U.S. state fish and wildlife agencies have responded to perceived ecological and social threats posed by mute swans by implementing population control strategies. In Michigan, some stakeholder groups have vocally opposed particular control activities. To better understand and characterize key aspects of the underlying media dialogue about mute swan management in Michigan, one factor in the development of public perceptions about wildlife issues, we conducted a qualitative content analysis of Michigan-specific online news articles. Results revealed tensions between stakeholder groups about management priorities were rooted in differing ideas about healthy social and ecological systems, appropriate wildlife behavior, and the acceptability of lethal control (i.e., shooting living birds). Characterizing which stakeholder groups aligned with particular arguments might allow managers to tailor and direct messaging to specific audiences.  相似文献

Post-recovery wolf management remains controversial. In Michigan, dialogue centers on hunting wolves but controversy may be more nuanced than simple pro- or anti-hunting positions. Social identity may cause stakeholders to organize in groups and identity differences may be driving controversy. To explore stakeholder disagreement over wolf management, we conducted semi-structured key informant interviews (n = 21) about identity and stewardship in August–September 2012. Four overarching identity themes emerged regarding: (a) management objectives, (b) focal levels, (c) control methods, and (d) justifications for methods. Themes indicated two main researcher-defined identity groups. Interviewees from both groups identified six common stewardship themes: (a) bequest values, (b) ecosystem health, (c) education, (d) existence values, (e) pride in natural resources, and (f) sustainability. Findings suggest stakeholders may be conflicting over opposing identities vis-à-vis organization affiliation, which may be related to what management strategies individuals oppose. Establishing common stewardship objectives through established participation methods may help alleviate controversy.  相似文献

DNA translocases are molecular motors that move rapidly along DNA using adenosine triphosphate as the source of energy. We directly observed the movement of purified FtsK, an Escherichia coli translocase, on single DNA molecules. The protein moves at 5 kilobases per second and against forces up to 60 piconewtons, and locally reverses direction without dissociation. On three natural substrates, independent of its initial binding position, FtsK efficiently translocates over long distances to the terminal region of the E. coli chromosome, as it does in vivo. Our results imply that FtsK is a bidirectional motor that changes direction in response to short, asymmetric directing DNA sequences.  相似文献

A total of 404 isolates of Xanthomonas campestris pv. vesicatoria , obtained from Capsicum chinense cv. West Indian Red grown in Barbados and Grenada, were differentiated into pathogenic races, and of these, 96 were tested also for selected taxonomic group phenotypes. The response of C. chinense to infection by several X. campestris pv. vesicatoria races and the contribution of races isolated from this cultivar to severity of bacterial spot of bell pepper and tomato were also investigated. P4T2, P5T2 and P6T2 were the predominant races of X. campestris pv. vesicatoria isolated from C. chinense grown in Grenada, whereas nine races (T1, P4, P6, P0T2, P1T2, P4T1, P4T2, P6T1 and P6T2) were isolated in Barbados. Race P4T2 comprised 46.0 and 71.4% of the isolates from Barbados and Grenada, respectively. The 96 isolates, all of which overcame resistance conferred by the gene Bs2 , shared taxonomic group B strain characteristics, including the presence of the β-protein band, positive amylolytic activity and inability to oxidize cis -aconitate. The C. chinense cv. West Indian Red was susceptible only to races of X. campestris pv. vesicatoria that can overcome Bs2 gene resistance. Of six such races identified in Barbados, only P4T1, P4T2 and P6T1 affected bacterial spot-susceptible bell pepper or tomato in the field, and they amounted to only 1.5–2.1% of each sample of isolates from these plant species. Moreover, they were confined to the smallest bacterial spot lesions. Bell pepper was most severely affected by combinations of races T1 with P3T2 and T2 with P0T1, and tomato by race T1 only and combinations of races P0T1 with P0T2 and P1T1 with P1T0, all of which prevailed in the field despite selection against them by C. chinense cv. In several previous observations on peat soils, redox potentials appeared to vary with season and depth. To clarify this variation, a systematic study was made over 1 yr at four peat sites, including wet and dry areas at high and low altitude in northern England.The coefficients of linear regressions of redox potential on seasonal temperatures became progressively more negative with increasing depth at all four sites. These observations support an hypothesis that increased aeration accompanying increased temperature played a role at the surface, but was of less importance in the deeper layers, where the reducing conditions that result from microbial activity were dominant.By using orthogonal polynomials, it was possible to demonstrate the existence of redox minima in the profiles at all sites. There were rather more minima present using this method at the lowland sites that at the upland ones where trends of redox with depth were otherwise linear or non-significant. Although the mean redox potentials over whole profiles were lower in the wetter sites than the drier, the mean depths of the redox minima were similar. These mean depths were poorly defined however having standard deviations of 30–50 per cent of the means calculated from the year's observations.Finally, it was shown that although the potentials were measured 60 s after closing the circuit, very similar conclusions would have been reached if the readings had been taken after 10 s.  相似文献

Incidence and control of subclinical mastitis in intensively managed ewes   总被引:1,自引:0,他引:1  
Ninety-three intensively managed, mature ewes were followed through three lactations to study effects and interrelationships of postweaning fast, prophylactic antibiotic injection, number of lactations and breed on subclinical mastitis, milk production and milk composition. Animals used in a 2 x 2 factorial design were allotted to treatment combinations of 1) postweaning 72 h fast or no fast, and 2) 1 x 10(6) units of procaine penicillin or sham i.m. injection at weaning. Treatments were implemented at weaning in yr 1; each ewe received the same treatment in each of the subsequent 2 yr of the study. Breeds included Finnish Landrace (F), Dorset (D), Rambouillet (R), Finnish Landrace x Dorset (FD) and Barbados Blackbelly x Dorset (BD). Milk production was greatest for D, followed by BD, FD, R and F during yr 1 (P less than .01) and overall (P less than .05). Milk production data were not collected in yr 2, but similar results were obtained in yr 3, except that the R and F exchanged rank positions. Milk composition was affected by breed. Breeds differed (P less than .05) in percentage lactose, fat and total nonfat solids in yr 1 and differed (P less than .01) in percentage lactose, fat, solids and protein across the 2 yr. A breed difference (P less than .05) was observed for incidence of mastitis at 3 wk postpartum and at weaning. Milk production was negatively correlated with incidence of mastitis (r = -.22, P less than .05). Enrofloxacin pharmacokinetics were studied in European cuttlefish, Sepia officinalis, after a single 5 mg/kg i.v. injection or a 2.5 mg/L 5 h bath. A pilot study with two animals was also performed following a 10 mg/kg p.o. administration. The concentration of enrofloxacin in hemolymph was assayed using high-performance liquid chromatography (HPLC) and pharmacokinetic parameters were derived from compartmental methods. In the i.v. study, the terminal half-life (t(1/2)), apparent volume of distribution, and systemic clearance were respectively 1.81 h, 385 mL/kg, and 4.71 mL/min/kg. Following bath administration the t(1/2), peak hemolymph concentration (C(max)), and area under the curve to infinity (AUC(0-infinity)) were 1.01 h, 0.5 +/- 0.12 mug/mL, and 0.98 microg.h/mL, respectively. After oral administration, the t(1/2), C(max), and AUC(0-infinity) were 1.01 h, 10.95 microg/mL, 26.71 mug.h/mL, respectively. The active metabolite of enrofloxacin, ciprofloxacin, was not detected in any samples tested. The hemolymph concentration was still above minimum inhibitory concentration (MIC) values for shrimp and fish bacterial isolates at 6 h after i.v. administration, therefore, a dose of 5 mg/kg i.v. every 8-12 h is suggested for additional studies of efficacy. The C(max) value for the water bath was lower than for the i.v. study, but a bath of 2.5 mg/L for 5 h once to twice daily is suggested for additional studies to test efficacy against highly susceptible organisms. Although only two animals were used for the oral study, a dose of 10 mg/kg produced hemolymph concentrations of enrofloxacin that were in a range consistent with therapeutic efficacy in other species.  相似文献
