus infection in dogs. For use of the assay in hydatid control or eradication campaigns, its sensitivity can be increased by choosing a lower absorbance discrimination value above which serums are regarded as having positive reactions. However, this does introduce positive reactions of some serums from dogs infected with helminths other than E. granulosus. Calves were vaccinated with antigens collected during in vitro cultivation of the larval stages of Taenia ovis, T. hydatigena or T. saginata, and challenged 4 weeks later with 4 000 T. saginata eggs. Calves vaccinated with T. saginata antigen were highly resistant to the challenge infection and those groups vaccinated with T. ovis and T. hydatigena were also significantly resistant.  相似文献
