Among extant large reptiles, juveniles often occupy different habitats from those of adults or subadults and thus avoid competition with and predation from the older animals; small juveniles often choose cryptic habitats because they are vulnerable to a wide variety of predators. Evidence from fossil humeri and femora of Early Permian reptiles collected from sediments of several distinct habitats indicate that similar shifts in habitat occurred. Juvenile Dimetrodon seem to have favored cryptic habitats around swamp and swampy lake margins; adults favored open habitats on the floodplains. Similar patterns of habitat shift seem to be present in the reptile Ophiacodon and the amphibian Eryops and may have been common in fossil tetrapods of the Permian-Triassic.  相似文献

Engineering resistance against various diseases and pests is hampered by the lack of suitable genes. To overcome this problem we started a research program aimed at obtaining resistance by transfecting plants with genes encoding monoclonal antibodies against pathogen specific proteins. The idea is that monoclonal antibodies will inhibit the biological activity of molecules that are essential for the pathogenesis. Potato cyst nematodes are chosen as a model and it is thought that monoclonal antibodies are able to block the function of the saliva proteins of this parasite. These proteins are, among others, responsible for the induction of multinucleate transfer cells upon which the nematode feeds. It is well documented that the ability of antibodies to bind molecules is sufficient to inactivate the function of an antigen and in view of the potential of animals to synthesize antibodies to almost any molecular structure, this strategy should be feasible for a wide range of diseases and pests.Antibodies have several desirable features with regard to protein engineering. The antibody (IgG) is a Y-shaped molecule, in which the domains forming the tips of the arms bind to antigen and those forming the stem are responsible for triggering effector functions (Fc fragments) that eliminate the antigen from the animal. Domains carrying the antigen-binding loops (Fv and Fab fragments) can be used separately from the Fc fragments without loss of affinity. The antigen-binding domains can also be endowed with new properties by fusing them to toxins or enzymes. Antibody engineering is also facilitated by the Polymerase Chain Reaction (PCR). A systematic comparison of the nucleotide sequence of more than 100 antibodies revealed that not only the 3′-ends, but also the 5′-ends of the antibody genes are relatively conserved. We were able to design a small set of primers with restriction sites for forced cloning, which allowed the amplification of genes encoding antibodies specific for the saliva proteins ofGlobodera rostochiensis. Complete heavy and light chain genes as well as single chain Fv fragments (scFv), in which the variable parts of the light (VL) and heavy chain (VH) are linked by a peptide, will be transferred to potato plants. A major challenge will be to establish a correct expression of the antibody genes with regard to three dimensional folding, assembly and intracellular location.  相似文献

Forage technology has been successfully introduced into smallholder cattle systems in Cambodia as an alternative feed source to the traditional rice straw and native pastures, improving animal nutrition and reducing labour requirements of feeding cattle. Previous research has highlighted the positive impacts of forage technology including improved growth rates of cattle and household time savings. However, further research is required to understand the drivers, challenges and opportunities of forage technology for smallholder cattle households in Cambodia to facilitate widespread adoption and identify areas for further improvement. A survey of forage-growing households (n = 40) in July–September 2016 examined forage technology adoption experiences, including reasons for forage establishment, use of inputs and labour requirements of forage plot maintenance and use of forages (feeding, fattening, sale of grass or seedlings and silage). Time savings was reported as the main driver of forage adoption with household members spending approximately 1 h per day maintaining forages and feeding it to cattle. Water availability was reported as the main challenge to this activity. A small number of households also reported lack of labour, lack of fencing, competition from natural grasses, cost of irrigation and lack of experience as challenges to forage growing. Cattle fattening and sale of cut forage grass and seedlings was not found to be a widespread activity by interviewed households, with 25 and 10% of households reporting use of forages for these activities, respectively. Currently, opportunities exist for these households to better utilise forages through expansion of forage plots and cattle activities, although assistance is required to support these households in addressing current constraints, particularly availability of water, if the sustainability of this feed technology for smallholder cattle household is to be established in Cambodia.  相似文献

Summary The effect of soil texture and CaCO3 content on water infiltration rate in crusted soil was studied with the use of a rain simulator. Two types of soils with low exchangeable sodium percentage (ESP < 3.0%) were studied: (i) calcareous soils (5.1–16.3% CaCO3) with a high silt-to-clay ratio (0.82–1.47) from a region with < 400 mm winter rain; and (ii) non-calcareous soils with a low silt-to-clay ratio (0.13–0.35) from a region with > 400 mm winter rain. Soil samples with clay percentages between 3 and 60 were collected in each region. Distilled water (simulating rainfall) and saline water (simulating irrigation water) were sprinkled on the soil. The soils were exposed to rain until steady state infiltration and corresponding crust formation were obtained. For both types of soils and for both types of applied water, soils with 20% clay were found to be the most sensitive to crust formation and have the lowest infiltration rate. With increasing percentage of clay, the soil structure was more stable and the formation of crust was diminished. In soils with lower clay content (< 20%), there was a limited amount of clay to disperse and, as a result, undeveloped crust was formed. Silt and CaCO3 had no effect on the final infiltration rate for either type of applied water, whereas with saline water, increasing the silt content increased the rate of crust formation.Contribution from the Agricultural Research Organization, The Volcani Center, Bet Dagan, Israel. In many countries, irrigation water is usedfor several purposes other than irrigatingfield crops. In Sri Lanka, irrigationwater from canals, wells, and reservoirs isused for domestic purposes, industry,livestock, and fisheries, and it alsocontributes to sustain the environment. However, policy makers and water managersin irrigation systems only take intoaccount water used for irrigating the fieldcrops and sometimes water used for domesticpurposes. Owing to the failure torecognize the different uses and users ofwater, the water in irrigation systems hasbeen undervalued. This paper presents theenvironmental impact of irrigation and aframework for valuing water for itsmultiple and often competing uses,especially focusing on water uses forecological services, in this case wetlands.The south coastal area of Sri Lanka wasselected as a site for a case study, including2,610 ha of irrigated area and adownstream wetland area of 2,250 ha whichhas five lagoons. This wetland areacombines coastal, marine and freshwaterecosystems in a tropical environment inwhich distinct plant and animal speciescoexist in a delicate balance. The existing standardised test systems for assessing the toxicity of crop protection products to the non-target arthropods Typhlodromus pyri (Acari: Phytoseiidae) and Aphidius rhopalosiphi (Hymenoptera: Aphidiidae) are limit tests designed to compare a single-use rate of the product with a water control. The suitability of these test systems for generating dose-response data as required for refined ecotoxicological risk assessment was evaluated. Data on dose-response toxicity of crop protection products to T. pyri and A. rhopalosiphi were generated under worst-case laboratory and to T. pyri under extended laboratory conditions and analysed using the standard Probit method, a logistic regression, a generalised Probit analysis, and the moving average-angle method in order to calculate the LR50-values (application rate killing 50% of the exposed organisms). The fit of the models, the precision of the resulting LR50 values, and the required minimum number of replicates were compared. In 85% of the studies, at least one of the statistical methods led to satisfactory results. The moving average-angle method was the most widely applicable method. The results show that the existing guidelines can be used to perform dose-response tests. Implications for risk assessment are discussed.  相似文献

Leptin may play a role in the endocrine-metabolic processes that guarantee the physiological course of lactation in dairy cattle. This study was aimed at determining the changes in plasma concentrations of leptin and some of the main hormones and metabolites involved in the lactogenetic process in high-yielding dairy cows throughout lactation; we also wanted to assess whether leptin secretion is subjected to seasonal influences. Blood samples were collected from 23 Italian Friesian dairy cows from the end of a lactation to the ninth month of the subsequent one; in addition, blood was sampled from 47 dairy cows in different phases of lactation during February and July. Plasma concentrations of leptin, growth hormone (GH), insulin, prolactin (PRL), glucose, non-esterified fatty acids (NEFA) and urea were quantified by either validated radioimmunoassay (RIA) or enzymatic colorimetric methods. At the beginning of lactation, GH concentrations significantly increased, while a significant reduction occurred in leptin and insulin. This endocrine condition, such as the significant increase in NEFA plasma concentrations, is indicative of a marked lipid mobilization. In the more advanced stages of lactation, when both energy and protein balances become positive, leptin plasma concentrations increased, whereas GH and NEFA concentrations declined. During the summer months, a significant increase in leptin plasma concentrations, irrespective of the phase of lactation, was observed. Collectively, our findings suggest that, in dairy cows, leptin may represent a 'metabolic signal' of animal's status of fattening and nutritional level; in addition, leptin seems to be influenced by photoperiod and environmental temperature.  相似文献
