Matrix protein is known as a type-specific structural protein of influenza viruses. An attempt has been made to find out whether or not strain-specific components could be detected from matrix protein, in addition to its type-specific antigen determinants. The technique of enzyme immune assay was chosen as the optional method to differentiate between matrix proteins of various influenza-A viruses. Antigen titration was undertaken of several matrix proteins, using two specific anti-matrix-protein sera in each case. Information regarding serological relationships between the tested matrix proteins of various influenza-A viruses was obtained from a quotient between the titres of one antigen, on the one hand, and the two anti-matrix-protein sera used in titration, on the other. Two matrix protein sub-types were established in the context of the influenza-A viruses tested. Sub-type M1 was attributable to older strains (A/PR/8 and A/FM/1), whereas the matrix protein of sub-type M2 was found to be present in more recent strains (A/Hongkong and A/Port Chalmers). 相似文献

Adult C57BL/6J male mice received either a primary ectopic transplant or a primary and a challenge ectopic transplant of trophoblast tissue obtained from the ectoplacental cones of 7(1/2)-day-old C3H/HeJ x C3H/HeJ embryos. Gross and histologic examinations of these grafts at 5, 7, and 12 days of growth indicated that the challenge grafts were inhibited in growth; there were a smaller percentage of grossly successful grafts, smaller hemorrhagic reactions, fewer viable cells at all stages of growth, and better host containment of the colony. The evidence indicates that exposure to pure trophoblast alters an animal's subsequent reaction to grafts syngeneic with the original trophoblast and can best be explained as evidence of immunologic sensitization of the host. 相似文献

A young adult, 28 kg, intact female Siberian husky-mixed breed dog was presented to the Veterinary Medical Teaching Hospital (VMTH) Emergency Service for evaluation of a dysrhythmia and an abnormal cardiac structure. The dog had been adopted from an animal shelter approximately 3 months before presentation. The dog occasionally showed exercise intolerance, and the owners suspected that she had impaired vision. Initial clinical signs appeared 2 days before presentation during strenuous activity—the dog uncharacteristically stopped and lay down while tachypneic. In a 5-year retrospective study of dogs presenting to the Veterinary Medical Teaching Hospital at the University of California, Davis, there were 31 histologic diagnoses of valvular endocarditis. By polymerase chain reaction (PCR) amplification of embedded valvular tissue, Bartonella organisms were exclusively associated with 6 out of 31 cases (19%). Confirmed Bartonella cases involved the aortic valve alone (five out of six) or in combination with the mitral valve (one of six). Microscopic features of Bartonella endocarditis were compared with valves from non-Bartonella endocarditis and with valvular change unrelated to infectious agents (endocardiosis). Features of Bartonella endocarditis included a combination of fibrosis, mineralization, endothelial proliferation, and neovascularization with variable inflammation. None of these features is specific; however, the combination is distinct both from endocarditis caused by culturable bacteria and from endocardiosis. Ultrastructural analyses revealed both extracellular and intraendothelial bacteria. Clinical history, serology, and PCR are currently necessary to establish an etiologic diagnosis of Bartonella endocarditis. 相似文献

The Southern sea otter (Enhydra lutris nereis) is listed as threatened under the Endangered Species Act. The population began a pattern of slow decline in 1995. The decline was attributed to high adult mortality rates with infectious disease being the major cause of death. Multiple pathogens were implicated in these deaths including opportunistic pathogens such as Coccidiodes immitis and Toxoplasma sp. These findings suggested that the immunological health of mature animals in this population might be compromised. The primary goal of this study was to establish techniques for assessing phenotypic and functional baseline data for peripheral blood mononuclear cells (PBMC) in free-ranging sea otters. Standard total and differential white blood cell counts were augmented by emumeration of T and B lymphocyte subsets. Lymphocyte function was determined by both mitogen-induced proliferation and expression of IL-2 receptors. In addition to establishing normal ranges for adult animals, age-related changes were identified in B lymphocyte numbers and cell-surface density of major histocompatability complex class II (MHC II) proteins. The predominant lymphocyte subpopulation in Southern sea otters is the T lymphocyte. Substantial variation among individual animals was observed within the B lymphocyte population both in cell number and density of MHC II expression. Pups had greater numbers of T and B lymphocyte, as well as, greater MHC II expression on B lymphocytes than adults. Mitogen-induced proliferation of peripheral blood mononuclear cells (PBMC) was variable among individual animals with no significant difference in cell response between age class and gender. Concanavalin (ConA) was a more effective mitogen in stimulating proliferation and interleukin (IL)-2 receptor expression than pokeweed. This data can be used to augment routine hematology profiles and aid in the identification of animals with immunologic perturbations. 相似文献

Most pathogens that cause bovine mastitis invade the udder lumen through the teat canal. Amino acids and intercellular lipids may support microbial colonisation of the teat canal epithelium by pathogenic microorganisms. The aim of this study was to investigate the association between teat end hyperkeratosis, which is induced by machine milking, and teat canal microbial load. Contralateral teats, which differed in teat end hyperkeratosis scores, were identified in a split-udder experiment. The teat canal's microbial load was evaluated using the wet and dry swab technique. Staphylococcus (S.) aureus, Streptococcus (Sc.) uberis, Escherichia (E.) coli and other coliforms were detected by agar plate cultures. The positive detection of E. coli and the log(10)-transformed E. coli load of a teat canal were significantly associated with the teat end hyperkeratosis score (P<0.05). There were significant differences with respect to positive findings for E. coli, as well as the microbial load of E. coli and Sc. uberis, between the less-calloused and the more-calloused teat of a pair. For S. aureus, no significant associations between hyperkeratosis score and teat canal microbial load were detected. In general, a teat with a highly calloused teat end had an increased teat canal microbial load compared with the contralateral teat, characterised by a lower callosity. The results of the present study indicate that the environmental pathogen load is associated with teat end hyperkeratosis. Further research is needed to identify factors that may affect teat canal microbial load in lactating dairy cattle. 相似文献

Nitrogen fertilizer practices affect nitrous oxide (N2O) emissions from agricultural soils. The “4R” nutrient stewardship framework of using N fertilizer at the right rate, right source, right placement and right time can reduce N2O emissions while maintaining or improving yield of field crops, but understanding of how the various factors affect N2O emissions from irrigated processing potato is lacking. We examined the effects of selected 4R practices on emissions, using results from two irrigated processing potato studies each conducted in 2011 and 2012 in Manitoba, Canada. Experiment 1 examined combinations of source (urea, ESN), placement (pre-plant incorporation [PPI], banding), and rate (100 and 200 kg N ha-1) on a clay loam soil. Experiment 2 examined timing and source treatment combinations (urea PPI, ESN PPI, urea split, urea split/fertigation) on a loamy fine sandy soil. For Experiment 1, use of ESN at 200 kg ha-1 did not reduce area-, yield- and applied fertilizer N- based N2O emissions compared to urea at 200 kg ha-1, irrespective of placement. Emissions from pre-plant banding ESN at 200 kg ha?1, however, were 32% lower than from PPI ESN. For Experiment 2, compared to single pre-plant urea application, fertigation simulated by in-season application of urea ammonium nitrate (UAN) gave lower area-, yield- and applied fertilizer N- based emissions. Split urea ( \( \raisebox{1ex}{$2$}\!\left/ \!\raisebox{-1ex}{$3$}\right. \) pre-plant, \( \raisebox{1ex}{$1$}\!\left/ \!\raisebox{-1ex}{$3$}\right. \) hilling) also reduced area- and yield- based N2O emissions compared to single pre-plant urea application. Emissions were generally lower at the site with loamy fine sandy soil than the site with clay loam soil. This study aims to investigate the expression of metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) in chronic doxorubicin cardiomyopathy in a rabbit model and to evaluate the effects of bone marrow-derived mesenchymal stem cell (MSC) transplantation in this disease. Thirty-nine 3-month-old New Zealand rabbits were divided into 4 groups: group 1 (n = 9) was the untreated control. Groups 2-4 were treated with 6 weeks of doxorubicin (3 mg/kg). Group 2 (n = 6) received no further treatment. In group 3 (n = 9), animals were treated with culture medium (CM) alone. In group 4 (n = 15), autologous MSCs (1.5-2.0 x 10(6)/ml) were injected in the left ventricular (LV) wall. Hearts were stained with HE and picrosirius red. MMP-1, -2, -3 and -9 and TIMP-2 and -3 were detected immunohistochemically. The mRNA levels were determined by real-time polymerase chain reaction. The results confirmed that doxorubicin treatment resulted in minimal myocardial fibrosis and showed that expression of MMPs increased and TIMP-3 decreased. The injection procedure resulted in increased myocardial fibrosis in groups 3 and 4. After MSC injection, MMP-1, MMP-2, and TIMP-3 expression was higher than that in group 2. CM injection led to more fibrosis, elevated TIMP-3, but diminished MMP-1 and MMP-2 expression compared with MSC injection. The mRNA levels of MMPs and TIMPs were not significantly different among all groups. In conclusion, chronic doxorubicin cardiomyopathy was characterized by increased MMP and decreased TIMP-3 expression. MSCs injection into the LV resulted in marked differences of collagen content and MMP/TIMP expression in the whole heart, although significant numbers of living MSCs were not detected after 4 weeks. 相似文献
