Interferon (IFN)-γ has been shown to be associated with immunity to Marek’s disease virus (MDV). The overall objective of this study was to investigate the causal relationship between IFN-γ and vaccine-conferred immunity against MDV in chickens. To this end, 3 small interfering RNAs (siRNAs) targeting chicken IFN-γ, which had previously been shown to reduce IFN-γ expression in vitro, and a control siRNA were selected to generate recombinant avian adeno-associated virus (rAAAV) expressing short-hairpin small interfering RNAs (shRNAs). An MDV challenge trial was then conducted: chickens were vaccinated with herpesvirus of turkey (HVT), administered the rAAAV expressing shRNA, and then challenged with MDV. Tumors were observed in 4 out of 10 birds that were vaccinated with HVT and challenged but did not receive any rAAAV, 5 out of 9 birds that were administered the rAAAV containing IFN-γ shRNA, and 2 out of 10 birds that were administered a control enhanced green fluorescent protein siRNA. There was no significant difference in MDV genome load in the feather follicle epithelium of the birds that were cotreated with the vaccine and the rAAAV compared with the vaccinated MDV-infected birds. These results suggest that AAAV-based vectors can be used for the delivery of shRNA into chicken cells. However, administration of the rAAAV expressing shRNA targeting chicken IFN-γ did not seem to fully abrogate vaccine-induced protection.  相似文献

Relative abundance and density estimates of Asian houbara were assessed during the breeding season in two breeding grounds of migrant populations (China and south Kazakhstan) and in one breeding ground of a resident population (Oman), between 1998 and 2002. For the study period, the relative abundance was from 0.004 to 0.06 individual per km driven and density estimates varied from 0.01 to 0.2 houbara per km2. Relative abundance and density of houbara declined by 63% and 69%, respectively in China, by 60% and 49%, respectively in Kazakhstan and by 50% and 75%, respectively in Oman. Overall, an average of 27-30% annual decline in both relative abundance and density was observed for the three regions. Despite being legally a strictly protected species in China, Kazakhstan and Oman, houbara are heavily hunted and poached on all their migration routes and wintering grounds. The current levels of hunting and poaching are not sustainable and without the immediate agreement and implementation of international conservation measures, the Asian houbara may face extinction in the wild in the foreseeable future.  相似文献

Subcycle strong-field ionization (SFI) underlies many emerging spectroscopic probes of atomic or molecular attosecond electronic dynamics. Extending methods such as attosecond high harmonic generation spectroscopy to complex polyatomic molecules requires an understanding of multielectronic excitations, already hinted at by theoretical modeling of experiments on atoms, diatomics, and triatomics. Here, we present a direct method which, independent of theory, experimentally probes the participation of multiple electronic continua in the SFI dynamics of polyatomic molecules. We use saturated (n-butane) and unsaturated (1,3-butadiene) linear hydrocarbons to show how subcycle SFI of polyatomics can be directly resolved into its distinct electronic-continuum channels by above-threshold ionization photoelectron spectroscopy. Our approach makes use of photoelectron-photofragment coincidences, suiting broad classes of polyatomic molecules.  相似文献

The attosecond time-scale electron-recollision process that underlies high harmonic generation has uncovered extremely rapid electronic dynamics in atoms and diatomics. We showed that high harmonic generation can reveal coupled electronic and nuclear dynamics in polyatomic molecules. By exciting large amplitude vibrations in dinitrogen tetraoxide, we showed that tunnel ionization accesses the ground state of the ion at the outer turning point of the vibration but populates the first excited state at the inner turning point. This state-switching mechanism is manifested as bursts of high harmonic light that is emitted mostly at the outer turning point. Theoretical calculations attribute the large modulation to suppressed emission from the first excited state of the ion. More broadly, these results show that high harmonic generation and strong-field ionization in polyatomic molecules undergoing bonding or configurational changes involve the participation of multiple molecular orbitals.  相似文献

Variations in soil acidity and in biodiversity were analysed in the National Natural Park "Russian North", European Russia. Improving soil quality from podzol, podzolic soil, derno-podzolic soil, brown earth to pararendzina leads to increase in diversity and changes in floristical composition, followed by changing of pine and spruce forest to mixed and birch forests. In PCA ordination species diversity, richness and evenness of trees, shrubs and vascular plants are closely connected with each other, and are represented by the first principal component. They are strongly correlated to the thickness of Al horizon, pHH20 and pHCaC12 in organic, surface and subsurface mineral horizons. Only bryophyte species richness and diversity are directly related to the thickness and weight of organic horizon, soil exchangeable acidity, and inversely related to the thickness of Al horizon and pH. Thus, the ordination of the major species diversity variables is highly related to soil pH, suggesting that pH is the best soil related predictor of species diversity parameters. Our study shows that plants notably respond to soil acidification in boreal forest ecosystems.  相似文献

Air pollution induced changes in pine needle chemistry were observed at sample sites in the surroundings of the Pechenganikel smelter. Close to the smelter, elevated concentrations of Ni, Cu and S were found (Ni: 0.7–1 mmol/kg, Cu: 0.4–0.5, and S 40–60 mmol/kg). Close to the pollution source, needles were enriched in Ni and Cu by needle age. Correlation and principal component analyses show that changes in the element composition of pine needles depended on air pollution and on natural factors as well. The contribution from air pollution increased with needle age. Besides direct input of pollutants from atmosphere, soil contamination and nutritional disturbance contributed significantly to the observed changes.  相似文献

This article presents methods and devices developed by the authors for protecting cattle from high solar activity in the hottest period of the year. A new method for determining the individual resistance of cattle to a thermal stimulant has been developed. The research was carried out on cows and heifers of the blackmotley breed crossed with Holstein. The body temperature, respiratory rate, pulse rate, and cycle of cow’s rumen were determined. Highly resistant cows identified by this method better maintain homeostasis and show increased milk productivity compared to less resistant cows. It has been found that both the young and adult animals with the dominance of white color are characterized by a high resistance to solar radiation during the hottest period of the year compared to black-colored individuals. We studied the ways of attenuating a high sunlight intensity in loafing areas by shelters with different air-permeable coatings, which protect animals from direct sunlight, reducing its negative impact by 86.2%. The use of these shelters will make it possible to improve the zoohygienic cattle management conditions and reduce costs using a design with a low material consumption.  相似文献

In addition to strong natural stresses forest ecosystems in the Kola Subarctic, Russia, receive high loads of sulphur and heavy metals from the nickel smelter. To estimate soil response to acid deposition we compared the soil field data along a pollution gradient and simulated time effects. Multivariate technique was applied to investigate spatial distribution of soil field data. Time response of soils to acid deposition was evaluated with the SMART model. According to field observations there is no evidence for strong soil acidification effects close to the smelter. Concentrations of exchangeable Ca and base saturation increase, while acidity decrease in lower soil mineral horizons towards the pollution source. However, some features seem to reflect the early stages of the started acidification. Most soil profiles have low pH values. Despite increasing of exchangeable Ca and Mg towards the smelter in lower mineral horizons due to geological inheritance, they do not reveal the same trends in the upper ones. Concentration of exchangeable K in organic horizons decreases towards the smelter, thus confirming the starting acidification. As result, exchangeable base cations are depleted in the considerable part of shallow soil profiles. According to model simulation the present acid load does not effect considerably on forest soils in background areas, however, dramatic shift in soil chemistry near the smelter is expected within several decades. Due to low pool of exchangeable base cations and low weathering rate continued acid deposition can lead to increased soil acidification and nutrient imbalance.  相似文献
