The study was conducted to investigate the effect of relaxin on motility, acrosome reaction (AR), viability and utilization of glucose in fresh and frozen‐thawed bovine spermatozoa. Both semen samples were washed twice through centrifugation (5 min at 600 g), and preincubated for 1 h at 39°C for swim up. The swim‐up separated spermatozoa were resuspended in a sperm Tyrode's albumin lactate pyruvate (Sp‐TALP) medium containing 0 (control) and 40 ng/mL porcine relaxin and incubated for 0–6 h. Sperm motility was determined on the basis of movement quality examined by a phase contrast microscope. Sperm viability and AR were evaluated by using the triple staining technique. The incorporation and oxidation of 14C‐glucose was assessed by a liquid scintillation counter. Motility was improved (P < 0.05) in both fresh and frozen‐thawed spermatozoa by the addition of relaxin to the Sp‐TALP medium, whereas relaxin showed no significant effect on viability in either fresh or frozen‐thawed spermatozoa. The percentage of AR increased (P < 0.05) when fresh or frozen‐thawed spermatozoa were incubated with relaxin. In contrast, the incorporation and oxidation of 14C‐glucose increased (P < 0.05) in both kinds of spermatozoa incubated with relaxin. Thus the results demonstrated that the addition of relaxin to the Sp‐TALP medium increased the motility, AR and utilization of glucose in fresh and frozen‐thawed bovine spermatozoa.  相似文献

The aims of this study was to assess the effect of two lactic acid bacteria (LAB), Lactobacillus curvatus and Leuconostoc mesenteroides, originally isolated from gastrointestinal (GI) tract of beluga (Huso huso) and Persian sturgeon (Acipenser persicus), respectively, on growth, survival and digestive enzyme (amylase, lipase and protease) activities and the population level of LAB in the GI tract. The treatments included 10 different groups; control, separate supplements of Lcurvatus and Leumesenteroides at three different counts [2 × 109, 5 × 109 and 9 × 109 colony forming units (CFU) per gram food] and three combinations of the two LAB (2 × 109 + 2 × 109, 5 × 109 + 5 × 109 and 9 × 109 + 9 × 109 CFU per gram food). The bacteria used in this study were added in lyophilized form to chopped Chironomidae. In the beluga study, highest specific growth rate, survival and improved intestinal enzyme activities were noted in the rearing group fed 9 × 109 L. curvatus per gram food. In Persian sturgeon, the inclusion level of 2 × 109 Leu. mesenteroides had similar positive effect. The ability of LAB to colonize the digestive tract seems to involve host specificity, and our bacteriological results are relevant to initiate future probiotic studies in sturgeons and future directions will be discussed.  相似文献

This comparative study was to investigate the hypocholesterolemic effects of dietary Rhodopseudomonas palustris and Rhodobacter capsulatus on rats fed a high cholesterol diet. Thirty male Wister–Imamichi rats were assigned to three groups and fed on either a high cholesterol diet, or a high cholesterol diet supplemented with 2.0% R. palustris or R. capsulatus for 4 weeks. Compared to the control diet, both of the R. palustris and R. capsulatus supplemented diets significantly reduced the serum cholesterol, triglycerides, low‐density lipoprotein, very low density lipoprotein cholesterol and hepatic triglycerides, but increased hepatic cholesterol in rats. In addition, both of the R. palustris and R. capsulatus supplemented diets may reduce the risk of atherosclerosis, as the ratio of high density lipoprotein cholesterol to the total cholesterol was significantly higher than in the control group (P < 0.05). Both the R. palustris and R. capsulatus supplemented diets led to an increase in the serum palmitic acid, compared with the oleic acid and linoleic acid. No significant differences were postulated between the rats fed R. palustris and R. capsulatus supplemented diets during the 4 weeks of the experimental period. Thus, the results may suggest that both R. palustris and R. capsulatus can contribute significant health benefits and seems to be feasible to investigate in future research.  相似文献

The study was designed to investigate the effect of Rhodobacter capsulatus on serum and meat cholesterol, fatty acid composition in meat, as well as meat quality of finishing pigs. A total of 16 120-day-old Landrace female pigs of about 60 kg initial body weight were randomly assigned into two groups. The pigs were fed a supplemented diet with 0.04% dietary R. capsulatus or a control diet for 60 days. Total cholesterol and low density lipoprotein-cholesterol concentration in serum was significantly lowered ( P  < 0.05) in the pigs fed the R. capsulatus supplemented diet compared to the control diet. Carcass weight, carcass length, shoulder fat, back fat and loin fat thickness, longissimus muscle (LM) area, and color score did not differ significantly between the pigs fed the R. capsulatus supplemented diet and control diet. Among the nutrient composition of LM meat, neutral fat and triglyceride concentration were significantly ( P  < 0.05) reduced in the finishing pigs by dietary supplementation of R. capsulatus . The proportions of n-6 PUFA were higher ( P  < 0.05) in the pigs fed the R. capsulatus supplemented diet than in the pigs fed the control diet. The supplementation of dietary R. capsulatus to finishing pig diet played important roles in reducing serum cholesterol and meat triglycerides, as well as in increasing polyunsaturated fatty acid content in LM meat.  相似文献
