Fasted Brazilian squirrel monkeys (BrSMs) exhibited slightly higher serum bilirubin levels (0.30±0.05 mg/dl) than others in the fed state (0.13±0.01). The mean liver weight was 50% lower following a 22 h fast. The rate of bile flow was unaffected by fasting and averaged 13.8 l/min/kg and 47.5 l/min/100g liver in six BrSMs. No significant difference in mean bilirubin excretion/min was observed on a body weight basis following fasting. When the mean rate of bilirubin excretion was calculated as a function of liver weight, a two-fold higher rate was present in fasted monkeys, but only at the p=0.06 level of statistical significance. From data collected in this and earlier studies, it would appear that BrSMs represent the best animals studied to date to serve as experimental controls in comparative studies with Bovilian squirrel monkeys which exhibit a Gilbert-like syndrome.  相似文献

Five hundred forty crossbred pigs were utilized in four trials (10 replications) at two stations to determine the separate and interacting effects of decreasing floor space allowance with or without the addition of virginiamycin to the diet on performance of growing-finishing pigs. Pigs were allowed .37, .33 or .28 m2/pig during the growing phase (23 to 55 kg) and .74, .66, .56 m2/pig during the finishing phase (55 to 100 kg) and fed either a corn-soybean meal control diet or the control diet plus 11 mg of virginiamycin/kg of diet. Floor space allowances were achieved by varying pen size, so the number of pigs and feeder space per pig were constant for all pens. During the growing phase, daily gain (P less than .01) daily feed intake (P less than .05) and feed conversion ratio (P less than .05) decreased as floor space allowance decreased. During the finishing phase and for the total test period, daily gain (P less than .01) and feed conversion ratio (P less than .05) were reduced but daily feed intake was not affected (P greater than .05) by restricted floor space allowance. The addition of virginiamycin to the diet had no effect on pig performance. The virginiamycin X floor space allowance interaction was not significant, suggesting that pigs fed a diet with or without virginiamycin responded similarly to restricted floor space allowance. The data suggest that the addition of virginiamycin to the diet was ineffective in overcoming the decrease in performance of growing-finishing pigs caused by crowded conditions.  相似文献

Sera obtained from pigs before and after subtherapeutic levels of ASP250 supplementation (pre and post serum pools) have been subjected to comparative fractionation by using gel filtration and affinity chromatography on immobilized Cibacron Blue F3G-A. Comparable serum fractions obtained from pre- and post-ASP250 blood sera were assayed in muscle cell culture bioassays designed to measure their effect on proliferation. Pre- and post-ASP250 sera were subjected to gel filtration and divided into the following fractions: fraction 1, Kav less than .17; fraction 2, Kav = .17 to .41; fraction 3, Kav = .41 to .59. Post-ASP250 fractions 2 and 3 increased proliferation rate in cultured muscle cells to a greater extent than comparable pre-ASP250 fractions (P less than .001). Chromatography of fraction 3 on immobilized Cibacron Blue F3G-A showed that both pre- and post-ASP250 fraction 3 contained a putative inhibitor of myogenic cell proliferation as well as mitogenic factors. However, negative growth factor activity was greater in pre-ASP250 fraction 3 than in post-ASP250 fraction 3 (P less than .05). Additionally, positive growth factor activity was lower in pre-ASP250 fraction 3 than in post-ASP250 fraction 3 (P less than .05). These data suggest that levels and(or) activities of both positive and negative muscle growth factors in serum may be altered by the addition of antimicrobials to the diets of growing pigs.  相似文献

A liquid chromatography–tandem mass spectrometry method was developed to confirm the presence of androstenedione-19-oic acid in intact male equine plasma and to show the source of 19-norandrostenedione in equine plasma. Androstenedione-19-oic acid was recovered from acidified plasma by liquid–liquid extraction using methyl tert-butyl ether and separated on an Ace 5 C8 column. A triple quadrupole mass spectrometer was used to detect the analytes in negative electrospray ionization mode. Limits of detection, quantification, and confirmation of the method were 0.1, 0.5, and 1.0 ng/mL, respectively. The linear dynamic range of quantification was 0.5–50 ng/mL. The presence of androstenedione-19-oic acid was confirmed in all plasma samples obtained from intact male horses but not those from gelded and female horses; the average concentration was 3.1 ± 1.6 ng/mL, suggesting androstenedione-19-oic acid is an endogenous compound only in intact male horse plasma samples. The conversion of androstenedione-19-oic acid to 19-norandrostenedione in equine plasma was demonstrated by spiking androstenedione-19-oic acid into blank plasma and monitoring the generation of 19-norandrostenedione and its increase in concentration during storage. Results indicated that androstenedione-19-oic acid was readily converted into 19-norandrostenedione; the higher the storage temperature, the faster the conversion. The conversion was not affected by the types of plasma samples collected from gelded and female horses or by anticoagulants used in blood collection to harvest plasma. Compared with other matrices such as water, methanol, and phosphate-buffered saline, the conversion of androstenedione-19-oic to 19-norandrostenedione in equine plasma was faster, suggesting that there is an unknown factor(s) in equine plasma that enhances the conversion.  相似文献

Summary These investigations were conducted to determine the effectiveness of first- and second-season selection programs for increasing forage and seed yield in kura clover (Trifolium ambiguum Bieb.).Rhizo kura clover, the base population, was grown as spaced plants in two programs for six generations of selection for flowering in the seedling and second seasons. Remnant seed for each generation of selection was used to establish spaced plant and broadcast evaluation trials.The first-season selection program was effective in increasing flowering not only in the first season, but for two subsequent seasons. The second-season selection for flowering had little or no effect. However, vigor, forage and seed yields were not increased by either program, and the first-season program progressively reduced vigor and forage yields as generations of selection increased. The cause of this is unknown but may be associated with inbreeding depression or detrimental physiological effects of first-season flowering. Reselection to eliminate possible inbreeding effects is being conducted to further examine the possibility of increasing seedling and aftermath vigor in kura clover.  相似文献

Summary This study was conducted using the isozymes ACP-1, ADH-1, GOT-2, GOT-3, MDH, 6-PGD-1 and PGI-2 to: a) compare isozyme banding patterns of tall fescue somaclones with parents and b) correlate tissue culture-induced chromosome abnormalities with variant banding patterns. The 174 somaclones were grouped into seven categories based on their meiotic analyses and time of regeneration from culture. Differences in isozyme frequency between categories compared by chi-square tests were greatest for MDH, 6-PGD-1 and PGI-2, and least for ACP-1. The most significant differences in frequency were found between somaclones and parents. In comparisons of somaclone categories, the most different isozyme distributions were between the early vs. late regenerated somaclones. No significant differences in isozyme frequencies were found between all 42-chromosome somaclones vs. aneuploid somaclones and the three somaclone groups (42-normal, 42-abnormal, aneuploid) compared to each other. This study suggests that culture-induced isozyme variation alters the distribution of the isozyme phenotypes, but is not directly correlated with chromosome abnormalities.  相似文献

Soil properties were evaluated after 10 years of continuous non-tilled and conventionally tilled corn (Zea mays L.) production on a Maury silt loam (Typic Paleudalfs) soil, which had been in bluegrass (Poa pratensis L.) for 50 years. On limed and nonlimed plots soil samples from 0, 84, 168 and 336 kg/ha N treatments were taken in the 0–5, 5–15 and 15–30 cm layers for determination of organic C and N, soil pH, and exchangeable Al, Mn, Ca, Mg, K.Tillage treatments had no effect on soil bulk density in the 0–15 cm layer. In the 0–5 cm surface layer, organic C and N were approximately twice as high with no-tillage as with conventional tillage; N fertilizer induced a high level of both organic C and organic N. No-tillage decreased soil pH for unlimed plots as compared to conventional tillage, especially at high N-rates, which produced an increase in exchangeable Al and Mn and a decrease in exchangeable Ca down to the 30 cm depth. When lime was applied, the pH of the surface soil was slightly higher under no-tillage. On treatments receiving lime, exchangeable Al and Mn levels were very low with no significant difference in tillage systems. At low rates of N fertilization the 10-year average corn yield was higher for conventional tillage than for no-tillage, but at high rates of N fertilization it was equal or higher for no-tillage treatments receiving lime. Unlimed no-tillage treatments produced lower yields at all N levels during 1975–1979. Deterioration of soil physical properties was not observed.  相似文献

Utilizing soil electrical conductivity (EC) measurements and terrain attributes for precision management will require secondary soil information for adequate interpretation. The objective of this study was to determine whether readily available second-order soil surveys were of adequate quality to aid with interpreting soil EC and terrain data. For three locations in Kentucky, USA, first-order soil surveys were created, second-order surveys reports were obtained, elevation was measured and used to calculate terrain attributes (slope, aspect, plan curvature, profile curvature), and bulk soil electrical conductivity was measured. Three analytical methods (an ordinary least squares analysis and two random field analyses), visual map assessment, and examination of least-squares means were used to assess the relationships between soil EC measurements, terrain attributes and first- and second-order soil surveys. The OLS and random field analyses were problematic. However, the ranking of the OLS F-statistics appeared to reflect the general relationship between landscape variables and first-order soil surveys. The landscape variables related particularly well with soil properties that had been impacted by past soil erosion. Unfortunately, however, second-order soil surveys in this study were not created at suitable scales to adequately interpret EC and terrain data regarding erosion history or other attributes. While these surveys may provide some useful information, field measurements, sampling, and observations will likely be required to develop high quality interpretations of soil EC and terrain attribute data.  相似文献

In agriculture and plant breeding the most important genotype × environment interaction pattern is crossover genotype × environment interaction. Procedures using linearbilinear models are used to find disjoint subsets of sites or genotypes with negligible crossover genotype × environment interaction. In terms of correlations, these subsets of sites or genotypes should be such that pairs of sites have nearly perfect positive genetic correlations. Perfect positive genetic correlation between sites is a sufficient, but not a necessary, condition for a non-COI pattern. The main objective of this study was to use the mixed model theory to confirm that the subsets of sites and genotypes formed by using the linear-bilinear models have negligible crossover genotype × environment interaction. If there is no significant crossover interaction in a subset of sites or genotypes, then the mixed model should be able to confirm this by nonrejection of the hypothesis that the covariance structure has perfect genetic correlation. Mixed model analysis of results from two multisite cultivar trials confirmed the validity of the procedures using linear-bilinear model methods for clustering sites and genotypes into noncrossover genotype × environment interaction subsets.  相似文献

Resistance of Salmonella to extended-spectrum cephalosporins (ESCs) is being reported with increasing frequency. In humans, infections with Salmonella resistant to ESCs threaten the efficacy of ceftriaxone, the drug of choice for treating salmonellosis in children. To determine the occurrence of resistance to ESCs, we examined 8426 strains isolated from food-producing animals in Canada in 1994–99 for reduced susceptibility or resistance to ceftriaxone. Of the 8 such strains identified (7 from turkeys and 1 from cattle), 5 had reduced susceptibility, and 3 were resistant; 2 were isolated in 1995, 1 was isolated in each of 1996 and 1997, and 4 were isolated in 1999. Isoelectric focusing showed that all 8 isolates produced a β-lactamase with a pI ≥ 9. The strains were resistant to cefoxitin and not inhibited by clavulanic acid. Primers specific for the Citrobacter freundii blaAmpC gene produced the expected product in the polymerase chain reaction. DNA sequencing showed that all isolates possessed the blaCMY-2 gene. Plasmid DNA from all 8 isolates transformed Escherichia coli DH10B, whereas only 1 isolate transferred blaCMY-2 conjugally. All transformants and the transconjugant were resistant to ampicillin, cefoxitin, ceftiofur, cephalothin, streptomycin, sulfisoxazole, and tetracycline. Southern blots of plasmids from the isolates, the transformants, and the transconjugant showed that blaCMY-2 was located on similar-sized plasmids (60 or 90 MDa) in the transformants and the transconjugant. In the S. Typhimurium DT104 and S. Ohio isolates, the floSt gene was found on the same plasmid. Class 1 integrons with the aadB gene cassette were detected in the S. Bredeney isolates but not in their transformants or the transconjugant. Pulsed-field gel electrophoresis and plasmid profiles indicated that both clonal dispersion and horizontal transfer of blaCMY-2 may have caused dissemination of the resistance determinant.  相似文献
