In the present study, embryogenic calli of sugarcane variety BL4 were induced using 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin in different concentrations and combinations. In contrast to earlier studies, embryogenic callus sectors were identified and isolated microscopically within 1–2 weeks. Subsequently, 51 media formulations were used for regeneration of proliferated embryogenic callus, using MS medium supplemented with three different cytokinins [kinetin, 6-Benzylamino purine (BAP), and thidiazuron (TDZ)] and auxins (IAA/NAA and IBA) in different combination and concentrations. After acclimatization, the genomic DNA of regenerated plants was studied to explore the insertion polymorphism of transposable elements in order to ascertain the variation among somaclones. Though low concentration of kinetin with 2,4-D was found supportive to embryogenic callus development, the highest number of regenerated plantlets was observed using BAP (1 μM), however the plantlets had very low fresh weight (2.2 g). Conversely, TDZ alone supported a significant increase in the number of plantlets regenerated (38–40) with higher fresh weight. The somaclones generated during this study showed considerable positional polymorphism of activator-like transposable elements possibly due to the stress associated with in vitro culture. This study provides a procedure to produce regenerated sugarcane plants from embryogenic callus in a relatively short time.  相似文献

采用样带和样方相结合的方法沿海拔梯度取样,利用数量生态学方法对小五台山国家级自然保护区亚高山草甸群落进行分析。结果表明,1)双向指示种分析(TWINSPAN)将该区的草甸群落分为9个类型;2)RDA排序结果较好地反映出群落分布格局与环境梯度的关系,各个群落类型在前两轴分异明显;第一轴反映了群落分布格局与坡向、土壤温度、海拔、土壤厚度极显著相关(P0.01),与土壤湿度显著相关(P0.05);第二轴反映了群落分布格局与坡度、土壤厚度极显著相关(P0.01);3)环境因子的蒙特卡罗随机置换检验表明,坡向、海拔、坡度显著影响群落分布(P0.05),对植被类型的形成具有关键作用;4)草甸群落Patrick丰富度指数随着海拔的升高呈下降趋势,随着坡向值的增加、土壤温度的升高呈逐渐上升趋势;Shannon-Wiener多样性指数随土壤厚度的增大呈微小下降趋势;Pielou均匀度指数随着海拔的升高、土壤湿度的增大呈上升趋势,随着坡向值的增加、土壤温度的升高呈逐渐下降趋势;Simpson优势度指数随着坡度的增大表现出微小的下降趋势,随土壤厚度的增大呈上升趋势。  相似文献

We analyzed guava fruits (Psidium guajava L. cv. Red Suprema) from Cuba to determine their chemical composition, total antioxidant capacity, as well as their protective effect against oxidative damage using an in vitro model of human dermal fibroblasts. The guava fruit is a natural source of bioactive compounds, such as polyphenols, vitamin C, folates and beta carotenes with proven health benefits. Human dermal fibroblasts were pre-incubated with different concentrations of guava crude extract and then subjected to oxidative stress using the AAPH stressor. The number of apoptotic and dead cells, as well as the markers of oxidative damage such as lipid and protein oxidation significantly decreased when cells were pre-incubated with guava crude extract and then exposed to the stressor. The activity of antioxidant enzymes also improved when cells were pre-incubated with guava crude extract in comparison to cells subjected to stress without prior pre-incubation with the guava extract. The results obtained in this study highlight the health benefits of guava regarding oxidative stress, proving it to be an important source of bioactive compounds associated with important biological properties.  相似文献

A study was conducted to determine the artificial breeding response, embryonic and larval development of giant snakehead, Channa marulius, with the application of various dosages of stimulatory hormones: Suprefact® (LHRH) agonist and Ovaprim® (GnRH + dopamine inhibitor). First and second hormonal dosages of Suprefact® (diluted) and Ovaprim® (0.3, 0.4, and 0.5 ml for male and 0.8, 0.9, and 1.0 ml for females per kilogram body weight) were used. There were three treatments T1, T2, and T3 and each treatment had three replicates. Male fish were treated with T1 (0.3), T2 (0.4), T3 (0.5) ml kg−1 of body weight while female fish with T1 (0.8), T2 (0.9), T3 (1.0) ml kg−1 of body weight. The results showed that fish stimulated with T3 (1.0) obtained better fecundity rate (2951.7) followed by T2 (0.4) (2678.3), while the lowest fecundity (466.7) was recorded in T1. The numerically higher values of gonado-somatic index (males and females) and fertilization rate appeared in T2. The highest survival rate (97.1%) was found in T3 followed by T2 (97.0%), while the lowest (32.2%) in T1. In conclusion, the application of Suprefact® and Ovaprim® at 0.4 and 0.5 ml kg−1 body weight for male and 0.9 and 1.0 ml kg−1 for female fish successfully stimulated snakehead fish induced breeding.  相似文献

Semen from Black Bengal bucks was collected to establish a cooling protocol (to −196°C) for buck semen preservation, and to study the effect of freezing on sperm motility and morphology. Semen was diluted with diluents (Triladyl & Tris) and cryoprotectants, filled into straws, sealed, cooled (to 5°C) and equilibrated. After dilution, motility ranged from 75.00% to 76.67% and from 73.33% to 80.00% in Triladyl and Tris diluents, respectively. Motility of sperm after cooling to 5°C in Triladyl and Tris diluents ranged from 65.00% to 66.67% and from 63.33% to 70.00%, respectively. After equilibration in straws, the semen was subjected to a freezing protocol in a computer-controlled biofreezer CL-3000 (cooling at 10°C per minute, from 5°C to −80°C) and plunged into liquid nitrogen. Sperm motility of re-thawed semen varied from 38.33% to 43.33% and from 6.00% to−6.67% in Triladyl and Tris diluents, respectively. Sperm morphology of re-thawed semen was studied and head damage or cryoinjury was found in 2–3% of sperm in Triladyl diluents and 3–6% in Tris diluents. Whether the differences of sperm motility and head damage reflect fertility after artificial insemination is yet unknown and needs to be studied further.  相似文献

Plant hosts can be engineered to disrupt the development of sedentary plant-parasitic nematodes or proper functioning of the feeding sites the nematodes induce. The use of constitutive promoters to express dsRNAs or nematode inhibitor proteins may be unreliable because of possible silencing or yield penalty from continuous expression in a plant host. This ill-effect can be avoided if a root-specific, nematode-responsive promoter (NRP) is used to drive the target nematode-inhibitory message. This study used the In Plant Activation (INPACT) system to express a barstar-controlled barnase in galls of Meloidogyne javanica and assessed how the engineered tobacco lines affected the growth and development of the nematodes. Of the 11 combinations of four NRPs and the CaMV 35S promoter assessed, the AtCel1 and TobRB7 combinations activated specific expression of split β-glucuronidase (GUS) and barnase genes in and around giant cells. The same NRP combination directed expression of the barnase gene in tobacco roots also constitutively expressing the barstar gene (SPBB transgenic lines). On roots of six T1 SPBB lines, there was up to 94% reduction in the number of galls with significantly smaller adult females compared to those on wild-type plants. Some of the females on lines SPBB4-1 and SPBB-4-2, for example, were not associated with galls. The results indicated the engineered plants disrupted M. javanica development and demonstrate the potential for controlled and localized expression of peptides, such as those that could block specific effectors, to disrupt initiation, formation, establishment, or proper functioning of feeding cells induced by damaging sedentary nematodes.  相似文献

Farmers normally practice conventional tillage ((CT), disk plowing, cultivator, rotavator, and leveling) in cotton (Gossypium hirsutum L.) with 15 cm intra-row spacing to avoid risks of poor plant stand and obtain higher yield. However, CT is costly besides it has adverse effects on soil and crop when sown after wheat. Conservation tillage [zero tillage (ZT) or reduced tillage (RT)] with suitable spacing can reduce production cost, increase cotton yield and quality, and it has favorable effects on soil properties. Field experiments were conducted to evaluate cotton response to tillage (ZT, RT, and CT) and intra-row spacing (15.0, 22.5, 30.0, 37.5 cm). Results revealed that RT produced higher bolls plant?1, boll weight, seed cotton yield, ginning out turn, fiber length and strength than ZT and CT. Mean boll weight, seed cotton yield, earliness, and fiber qualities were optimum at 22.5 cm spacing. Tillage × spacing interaction showed optimum boll weight, earliness, and fiber strength with 15.0–22.5 cm spacing under RT. CT with 22.5 cm spacing also performed better in terms of boll weight and fiber strength; however, 15.0 cm spacing resulted in earlier maturity. RT with 22.5 cm spacing is an alternative to CT for higher yield, earliness, and quality of cotton besides environmental safety.  相似文献

Sewage contains several trace elements of environmental concern, and cadmium (Cd) is one of the most mobile elements in soil–plant system that can pose drastic effects on plants and human health due to its long persistence and non-biodegradability nature in environment. It is necessary to prevent its entry into food chain for better food quality and human health. Present study was designed to evaluate the effectiveness of different water management practices, viz. W1: flooding throughout the growing season, W2: flooding after 4 days of disappearance of standing water (DAD), W3: flooding till heading and after that flooding of soil after 4 days of DAD, W4: Aerobic condition throughout growing season (flooding after 8 days) for reducing Cd concentration in rice grain grown under varying levels of Cd (0, 20, 40 mg kg?1) spiked soil. Results revealed that grain yield declined with increasing Cd levels but maximum plant height (89.3 cm), straw yield (16.9 g) and grain yield (22.5 g pot?1) was observed where pots were flooded till heading and thereafter flooding. Cadmium concentration increased with increasing concentration of Cd in soil. Further, it is added that the lowest Cd concentration in shoot, grain and husk and translocation factor were observed under W3 when the soil was spiked with 40 mg kg?1. In crux, continuous flooding till heading and thereafter flooding after 4 DAD can significantly decrease the grain Cd concentration without compromise on yield.  相似文献

Hepatitis E is a rare human disease in Central Europe commonly imported from endemic regions. For autochthonous infections a zoonotic transmission from pigs, deer and wild boar is assumed. Using three different RT-PCR protocols, hepatitis E virus (HEV) RNA was detected in 10 out of 189 (5.3%) serum samples collected in 1995/1996 from wild boars in Germany. Sequence analysis indicates a close relationship with genotype 3 isolates of pigs and humans from the Netherlands and Japan. The results indicate that HEV is present in Germany since more than 10 years and that wild boar may function as a reservoir for HEV.  相似文献
