The effect of take-all root lesions on nitrate uptake of wheat was investigated in two experiments under controlled conditions. Plants were supplied with a nutrient solution labelled with 15N during stem elongation and flowering to assess the distribution of the isotopic tracer in the different plant organs, and particularly in root segments located on both sides of take-all lesions. The 15N atom percentage excess measured in root segments located below lesions longer than 1 cm was reduced on average by half compared with that in healthy roots and root segments above lesions, reflecting a reduction in nitrogen uptake by these root segments. This reduction probably resulted from the invasion and breakdown of phloem vessels by the fungus hyphae, interrupting energy supply and thus the uptake process. Severely infected plants showed an increase in the uptake rate per unit of efficient root, which appeared to be a compensatory response to reduction of efficient root biomass in order to satisfy shoot nitrogen demand. However, this compensatory response was insufficient to ensure nitrogen accumulation equivalent to that of healthy plants, as reductions in nitrogen accumulated in roots and aerial parts at flowering were up to 56 and 49%, respectively, for plants with more than 50% of the root system below lesions longer than 1 cm.  相似文献

A soybean lecithin‐based extender supplemented with hyaluronic acid (HA) was assayed for effectiveness to improve the quality of frozen–thawed ram semen. HA has not been tested yet in an extender containing soybean lecithin for freezing ram semen. Thus, the aim of this study was to analyse the effects of soybean lecithin at 1% or 1.5% along with HA at 0, 0.5 and 1 mg ml‐1 in a Tris‐based extender on the motion characteristics, membrane integrity (HOST), viability, GSH peroxidase (GSH‐PX) activity, lipid peroxidation and acrosomal status after freezing–thawing. Semen was collected from four Mehraban rams during the breeding season and frozen in the six lecithin×HA extenders. The extender containing 1.5% lecithin supplemented with no HA yielded higher total motility (52.5%±1.6), viability (55.8%±1.6) and membrane integrity (44.5%±1.7), but the effects of the lecithin concentration did not reach signification. Linearity‐related parameters, ALH, BCF, lipid peroxidation, GSH‐PX activity, morphology and acrosomal status were not affected by the extender composition. In general, adding HA significantly decreased sperm velocity (1 mg ml‐1 HA), total motility (only with 1.5% lecithin), viability (1 mg ml‐1 HA for 1% lecithin; both concentrations for 1.5% lecithin) and membrane integrity. In conclusion, adding HA to the freezing extender supplemented with soybean lecithin failed to improve quality‐related variables in ram semen. Increasing the lecithin content could have a positive effect, but further studies are needed.  相似文献

Recently, isolation and in vitro culture of putative spermatogonial stem cells (SSCs) in the domestic cat have been conducted. However, the cellular niche conditions that facilitate the establishment and long‐term maintenance of feline SSCs (FSSCs) have not been described. Therefore, we investigated the type of feeder cells used to stimulate colony formation and growth of FSSCs among the various factors in the FSSC niche. Spermatogonial stem cells isolated from feline testes were cultured on mitotically inactivated testicular stromal cells (TSCs) derived from cats, dogs and mice, and mouse embryonic fibroblasts (MEFs). The formation and growth of colonies derived from SSCs cultured on each type of feeder cell were identified at passage 0, and the morphology, alkaline phosphatase (AP) activity and expression of SSC‐specific genes in surviving colonies were investigated at passage 4. Among these diverse feeder cells, TSCs from cat showed the greatest colony formation, growth and maintenance of FSSCs, and SSC colonies cultured by passage 4 showed a typical dome‐shaped morphology, AP activity and expression of SSC‐specific genes (NANOG, OCT4, SOX2 and CD9). Accordingly, these results demonstrate that feline TSCs could be used as feeder cells to support the establishment and maintenance of SSCs from domestic cats.  相似文献

The coalescence of a neutron star and a black hole in a binary system is believed to form a torus around a Kerr black hole. A similarly shaped magnetosphere then results from the remnant magnetic field of the neutron star. In the strong-field case, it contains a cavity for plasma waves located between the barrier of the gravitational potential and the surrounding torus. This cavity may be unstable to superradiance of electromagnetic waves. Superradiant amplification of such waves, initially excited by turbulence in the torus, should inflate into a bubble in a time as short as approximately 0.75 (1 percent/&cjs3539;epsilon&cjs3539;2)(M/7M middle dot in circle) seconds approximately 0.15 to 1.5 seconds, assuming an efficiency &cjs3539;epsilon&cjs3539;2 = 0.5 to 5 percent and a mass M = 7M middle dot in circle. These bubbles may burst and repeat, of possible relevance to intermittency in cosmological gamma-ray bursts. The model predicts gamma-ray bursts to be anticorrelated with their gravitational wave emissions.  相似文献

The critical crop nitrogen uptake is defined as the minimum nitrogen uptake necessary to achieve maximum biomass accumulation (W). Across a range of crops, the critical N uptake is related to W by a power function with a coefficient less than unity that suggests crop N uptake is co-regulated by both soil N supply and biomass accumulation. However, crop N demand is also often linearly related to the expansion of the leaf area index (LAI) during the vegetative growth period. This suggests that crop N demand could be also linked with LAI extension. In this paper, we develop theory to combine these two concepts within a common framework. The aim of this paper is to determine whether generic relationships between N uptake, biomass accumulation, and LAI expansion could be identified that would be robust across both species and environment types. To that end, we used the framework to analyze data on a range of species, including C3 and C4 ones and mono- and di-cotyledonous crops. All crops were grown in either temperate or tropical and subtropical environments without limitations on N supply. The relationship between N uptake and biomass was more robust, across environment types, than the relationship of LAI with biomass. In general, C3 species had a higher N uptake per unit biomass than C4 species, whereas dicotyledonous species tended to have higher LAI per unit biomass than monocotyledonous ones. Species differences in N uptake per unit biomass were partly associated with differences in LAI and N-partitioning. Consequently the critical leaf-N uptake per unit LAI (specific leaf nitrogen, SLN) was relatively constant across species at 1.8–2.0 g m−2, a value that was close to published data on the critical SLN of new leaves at the top of the canopy. Our results indicate that critical N uptake curves as a function of biomass accumulation may provide a robust platform for simulating N uptake of a species. However, if crop simulation models are to capture the genotypic and environmental control of crop N dynamics in a physiologically functional manner, plant growth has to be considered as the sum of a metabolic (e.g. leaves) and a structural (e.g. stems) compartment, each with its own demand for metabolic and structural N.  相似文献

Several disease indices (DI) have been proposed to assess the severity of phoma stem canker at late growth stages. The DI commonly used in France (called G2 DI) consists of integrating the distribution of the proportions of plants observed within six canker severity classes into a single index. This index uses arbitrarily chosen coefficients associated to each class of canker severity. A given value of this DI can therefore be obtained with different distributions among the canker severity classes. Consequently, it is important to assess to what extent the G2 DI is representative of the distribution of plants in canker severity classes. We show in this study that the G2 DI is a good indicator of the distribution of the observed canker severities, using a dataset from different years, sites and cultivars; this illustrates the robustness of the G2 DI.  相似文献

Dynamic crop models can be used to predict the occurrence of nitrogen deficiency during crop growth and optimize nitrogen fertilisation. However, prediction errors can be large and may lead to wrong recommendations. The objective of our work is to study the value of correcting the dynamic model Azodyn using transmittance measurements made with the N-Tester® (Yara) to predict the nitrogen status of a winter wheat crop. Our approach is to use a Bayesian method called the “interacting particle filter” to fit the model's state variables to measurements obtained over the course of the season. This approach was assessed on 44 experimental plots. Predictions of crop biomass, nitrogen uptake and nitrogen nutrition index were first performed for each plot by using the model without any correction. A second series of predictions was then performed for the same variables by correcting the model with N-Tester measurements at GS 7 on Feekes’ scale. The results showed that the second series of predictions were more accurate. Depending on the prediction dates, model corrections reduced the root mean squared error by 18.1–53.2% for nitrogen nutrition index, by 9.1–10.1% for biomass, and by 17.1–45.0% for nitrogen uptake. The predictions were improved up to 52 days after the measurement but the degree of improvement was higher when the prediction date was close to the measurement date. The results also showed that, when corrected, model predictions were very sensitive to values of N-Tester measurements. It is therefore necessary to use N-Tester measurements which are as precise as possible.  相似文献

The relationship between severity of blackleg, or phoma stem canker ( Leptosphaeria maculans/L. biglobosa ), and subsequent primary inoculum production on oilseed rape ( Brassica napus ) stubble was investigated at two sites in France over 3 years. The quantity of primary inoculum produced in the following year increased with canker severity, from 1·9 to 10·8 pseudothecia cm−2 on stubble with the least and most severe cankers, respectively. Stubble incubated at Le Rheu (cooler, more rain) had 1·7 times more pseudothecia than stubble incubated at Grignon. Stubble collected at Grignon had 2·7 times more pseudothecia than that collected at Le Rheu. The use of Darmor, a cultivar with a good level of quantitative resistance, reduced the severity of canker in the field, but not the subsequent inoculum production for stubble of the same canker severity class. At both sites, maturation of pseudothecia occurred after 63–75 days of incubation and increased with canker severity with a mean of 0·5 and 3% mature pseudothecia appearing per favourable day, on stubble with the least and most severe cankers, respectively. A simplified procedure for pseudothecial quantification proved satisfactory: for all three observers, most (91–96%) of the fructifications counted as pseudothecia were real pseudothecia. Only a few (4–14%) of the fructifications considered as non-pseudothecia were in fact pseudothecia of L. maculans . The total area occupied by pseudothecia, which was simpler and faster to evaluate, was correlated (coefficient of determination, R 2 = 71%) with the number of counted pseudothecia. The results presented here make it possible to forecast the quantity of available primary inoculum for a given disease severity.  相似文献

To optimize wheat segregation for the various markets, it is necessary to add to genotype segregation, a prediction before harvest of the values of yield and grain protein concentration (GPC) for the different fields of the collecting area. Different tools allowing a prediction of crop production exist. Among them, the evaluation of nitrogen concentration by a chlorophyll meter (Soil–Plant Analysis Development (SPAD) readings), classically used to adapt the nitrogen fertilizer application, has been used in few works to foresee grain yield and grain protein concentration. But the relationships between N crop status and SPAD measurements varies among varieties and this genotypic effect has rarely been incorporated in models of forecasting grain quality.

This paper compares several models to forecast yield, nitrogen uptake in grain (NUG) and grain protein concentration from trials carried out in 2001 and 2002 at the INRA experiment station of Grignon (West of Paris). Trials crossed nine varieties by four (2002) or five (2001) nitrogen rates. Input variables of those models are mainly chlorophyll meter measurements (SPAD) on the penultimate leaf at GS65 and on the flag leaf at GS71 Zadoks growth stages and ear number per square meter (NE).

A square root model of yield based on NE × SPAD gave the best fit (RMSE = 0.6 t ha−1 for both stages) if considering three different groups of genotypes. Based on the same variable, NE × SPAD, a quadratic model for NUG without significant effect of genotypes gave the best fit (RMSE, between 21 and 30 kg ha−1 depending of the growth stage). And, for GPC, considering the same three groups of genotypes, the slope of the linear model with the ratio of predicted grain nitrogen concentration to predicted yield, is the same at both stages and very close to the standard value used to calculate protein concentration from nitrogen concentration (5.7), but the predictive quality of the model is more than 10% higher at GS71 (R2 of 0.77) than at flowering (R2 of 0.64). Finally, the sensibility of the models to delay in the stage of measurement is discussed.  相似文献
