The role that environmental contamination might play as a reservoir and a possible source of Methicillin-resistant Staphylococcus aureus (MRSA) for patients and personnel at equine veterinary hospitals remains undefined, as the environment has only been monitored during outbreaks or for short periods. Therefore, the objectives of this study were to determine the monthly presence, distribution, and characteristics of environmental MRSA at an equine hospital, and to establish patterns of contamination over time using molecular epidemiological analyses. For this purpose, a yearlong active MRSA surveillance was performed targeting the environment and incoming patients. Antimicrobial susceptibility testing, SCCmec typing, PFGE typing, and dendrographic analysis were used to characterize and analyze these isolates. Overall, 8.6% of the surfaces and 5.8% of the horses sampled were positive for MRSA. The most common contaminated surfaces were: computers, feed-water buckets, and surgery tables-mats. Ninety percent of the isolates carried SCCmec type IV, and 62.0% were classified as USA500. Molecular analysis showed that new pulsotypes were constantly introduced into the hospital throughout the year. However, maintenance of strains in the environment was also observed when unique clones were detected for 2 consecutive months on the same surfaces. Additionally, pulsotypes were circulating throughout several areas and different contact surfaces of the hospital. Based on these results, it is evident that MRSA is constantly introduced and frequently found in the equine hospital environment, and that some contact surfaces could act as “hot-spots”. These contaminated surfaces should be actively targeted for strict cleaning and disinfection as well as regular monitoring.  相似文献

Trace elements (Cu, Fe, Pb, Hg, Cd, As, Mn, Zn) were analyzed quantitatively in 14 wheat samples collected from fields in all Serbian growing regions, harvested in 2002. Microelements were determined according to an atomic absorption spectrophotometric method. Principal component analyses (PCA) were performed on data matrices consisting of contents of trace elements in wheats (columns) and all Serbian wheat-growing regions (rows). It was found that four principal components account for 87.2% of the total variance in the data. The plot of component loadings showed significant groupings for concentration of some microelements. The component scores indicated the similarities among the Serbian wheat-growing regions. The loading plot reveals that there is no need to measure all of the variables to achieve the same classification. It is enough to measure one variable per group. Naturally, this conclusion is valid only within the limits of the present study of wheat grain samples from different parts of Serbia.  相似文献

Symptoms of brown blotch were observed on cultivated Agaricus bisporus on several mushroom farms in Serbia. Subsequently, samples were collected from nine localities during 2006?C2010 and fluorescent Gram-negative bacterial strains were isolated. All strains caused sunken brown lesions on A. bisporus tissue blocks after artificial inoculation. Isolated strains showed the LOPAT characteristics of the group Va being negative for levan production; the ability to produce a hypersensitivity reaction in tobacco; were oxidase and arginine dihydrolase positive; and lacked pectolytic activity. The results from other tests were as follows: oxidative metabolism of glucose; catalase activity positive; aesculin hydrolisis negative; Tween 80 hydrolysis positive; casein hydrolysis positive; gelatine hydrolysis and nitrate reduction negative; mannitol, erythritol, sorbitol, inositol and trehalose were used as a carbon source but not sucrose, arabinose and D(-) tartrate. Based on these features bacterial strains were identified as Pseudomonas tolaasii. The identity of isolated strains was confirmed by polymerase chain reaction. This is the first report of P. tolaasii in Serbia.  相似文献

This study compares the results and suitability of serological testing, microscopic examination, deoxyribonucleic acid (DNA) detection, and bacterial culture for detecting Mycobacterium avium subsp. paratuberculosis (Map) infection in asymptomatic farmed white-tailed deer (WTD) (Odocoileus virginianus). Deer were classified as infected if culture slants from their feces, lymph nodes, or ileum were positive, or if a polymerase chain reaction (PCR) assay detected Map DNA in any of its tissues. Deer identified as positive by agar gel immunodiffusion (AGID) testing or enzyme-linked immunosorbent assay (ELISA) but not by bacterial culture, Ziehl-Neelsen staining, or PCR assay were classified as suspect. Culture of tissues classified 10/16 (62.5%), histopathologic examination 1/16 (6.3%), tissue smears 4/16 (25%), culture slant (CS)-PCR on feces 12/15 (80%), CS-PCR on tissue 13/16 (81.3%), and direct PCR on uncultured tissues 5/16 (31.3%) deer as infected. The ELISA classified 2/15 (13.3%) deer as positive and therefore suspect. The AGID test was negative for all deer. Fifteen of 16 deer were positive by 1 or more tests; only 1 deer was negative on all 11 assays. The CS-PCR gave superior results on antemortem fecal testing as well as postmortem tissue testing and can be recommended for improving the detection of Map in WTD at every stage of infection.  相似文献

The essential oils of rosemary ( Rosmarinus officinalis L.) and sage ( Salvia officinalis L.) were analyzed by means of gas chromatography-mass spectrometry and assayed for their antimicrobial and antioxidant activities. Antimicrobial activity was tested against 13 bacterial strains and 6 fungi, including Candida albicans and 5 dermatomycetes. The most important antibacterial activity of both essential oils was expressed on Escherichia coli, Salmonella typhi, S. enteritidis, and Shigella sonei. A significant rate of antifungal activity, especially of essential oil of rosemary, was also exhibited. Antioxidant activity was evaluated as a free radical scavenging capacity (RSC), together with the effect on lipid peroxidation (LP). RSC was assessed by measuring the scavenging activity of essential oils on 2,2-diphenyl-1-picrylhydrazil (DPPH) and hydroxyl radicals. Effects on LP were evaluated following the activities of essential oils in Fe(2+)/ascorbate and Fe(2+)/H2O2 systems of induction. Investigated essential oils reduced the DPPH radical formation (IC50 = 3.82 microg/mL for rosemary and 1.78 microg/mL for sage) in a dose-dependent manner. Strong inhibition of LP in both systems of induction was especially observed for the essential oil of rosemary.  相似文献
