ABSTRACT Five areas in urban Miami were identified to study the spread of Xanthomonas axonopodis pv. citri to determine if the practice of removing exposed citrus trees within 38.1 m of trees affected by citrus canker was adequate to curtail further bacterial spread. To accomplish this, 18,769 trees in dooryards were surveyed, geo-referenced by differential global positioning systems (GPS), and assayed for disease severity, age of infection, citrus cultivar, location of infection in tree, and canopy size. For each tree, the date the tree became infected was estimated and used to separate trees into contiguous 30-day categories. For each area studied, distance measurements between focal trees and newly infected trees were calculated for various temporal periods of 30, 60, 90, and 120 days in duration, corresponding to intervals of inspection survey. A visual basic application was used to calculate the distances between each newly diseased tree and all prior focal trees. The nearest distance was used because it was considered the most conservative estimate possible. It is therefore likely to be an underestimate of spread but is a good estimate of the minimum possible distances of spread. For the first four 30-day periods among the five study areas, calculated maximum distances of spread ranged from 12 to 3,474 m, indicating a broad continuum of distance for bacterial spread was possible. Disease increased during the first two-thirds of the time studied and reached an asymptote due to dry conditions in the final one-third of the duration of the study. Cross correlation analysis indicated that disease was best visualized 107 days following rainstorms with wind. Analysis of regional spatial point patterns was performed temporally for each 30-day period via a modified Ripley's K-function. Spatiotemporal analyses between periods over areas larger than previously examined were accomplished via spatiotemporal semivariogram analysis. These methods in combination demonstrated rapid increases in range of spatial dependency and range of spatiotemporal dependency for all study sites. This corresponded to rapid spread of disease across the regions studied in response to rainstorms with wind followed by a "filling in" of disease on remaining noninfected susceptible trees through time by less intense rain events. A stochastic quadratization technique demonstrated that disease incidence and disease severity were not greatly affected by urban host density but were positively correlated to host susceptibility within local 0.25-km(2) quadrats.  相似文献

ABSTRACT Citrus tristeza virus (CTV) was monitored for 4 years by monoclonal antibody probes via enzyme-linked immunosorbent assay in four citrus orchards in northern Costa Rica and four orchards in the Dominican Republic following the introduction of the brown citrus aphid, Toxoptera citricida. The Gompertz nonlinear model was selected as the most appropriate in most cases to describe temporal increase of CTV. Ordinary runs analysis for association of CTV-positive trees failed to show a spatial relationship of virus status among immediately adjacent trees within or across rows. The beta-binomial index of dispersion for various quadrat sizes suggested aggregations of CTV-positive trees for all plots within the quadrat sizes tested. Spatial autocorrelation analysis of proximity patterns suggested that aggregation often existed among quadrats of various sizes up to four lag distances; however, significant lag positions discontinuous from the main proximity pattern were rare. Some asymmetry was also detected for some spatial autocorrelation proximity patterns. These results were interpreted to mean that, although CTV-positive trees did not often influence immediately adjacent trees, virus transmission was common within a local area of influence that extended two to eight trees in all directions. Where asymmetry was indicated, this area of influence was somewhat elliptical. The spatial and temporal analyses gave some insight into possible underlying processes of CTV spread in the presence of T. citricida and suggested CTV spread was predominantly to trees within a local area. Patterns of longer-distance spread were not detected within the confines of the plot sizes tested. Longer-distance spread probably exists, but may well be of a complexity beyond the detection ability of the spatial analysis methods employed, or perhaps is on a scale larger than the dimensions of the plots studied.  相似文献

An electroimmobilisation device has been developed to facilitate the automated shearing of sheep, but there is little information on its effects on the body. We have studied its effects on the cardiovascular system and on intermediary metabolism in sheep. Electroimmobilisation caused statistically significant increases in mean arterial pressure, heart rate, cardiac output, renal and hepatic and hindquarter glucose and lactate flux, organ and whole body oxygen flux, hindquarter blood flow and core temperature and decreases in arterial and posterior vena cava blood pH, renal and hepatic blood flow and PaCO2. Notably, no change occurred in PaO2. The metabolic changes demonstrated the capacity of sheep to respond to the increased muscular and cardiovascular work induced by electroimmobilisation. Pulmonary function was not compromised during electroimmobilisation as judged from blood gas changes, and the acid/base changes were rapidly reversed after electroimmobilisation. The recovery to control conditions for all perturbations generally took no longer than 30 min, consistent with a rapid and physiologically adequate reversal by the animal's homeostatic mechanisms.  相似文献

Early diagnoses are applied in selection so as to shorten breeding times. Somaclonal variations may occur relatively often on in vitro propagation; these need to be eliminated. ABSTRACT We evaluated the impact of roguing on the spread and persistence of the aggressive Plum pox virus strain M (PPV-M) in 19 peach orchard blocks in Southern France. During a 7- to 10-year period, orchards were visually inspected for PPV symptoms, and symptomatic trees were removed every year. Disease incidence was low in all orchards at disease discovery and was <1% in 16 of the 19 orchard blocks. The spread of Sharka disease was limited in all 19 blocks, with an annual disease incidence between 2 and 6%. However, new symptomatic trees were continuously detected, even after 7 to 10 years of uninterrupted control measures. An extended Cox model was developed to evaluate to what extent tree location, orchard characteristics, environment, and disease status within the vicinity influenced the risk of infection through time. Eleven variables with potential effect on tree survival (i.e., maintenance of a tree in a disease- free status through time) were selected from survey data and databases created using a geographical information system. Area of the orchard, density of planting, distance of a tree from the edge of the orchard block sharing a boundary with another diseased orchard, and distance to the nearest previously detected symptomatic tree had a significant effect on the risk for a tree to become infected through time. The combined results of this study suggest that new PPV-M infections within orchards subjected to roguing resulted from exogenous sources of inoculum, disease development of latent infected trees, as well as infected trees overlooked within the orchards during visual surveys. A revision of the survey and the roguing procedures used for more effective removal of potential sources of inoculum within the orchards and in the vicinity of the orchards would improve disease control suppression of PPV.  相似文献

Abstract

CASE HISTORY: A 15-mm diameter mass developed in the S/C tissue overlying the right lateral gluteal muscle of a 6½-year-old female Flat-coated Retriever.

PATHOLOGICAL FINDINGS: Cytological preparations following aspiration of the mass were highly cellular and consisted of a population of large polygonal cells containing single to multiple nuclei, large prominent nucleoli, and intracytoplasmic vacuoles. Histologically, the neoplasm consisted of similar large cells surrounded by thick fibrous connective tissue trabeculae. The large polygonal cells reacted positively with antibodies against vimentin, low- and high-molecular-weight variants of cytokeratin (AE1/AE3), but not with antibodies to desmin or glial fibrillary acidic protein (GFAP).

DIAGNOSIS: The clinical, gross, histological and immunohistochemical findings are similar to those reported for parachordomas in humans. Neither recurrence nor metastases were noted 18 months after surgical excision of the mass.

CLINICAL RELEVANCE: This is the first reported case of a possible parachordoma in a dog, a benign tumour with cytological features of malignancy.  相似文献

ABSTRACT An environmental chamber was designed to study aerial release of spores of ascomycetes and hyphomycetes, based on a device developed by C. M. Leach. Relative humidity (RH), temperature, red (660 nm) and infrared (880 nm) light, leaf wetness, wind speed, vibration, and rain events are controlled and monitored within the chamber via an RTC-HC11 real-time controller and data-acquisition system. A BASIC11 computer program is uploaded to and controls the system. The program requests values for environmental parameters that change through time according to user specifications. The controller interacts with a stepper motor, solenoids, and relay switches via a feedback system based on data received from solid-state RH, temperature, and leaf-wetness sensors. The data-acquisition system records environmental data from the chamber in RAM (random access memory) that can be downloaded to a personal computer for correlation with spore-release data. Spores released from fungal specimens on plant tissues and cultures placed in the chamber and subjected to the desired environmental conditions are collected on a continuous volumetric spore trap at an exhaust port from the chamber. The performance of the device was examined by measuring spore release of Mycosphaerella citri, Alternaria solani, and Venturia inaequalis under various environmental conditions.  相似文献

The objective of this study was to investigate in mares the effect of parturition on plasma and milk levels of retinol, beta-carotene, alpha-tocopherol and cholesterol over 12 weeks around parturition. In blood plasma of horses around parturition an increase of all these components was observed. This increase was most impressive in beta-carotene (P<0.05) and less pronounced for vitamin E, vitamin A and cholesterol. The magnitude of increase around parturition corresponded well with the magnitude of accumulation in colostrum; levels of beta-carotene in colostrum were 65 times higher compared to mature milk while vitamin A, vitamin E and cholesterol were only 3 to 8 times higher. Beta-carotene concentrations in colostrum were positively correlated with corresponding plasma levels (r = 0.9; P<0.001). Reasons for the increase in plasma beta-carotene around parturition may include an improved absorption of carotene and/or reduced conversion into vitamin A as well as mobilisation from tissue storages or a reduced uptake in tissues other than the mammary gland. In conclusion, the results may point to possible component- and species-specific differences involved in the transfer of fat-soluble vitamins, beta-carotene and cholesterol from blood plasma into colostrum.  相似文献

Citrus canker (Xanthomonas citri subsp. citri, Xcc) is one of the most serious diseases citrus in Florida, and elsewhere in the world. The disease causes yield loss and some fresh fruit trade restrictions may apply. Cultural management techniques such as windbreaks may work by not only reducing wind speed, but also reducing the period of exposure of susceptible foliage or fruit to those wind speeds that support infection from incoming inoculum. To investigate the effect of exposure period to inoculum of Xcc, seedlings of canker-susceptible Swingle citrumelo were exposed to sprayed inoculum for increasing periods at different wind speeds. The incidence and severity of citrus canker was assessed. In three experiments the incidence and severity of citrus canker most often increased with longer periods of exposure to inoculum, especially so at wind speeds of ≥16 m/s compared to wind speeds of ≤5 m/s (wind speed also increased disease incidence and severity). Regression analysis demonstrated relationships between period of exposure to inoculum and the percent infected leaves per plant, the number of lesions per plant, the number of lesions per infected leaf, and for the percent of infected leaves with lesions on the petioles at wind speeds of ≥16 m/s (R2?=?0.16–0.72). Due to the effect of inoculum exposure period and wind speed, attempts should be made to minimize exposure of canker-susceptible citrus when wind speed is highest and inoculum is available. Windbreaks should help minimize periods of exposure to splashed inoculum in high winds.  相似文献
