To produce large quantities of primary cell suspensions on efficient technique is described which is named "Biomix-Method". By this technique first tissue mince is performed automatically using tissue amounts between 300 g and 500 g. Trypsinization takes place in a three-litre trypsinization vessel using a stirring mechanism according to a defined trypsinization mode. In regard to optimal cell seeding concentration and a justifiable productivity one kidney of newborn or suckling calf yields 6--9 1 cell suspension and that of piglet 1,8-2,3 1, respectively. Proliferation behaviour and effectivity of primary cells are discussed and compared with other cell systems.  相似文献

Prolonged exposure to captive conditions has led to the development of a rainbow trout ‘farmed’ phenotype, which is different from that of wild trout. Selection for desirable productive traits in hatcheries has resulted in the development of some morphological traits that are maladaptive in nature. The recent development of organic aquaculture, guided by the well‐being of the fish, could potentially produce a new farmed phenotype that would be more adaptive in nature. In this study, rainbow trout reared in intensive and organic farms were compared by means of shape analysis, to detect patterns of shape variation associated with rearing environment. The results of this study highlight a significant effect of the rearing method on rainbow trout shape: organically reared trout showed a higher body profile, in particular in the head and trunk regions, shorter median fins and a deeper caudal peduncle. A combined effect of density and habitat complexity could have contributed to the observed shape differences: in organic rearing systems, lower densities and steady water could increase territoriality and aggressive interactions, promoting body designs more functional in rapid attacks and escapes.  相似文献

Plants of the family Brassicaceae contain high levels of glucosinolates. The latter compounds are degraded to isothiocyanates, some of which have been shown to be potent inducers of phase II detoxification enzymes in vitro. In the present study, the ability of six plant-derived isothiocyanates (allyl isothiocyanate, iberverin, erucin, sulforaphane, iberin, and cheirolin) to increase tissue levels of the phase II detoxification enzymes quinone reductase (QR) and glutathione S-transferase (GST) in a variety of rat tissues has been compared. At the low dose level employed (40 micromol/kg/day), cheirolin was without effect in any tissue. All of the other isothiocyanates, however, increased GST and QR activities in the duodenum, forestomach, and/or the urinary bladder of the animals, with the greatest effects being seen in the urinary bladder. With the exception of cheirolin, little difference was observed in the inductive activity of the various isothiocyanates. Phase II enzymes are known to protect against chemical carcinogenesis, and the selectivity of isothiocyanates in inducing such enzymes in the bladder is of interest in view of recent epidemiological studies showing a decreased incidence of cancer of this organ in individuals with a high dietary intake of Brassica vegetables.  相似文献

The relationship between pasta texture and physicostructural characteristics was determined in relation to potential starch degradation and subsequent glucose release. Pastas with added soluble and insoluble dietary fiber ingredients were evaluated in relation to biochemical composition, cooking properties, and textural characteristics. Results show that both the type and amount of added fiber influence the overall quality of both raw and cooked pasta. Glucose release may be significantly reduced by the addition of soluble dietary fiber.  相似文献

Pot plants of 12 black currant genotypes were assessed in frost simulation chambers for their tolerance of spring frosts during the flowering period. The most tolerant were some genotypes from the UK and Scandinavia. The inheritance of tolerance may be complex rather than simply additive. The results indicate that physiological frost hardiness at flowering is needed as well as later flowering, particularly when breeding for early ripening cultivars.  相似文献

An experiment was conducted to determine how an E. coil challenge and dietary clays affect the intestinal barrier of pigs. Two groups of 32 pigs (initial BW: 6.9 ± 1.0 kg) were distributed in a 2 x 4 factorial arrangement of a randomized complete block design (2 challenge treatments: sham or E. coil, and 4 dietary treatments: control, 0.3% smectite A, 0.3% smectite B and 0.3% zeolite), with 8 replicates total. Diarrhea score, growth performance, goblet cell size and number, bacterial translocation from intestinal lumen to lymph nodes, intestinal morphology, and relative amounts of sulfo and sialo mucins were measured. The E. coli challenge reduced performance, increased goblet cell size and number in the ileum, increased bacterial translocation from the intestinal lumen to the lymph nodes, and increased ileal crypt depth. One of the clays (smectite A) tended to increase goblet cell size in ileum, which may indicate enhanced protection. In conclusion, E. coli infection degrades intestinal barrier integrity but smectite A may enhance it.  相似文献

A mycosis in larval tiger prawn, Penaeus monodon Fabricius, is described for the first time from India. The hyphae of Lagenidium callinectes are contorted, irregularly branched, sparingly septate, and contain a cell wall and membrane, vacuoles, mito chondria, ribosomes, small and large vesicles, and Woronin bodies. The spores occur singly or in pairs. The fungal mycelium may either invade and embed itself in the tissues, or alternatively, replace all the muscle tissues of the infected larval P. monodon. Fungus infected, untreated populations of nauplii, zoea and myksis exhibited mortalities of 5.33 ± 0.55%, 24.68 ± 4.58% and 47.89 ± 0.27%, respectively. A 0.5 ppm treatment with trifluralin significantly reduced the mortality of infected larval populations (i.e. 1.1% nauplii, 3.28% zoea and 5.21% myksis mortality). Lagenidium sp. exhibited growth in potato dextrose agar medium and in Sabouraud's agar at 28 °C.  相似文献

The fatty acid composition and total fatty acid content of seeds from 36 blackcurrant genotypes developed at the Scottish Crop Research Institute were examined. A rapid small-scale procedure, involving homogenization of seeds in toluene followed by sodium methoxide transesterification and gas chromatography, was used. There was considerable variation between genotypes. The gamma-linolenic acid content generally varied from 11 to 19% of the total fatty acids, but three genotypes had higher values of 22-24%, levels previously not reported for blackcurrant seed and similar to those for borage seed. Other nutritionally important fatty acids, stearidonic acid and alpha-linolenic acid, varied from 2 to 4% and 10-19%, respectively. The mean total fatty acid contents ranged from 14 to 23% of the seed, but repeatability was poor. The results are discussed. Blackcurrant seeds are mainly byproducts from juice production, and the study shows the potential for developing blackcurrant genotypes with optimal added value.  相似文献
