Background: Lipase enzymes are of great importance in various industries. Currently, extensive efforts have been focused on exploring new lipase producer microorganism as well as genetic and protein engineering of available lipases to improve their functional features. Methods: For screening lipase-producing lactobacilli, isolated strains were inoculated onto tributyrin agar plates. Molecular identification of lipase-producing Lactobacilli was performed by sequencing the 16Sr DNA gene, and a phylogenetic tree was constructed. The LAF_RS05195 gene, encoding lipase protein in L. fermentum isolates, was identified using specific primers, amplified by PCR (918 bp) and cloned into the pET28a (+) vector. The recombinant proteins were expressed 2, 4, 6, and 12 hours after induction with IPTG and assessed using the SDS-PAGE. Enzymatic activity of the purified recombinant protein was measured at 410 nm in the presence of ρ-NPA and ρ-NPP. Results:Among five identified native lipase-producing isolates, one isolate showed 98% similarity with Enterococcus species. The other four isolates indicated 98% similarity to L. fermentum. After purification steps with Ni-NTA column, a single protein band of about 34 kDa was detected on SDS- PAGE gel. The enzymatic activity of purified recombinant protein alongside ρ-NPA and ρ-NPP was measured to be 0.6 U/ml and 0.2 U/ml, respectively. Conclusion:In the present research, a novel lipase/esterase from L. fermentum was cloned and expressed. The novel lipase/esterase has the merit to be further studied due to its substrate specificity. Drought and salt tolerances are complex traits and controlled by multiple genes, environmental factors and their interactions. Drought and salt stresses can result in more than 50% yield loss in Upland cotton (Gossypium hirsutum L.). G. barbadense L. (the source of Pima cotton) carries desirable traits such as tolerance to abiotic and biotic stress along with high fiber quality. However, few studies have been reported on mapping quantitative trait loci (QTL) for abiotic stress tolerance using a permanent bi-parental population in multiple tests. The transfer of drought and salt tolerance from Pima to Upland cotton has been a challenge due to interspecific hybrid breakdown. This issue may be overcome by using introgression lines with genes transferred from Pima to Upland cotton. In this study, four replicated tests were conducted in the greenhouse each for drought and salt tolerance along with another test conducted in a field for drought tolerance using an Upland recombinant inbred line population of TM-1/NM24016 that has a stable introgression from Pima cotton. The objectives of the study were to investigate the genetic basis of drought and salt tolerance and to identify genetic markers associated with the abiotic stress tolerance. A total of 1004 polymorphic DNA marker loci including RGA-AFLP, SSR and GBS-SNP markers were used to construct a genetic map spanning 2221.28 cM. This population together with its two parents was evaluated for morphological, physiological, yield and fiber quality traits. The results showed that drought under greenhouse and field conditions and salt stress in the greenhouse reduced cotton plant growth at the seedling stage, and decreased lint yield and fiber quality traits in the field. A total of 165 QTL for salt and drought tolerance were detected on most of the cotton chromosomes, each explaining 5.98–21.43% of the phenotypic variation. Among these, common QTL for salt and drought tolerance were detected under both the greenhouse and field conditions. This study represents the first study to report consistent abiotic stress tolerance QTL from multiple tests in the greenhouse and the field that will be useful to understand the genetic basis of drought and salt tolerance and to breeding for abiotic stress tolerance using molecular marker-assisted selection in cotton.  相似文献

Verticillium wilt (VW, caused by Verticillium dahliae Kleb) is a destructive fungal soil-borne disease in Upland cotton (Gossypium hirsutum L.). High levels of VW resistance can be transferred into Upland from Pima cotton (G. barbadense L.) through interspecific introgression breeding. In this greenhouse study, VW resistance was evaluated in a multi-parent advanced generation inter-cross (MAGIC) introgressed line (IL) population, derived from a random mated Barbadense Upland population with five generations of intermating (called RMBUP-C4) between three Upland cotton cultivars and 18 CS-B Upland lines each carrying a pair of G. barbadense chromosome or arm in the TM-1 background. The objectives of this study were to, (1) evaluate VW resistance of 530 MAGIC ILs in the greenhouse; and (2) to identify lines with VW resistance in the MAGIC population based on a total of three replicated greenhouse tests. Approximately 8 plants for each line in each replicate were grown and screened for VW resistance using three parameters i.e., disease leaf severity rating, percentage defoliated leaves, and percentage infected plants, with a total of ~?25,190 plants evaluated. A correlation analysis indicated that the three parameters were significantly and positively correlated with one another in each test. The disease leaf severity rating was the best parameter to assess VW resistance due to its relatively low coefficient of variation and its higher resolution to differentiate resistant genotypes from susceptible ones. Of the 530 genotypes, 5 showed resistance to VW, namely, NMIL348, NMIL518, NMIL405, NMIL290, NMIL307 and had higher levels of resistance to VW with mean disease leaf severity ratings, percentage of defoliated leaves, and percentage of infected plants across three tests ranging from 0.58–1.46, 9.46–26.74, and 25–95%, respectively. These lines can be used as parental lines to improve VW resistance in cotton breeding programs.  相似文献

Mesenchymal stem cells are undifferentiated cells that have the ability to divide continuously and tissue regeneration potential during the transplantation. Aging and loss of cell survival, is one of the main problems in cell therapy. Since the production of free radicals in the aging process is effective, the use of antioxidant compounds can help in scavenging free radicals and prevent the aging of cells. The aim of this study is evaluate the effects of L-carnitine (LC) on proliferation and aging of rat adipose tissue-derived mesenchymal stem cells (rADSC). rADSCs were isolated from inguinal region of 5 male Rattus rats. Oil red-O, alizarin red-S and toluidine blue staining were performed to evaluate the adipogenic, osteogenic and chondrogenic differentiation of rADSCs, respectively. Flow cytometric analysis was done for investigating the cell surface markers. The methyl thiazol tetrazolium (MTT) method was used to determine the cell proliferation of rADSCs following exposure to different concentrations of LC. rADSCs aging was evaluated by beta-galactosidase staining. The results showed significant proliferation of rADSCs 48 h after treatment with concentrations of 0.2 mM LC. In addition, in the presence of 0.2 mM LC, rADSCs appeared to be growing faster than control group and 0.2 mM LC supplementation could significantly decrease the population doubling time and aging of rADSCs. It seems that LC would be a good antioxidant to improve lifespan of rADSCs due to the decrease in aging.  相似文献

A comprehensive study on the colour reaction of Hashish applying the known reagents using some other aldehydes was carried out. Cinnamaldehyde reagent proved to be an excellent spray reagent for Hashish especially the diols.
Zusammenfassung Die Farbreaktionen der Haschisch-Komponenten wurden mit verschiedenen Reagenzien untersucht. Cinnamaldehyd stellte sich als ein sehr günstiges Haschish-Reagens insbesondere für Diole heraus.

Large quantities of brown algae have traditionally been exported from Chile as a raw material, of which Lessonia spp. has amounted to over 130 000 tons annually since 1995. To the export demand has recently been added the new demand for high‐quality Lessonia spp. as a foodstuff for the expanding abalone culture industry in Chile. The present study is based on efforts to produce significant quantities of Lessonia trabeculata in long‐line culture as food for tank‐cultured Haliotis rufescens Swainson and Haliotis discus‐hannae Ino, which accept it as an excellent source of nutrition. Small sporophytes of L. trabeculata were propagated in the laboratory from reproductive blades harvested by diving near Coquimbo (30°S). The best culture substrate was polyvinyl chloride (PVC) in small pieces, inserted into the nylon cord for final culturing. Enrichment of sea water with agricultural‐grade fertilizer produced no differences in growth and development of the Lessonia compared with results obtained using Provasoli medium. Sporophytes 1–2 cm in length cultured on 12‐mm cord were transferred to outdoor tanks with circulating sea water and strong aeration where they were outgrown to 15–20 cm length; at this size, they were transferred to a 50‐m long line in the ocean. In a 1‐year period, individual plants reached up to 1.7 kg in mass, with average values per cord of about 9 kg. Total production from the long line was about 500 kg fresh weight of the alga. In comparative testing, H. discus hannae grew as well on the cultured algae as on naturally occurring L. trabeculata.  相似文献

This work introduces, for the first time worldwide, undeinked recycled old newsprint as a new resource of electrical purposes paper. Impregnation of undeinked recycled old newsprint paper with linseed oil enhances the breaking length of paper and remarkably improves its electrical properties, i.e., the dielectric constant increases greatly and the a.c. conductivity decreases significantly due to impregnation. It was found that the electrical properties of the undeinked old newsprint paper and its linseed oil impregnated counterpart are close to the electrical properties of paper made from the more expensive virgin wood pulps and their linseed oil impregnated counterparts. Using the undeinked pulp is more privileged than using the deinked pulp; because eliminating the deinking step saves money, time, and reagents. In addition, eliminating the deinking step improved the breaking length of paper. Electron dispersive X-ray elemental analysis (EDX) was used to investigate the undeinked and deinked pulps for residual elements originating from the printing materials. EDX was correlated to the slight differences in electrical properties of paper made from undeinked and deinked pulps. However, impregnation was able to overcome these slight differences. It was shown that improvement in electrical properties, due to impregnation, is sustained at elevated temperatures.  相似文献

The cultivated tetraploid Gossypium barbadense L. cotton produces superior natural fibers for the textile industry in the world. However, the possibility in utilization of heterosis to further increase its lint yield has not been extensively explored. In this study, two commercial US Pima cotton cultivars and three exotic G. barbadense lines, together with all of their possible hybrids in F1 and F2 progeny without reciprocals, were tested for lint yield, yield components, and fiber quality traits in four environments in 2005–2007. With a few exceptions, genotype (G), environment (E), and G × E were all significant or highly significant for all the traits studied. General combining ability (GCA) variances for all the traits in both F1 and F2 were also significant, while specific combining ability (SCA) variances were detected only for lint yield, fiber length, and micronaire in both generations and boll weight in F1. GCA × E was also detected for lint percent, seed index, and fiber length in both F1 and F2, and boll weight in F1, but none of the traits had significant SCA × E. As a group, F1 and F2 out-yielded the parent group by 20–40% and 6–10%, respectively. Mid-parent heterosis (MPH) for lint yield in F1 was generally positive, ranging from ?4.7 to 116.4% with an average of 21.2–48.7%, while lint yield MPH in F2 ranged from ?23.3 to 69.4% with an average of 6.4–12.4%. However, useful heterosis in lint yield was only detected in the hybrid between the two US commercial cultivars Pima S-7 and DP 340. MPH for other traits was low or not detected. MPH in F2 was lower than that in F1 but they were generally positively correlated. The genetic distances (GD) of the parents (based on 467 polymorphic RAPD and AFLP markers) between the five parents was not consistently correlated with MPH and SCA of their hybrids and dominant effects for lint yield and other traits. However, significant and positive correlations between GD of parents and the performance of their hybrids were detected for lint yield, lint percentage, and lint index in both F1 and F2 in most of the tests. GD of parents was also correlated with their GCA and additive effects in lint yield, lint percent, lint index, micronaire, plant height, and elongation. The results suggest that the close correlation between GD and hybrid performance per se was mainly due to the existence of GCA and additive effects from parents.  相似文献

Abstract

Among major constraints to crop production, salt stress affects the morphological structure and yield in crop plants. Nano-fertilizers, have made their hierarchy to supplement the traditional fertilizers in mitigating the effect of abiotic stress due to their slow release and efficiency. Three products were tested namely lithovit®-standard(LITHO), monopotassium-phosphate(MKP:0-52-34), and aspirin(ASP) on salt-stressed tomato crop. These products were applied through foliar spraying (foliar) or fertigation (ferti) on tomato plants irrigated with 4 saline solutions (4, 6, 8 and 10dS/m). Control plants were subjected to five salinity levels (0, 4, 6, 8 and 10dS/m) without any treatments. Results showed that increasing in salinity levels adversely affected all parameters in the control plants. The ameliorating effect of MKP-ferti was prominent in plant height, stem diameter, number of fruits, fruit weight and yield/plant at 4 and 6S/m. Whereas, the effect of salinity was mitigated by LITHO-foliar regarding flowers number and total chlorophyll content at all salinity level. Leaf N, P and K contents were accumulated higher when MKP-ferti was used whereas, Ca2+ and Mg2+ contents were accumulated higher in the plants treated with LITHO-foliar. The cellular leakage of electrolyte was reduced at 4, 6 and 10dS/m with MKP-ferti and at 8dS/m with LITHO-foliar application. Among the two methods of application, foliar spraying of LITHO and ASP, and fertigation of MKP improved plant performance more than their complementary methods of application. The better performance of MKP necessitates the use of phosphorous and potassium-based nano-fertilizers. It can reduce the production cost of vegetables using conventional fertilizers.  相似文献
