A lupin seed γ-conglutin-enriched preparation was tested in a glucose overload trial with both murine models and adult healthy volunteers. The results with rats showed a dose-dependent significant decrease of blood glucose concentration, which confirmed previous findings obtained with the purified protein. Moreover, three test-product doses equivalent to 630, 315, and 157.5 mg γ-conglutin, orally administered 30 min before the carbohydrate supply, showed a relevant hypoglycemic effect in human trials. Insulin concentrations were not significantly affected. The general hematic parameters did not change at all.This is the first report on the glucose-lowering effect of lupin γ-conglutin in human subjects.  相似文献

The Aeta Magbukún of Mariveles are one of the least known and researched Indigenous peoples remaining on the fringe of the bay, and within the remaining forests in Bataan province on Luzon Island in the Philippines. This work describes the unique cultural systems and language of the Aeta Magbukún tribe in Biaan, Mariveles, and both the traditional forest resource use and the evolving new subsistence practices developed to adapt to the encroachment of non-Indigenous peoples onto ancestral lands. The Aeta's forest resource use practices are discussed from a sustainable Indigenous development context within unique socioeconomic, cultural, and environmental circumstances in Bataan.  相似文献

IL-1R/TLRs play an important role in the inflammatory responses. Their signaling pathway is tightly regulated to keep inflammation under control. The regulation involves both extracellular and intracellular mechanisms. TIR8, also known as SIGIRR (Single Immunoglobulin IL-1R-Related molecule), is an orphan receptor belonging to the IL-1R/TLRs super-family. Recent studies suggest its role in modulating the inflammatory response in the gastrointestinal tract in mice, acting as an IL-1 receptor family antagonist. We identified the sequence and analyzed the expression of TIR8 in a wide panel of organs and tissues in different animals. There was high homology of the cDNA sequence among human, mouse and the domestic species (74–85%). The pattern of expression of this receptor was similar in all the species examined (high levels in kidney and gastrointestinal tract) and similar to the human. These results demonstrate that TIR8 is conserved, in evolutionary terms, both with regard to sequence and pattern of expression, a finding consistent with a key function of this molecule in modulating inflammation in the gastrointestinal tract.  相似文献

The aim of the present study was to analyze the effects of drug detection training on behavior and blood neurotransmitter levels in drug detection dogs so as to investigate some variables influencing dog reactivity and responsiveness to training. In all, 20 dogs were sampled out of the Guardia di Finanza canine population. All the subjects were born, reared, housed, and trained in the same facility and followed the same training sessions. Dogs’ behavioral reactivity was scored according to a standardized working dogs test to evaluate natural dog attitudes. Plasma samples were analyzed by the high-performance liquid chromatography method to evaluate adrenaline, noradrenaline, L-3,4-dihydroxyphenylalanine, homovanillic acid (HVA), 3-methoxy-4-hydroxyphenylglycol acid (MHPG), 5-hydroxyindole acetic acid (5-HIAA), and 5-hydroxytryptamine (5-HT) levels. 5-HT and 5-HIAA were also analyzed from platelets. The analysis was carried out considering training, breed, and sex as independent variables. From a behavioral point of view, significant differences were recorded before and after training in “sociability,” “playfulness,” “predatory instinct,” and “aggressiveness” scores. Lower levels of platelet 5-HT and 5-HIAA were found after training. Plasma L-3,4-dihydroxyphenylalanine levels differed between sexes, with males showing higher concentrations. These results underline the importance of complete and objective evaluations protocols of the dogs before, during, and after drugs search training to determine effective and successful selection strategies and training procedures.  相似文献

In 1996, dichlorodiphenyltrichloroethane (DDT) pollution of industrial origin was discovered in Lake Maggiore. It was caused by industrial effluents on a tributary of the River Toce, one of the major affluents of the lake in correspondence of Pallanza Bay. This event is the worst case of environmental pollution that has occurred in Western countries in the last 25 years, not due to agricultural use of DDT, but because of an accidental industrial discharge. Heavy polychlorinated biphenyls (PCBs) pollution was also noticed in 2002, with concentration levels three to seven times higher than those measured in other Italian subalpine lakes. In this study, the current DDT and PCBs contamination levels were assessed according to their presence in zebra mussel (Dreissena polymorpha) specimens sampled in the last 5 years (2003–2008) in eight sampling stations of Lake Maggiore, chosen to cover the entire perimeter of the basin. Moreover, for two stations (Baveno and Pallanza-Villa Taranto) located inside and outside Pallanza Bay, respectively, it is possible to make comparisons starting from 1996. The results obtained show how Lake Maggiore is still an ecosystem with a severe environmental risk, more than 10 years after the original insecticide discharge. DDT contamination continues to evolve, and natural events, like lake overturn, floods, and heavy rains, can have a great influence on the insecticide levels in the lake. By contrast, PCB contamination is absolutely negligible, even if the peak of pollution revealed in 2002 seems to indicate that these pollutants are still present in large quantities in the Lake Maggiore watershed.  相似文献

This study evaluated the importance of the environment and spatial distribution of juvenile fish for the adoption of alternative migratory tactics in a partially anadromous population of rainbow trout (Oncorhynchus mykiss) from the Santa Cruz River. We captured young‐of‐the‐year fish along the river during autumn 2009, 2010 and spring 2010 and determined their maternal origin (anadromous vs. resident) using strontium to calcium ratios in the otolith core. Relative proportion of anadromous offspring, modelled with logistic regression, increased towards headwaters and in areas with deeper channels and larger substrate composition. Body length, modelled with linear multiple regression, varied positively with site depth, water velocity, substrate size and anadromous maternal origin. Based on evidence for limited juvenile movements (<25 km), the spatial extent of this study (240 Rkm) and the identification of large, contrasting reaches along the river, it is likely that the observed spatial distribution of juveniles and their association to sites with coarse substrate composition reflects maternal spawning activity. Results further indicate that anadromous females breed predominantly in middle and upper river sections in areas with coarse substrate. Given that body size in this system has been positively related to propensity for anadromy, we propose that female spawning choice affects their offspring's spatial distribution, providing the adequate physical template for anadromous offspring to reach or maintain larger body sizes and display anadromy themselves. Relevance of this study is also discussed in the context of alterations in response to future dam construction in one of the latest free‐flowing rivers sustaining anadromous O. mykiss.  相似文献

Effect of cooking time on starch retrogradation and water distribution was studied in pasta (spaghetti) and rice (parboiled and arborio) using differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) Optimum cooking times (OCT) were 8, 16, and 18.5 min for spaghetti, parboiled, and arborio rice, respectively. Swelling was observed by image analysis. OCT spaghetti and rice showed various starch retrogradation rates at various aging times and temperatures. Based on the classical Avrami function, the retrogradation rate at 5°C followed the order spaghetti > parboiled rice > arborio rice, while extent was in the opposite order. At higher temperature (20°C), the rates decreased by 20× in all cases. Thermogravimetric analysis (TGA) investigations were undertaken to check the distribution of water within these products and its relationship to starch retrogradation. During heating, water was released in two distinguishable steps at ≈80 and 100°C. Results supported the conclusion that the more tightly bound water might not participate or facilitate starch retrogradation. In this study, the overall water content did not change during storage, and water appeared to migrate from sites of stronger binding to sites of weaker binding. The temperature dependence of the Avrami constant was described with the Vogel‐Tamman‐Fulcher empirical expression.  相似文献

A blocking (B) dot enzyme-linked immunosorbent assay (ELISA), using a monoclonal antibody (mAb) against a group specific antigen of bluetongue virus (BTV) is described for the detection of BTV antibodies to BTV in cattle sera. Dots of BTV antigens were adsorbed to nitrocellulose (NC) strips and/or NC mounted in the windows of dipsticks. After blocking the remaining sites of the NC paper with milk powder solution and immersion in the test sample, the NC strips and dipsticks were exposed to mAb. Bound mAb was detected with peroxidase conjugated anti-mouse IgG (H and L). In the absence of anti-BTV antibody in the test sample, BTV antigen sites were reactive with mAb as indicated by a brown colored dot in the presence of the enzyme substrate, hydrogen peroxide and diaminobenzidine. In the presence of sufficient anti-BTV antibodies no color reaction was observed. The performance of these assays in detecting anti-BTV antibody in field blood eluate samples, prepared from whole blood dried on filter paper, from 395 bluetongue-free cattle in Canada and 635 sentinel cattle in Florida, USA, was evaluated and compared with the standard competitive (C) ELISA. The specificity of the dipstick B-dot ELISA was identical to that of the C-ELISA in testing of BT-free Canadian cattle but not in the testing of samples from the sentinel cattle in Florida, resulting in values of 100% diagnostic and 88.9% relative specificity, respectively. Based on the C-ELISA, the specificity of the NC strip B-dot ELISA was low and in the same order as that of the dipstick assay.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
