Myxomatosis is a major viral disease of the European rabbit (Oryctolagus cuniculus). Two forms of the disease (nodular and amyxomatous) exist. The clinical diagnosis of the nodular form is easily performed on the basis of typical skin lesions whereas that of amyxomatous forms must be based on virus isolation or detection of specific antibodies to myxoma virus (MV). The seroprevalence of MV was studied between March 1998 and February 1999 in 16 farms from three European countries considered free of myxomatosis on the basis of the absence of typical clinical signs. MV antibodies were detected by enzyme-linked immunosorbent assay (ELISA) (sensitivity 100%, specificity 90%) in all 16 farms; the seroprevalences corrected for test inaccuracy (95% confidence interval) were 55+/-7.7% and 37+/-6.1% for does and broilers, respectively. The association between herd sizes, types of rabbitries, and presence of recurrent respiratory or digestive troubles and seroprevalence of MV antibodies was tested in logistic multiple regressions. In all models, the seroprevalence of MV antibodies was significantly higher in herds (does and broilers) with recurrent respiratory or digestive troubles than in herds without these problems. The seroprevalence was also higher in herds (does and broilers) where animals were housed totally or partially in outdoors rabbitries than in totally enclosed rabbitries. The effect of herd sizes on the presence of MV antibodies was the same in does and broilers; intermediate sizes were at lower risk than the smaller and larger ones.  相似文献

From a total of 261 yellow sugarbeet leaves collected from 10 countries representing three continents, the incidence and distribution of strains of Beet mild yellowing virus (BMYV), Beet chlorosis virus (BChV) and Beet yellows virus (BYV) were analysed using serological and molecular methods. BMYV was found in all countries except Greece, and more frequently in the northern and western areas of Europe, whereas BYV predominated in Turkey, Spain, Greece, the USA and Chile. BChV, originally found in the USA and the UK in 1989, was identified in France, Spain, the Netherlands and Chile. Nine sugar beet poleroviruses, plus a reference isolate of Turnip yellows virus (TuYV, syn. Beet western yellows virus ), were further characterized and compared. Isolates obtained from sugar beet infected this species, but not oilseed rape or lettuce; all isolates except one infected Capsella bursa-pastoris . The coat-protein sequences of these isolates were highly similar, with the consensus sequence representing 89% of nucleotide residues. Within the coat-protein gene, two regions were identified that could represent specific epitopes to which monoclonal antibody BYDV-PAV-IL-1 could bind; this antibody is used to distinguish beet poleroviruses in ELISA. Comparison of the sequences at the 5' end showed that sequence homology existed only between isolates with the same host range. The first sequence data of polerovirus isolates from Chile are presented, showing that the coat protein and the 5' end of their genomes are highly similar to those of BMYV isolates found in Europe. Chilean polerovirus isolates may have been imported from the northern hemisphere in sugar beet breeding material.  相似文献

This study was conducted to determine whether young calves with maternal antibodies against bovine herpesvirus type 1 (BHV-1) but without antibodies against glycoprotein E (gE) can produce an active antibody response to gE after a BHV-1 infection. Five calves received at birth colostrum from gE-seronegative cows which had been vaccinated two or three times with an inactivated BHV-1, gE-deleted marker vaccine. After inoculation with a wild-type virulent strain of BHV-1, all the passively immunised gE-negative calves shed virus in large amounts in their nasal secretions. All the calves seroconverted to gE within two to four weeks after inoculation and then had high levels of gE antibodies for at least four months. The development of an active cell-mediated immune response was also detected by in vitro BHV-1-specific interferon-gamma assays. All the calves were latently infected, because one of them re-excreted the virus spontaneously and the other four did so after being treated with dexamethasone. The results showed that under the conditions of this work the gE-negative marker could also distinguish between passively immunised and latently infected calves.  相似文献

During a field trial to evaluate the efficacy of repeated vaccinations with bovine herpesvirus type 1 (BHV-1) marker vaccines, a glycoprotein E (gE)-negative BHV-1 strain was isolated from the nasal secretions of two cows, eight months after vaccination with a gE-negative live-attenuated vaccine, initially given intranasally, then intramuscularly. The strain isolated was characterised using immunofluorescence, restriction analysis and PCR. All the techniques used identified the isolated virus as a gE-negative BHV-1 phenotypically and genotypically identical to the Za strain used as a control.  相似文献

Yellowing diseases of sugar beet can be caused by a range of strains classified as Beet mild yellowing virus (BMYV) or Beet western yellows virus (BWYV), both belonging to the genus Polerovirus of the family Luteoviridae. Host range, genomic, and serological studies have shown that isolates of these viruses can be grouped into three distinct species. Within these species, the coat protein amino acid sequences are highly conserved (more than 90% homology), whereas the P0 sequences (open reading frame, ORF 0) are variable (about 30% homology). Based on these results, we propose a new classification of BMYV and BWYV into three distinct species. Two of these species are presented for the first time and are not yet recognized by the International Committee on Taxonomy of Viruses. The first species, BMYV, infects sugar beet and Capsella bursa-pastoris. The second species, Brassica yellowing virus, does not infect beet, but infects a large number of plants belonging to the genus Brassica within the family Brassicaceae. The third species, Beet chlorosis virus, infects beet and Chenopodium capitatum, but not Capsella bursa-pastoris.  相似文献

The characterization of stem elongation is of fundamental importance in C4 tropical grasses as it affects forage quality and determines optimal management practices. The objectives of this study were to analyse the determinants of stem elongation and leaf area production in shoots of Pennisetum purpureum cv. Napier (elephant grass) using unstressed isolated plants. Three experiments were conducted in Brazil during the spring, summer and autumn seasons. Regular measurements of leaf and pseudostem length were performed on the main and primary axes. Ten destructive measurements were also performed during each experiment to monitor apical meristem height, internode length and the number of initiated leaves. The onset of stem elongation occurred at the same vegetative stage (i.e., appearance of leaf 13) irrespective of the seasons and experiments. The first internode to elongate belonged to phytomer 8, and a constant lag of five phyllochrons was systematically observed between internode production and its rapid elongation period. Higher stem and internode elongation rates were observed during the reproductive phase (autumn) versus the vegetative phase (summer and spring group). Maximal internode length reached 8–10 cm in summer and spring and 20 cm in autumn, at approximately phytomers 12–13. A similar pattern was reported for all primary axes irrespective of the experiments, the position of the first internode to elongate descending regularly down the main axis. These results provide key elements to predict the onset of stem elongation in the field from simple measurements. They could contribute to improving crop models for perennial tropical C4 grasses.  相似文献

In order to determine the etiology of Syrah decline, virus detection was performed on 22 Syrah clones, chosen for their various levels of sensitivity to Syrah decline. All clones, including the sensitive ones, were free of 20 main grapevine viruses. In contrast, Grapevine Syrah virus-1 and Grapevine Rupestris stem pitting-associated virus (GRSPaV), were detected in 56 % and 100 % of the analysed Syrah clones respectively. This is the first report of GSyV-1 in a French vineyard. The genetic diversity of a 380 nt region within the GRSPaV coat protein gene was studied extensively in vines differing in their sensitivity to Syrah decline. Most GRSPaV variants were scattered between the four phylogenetic groups previously identified; 65 % of the sequences analysed were found to belong to the GRSPaV—group 1, 22 % to—group 2b, 10 % to—group 2a and 2 % to—group 3. Seventy percent of the 31 plants analysed harboured mixtures of genomic variants. Statistical analyses revealed no significant correlation between sensitivity and GRSPaV sequence variation. This suggests that GRSPaV is not the direct etiological agent of the Syrah decline.  相似文献

The critical crop nitrogen uptake is defined as the minimum nitrogen uptake necessary to achieve maximum biomass accumulation (W). Across a range of crops, the critical N uptake is related to W by a power function with a coefficient less than unity that suggests crop N uptake is co-regulated by both soil N supply and biomass accumulation. However, crop N demand is also often linearly related to the expansion of the leaf area index (LAI) during the vegetative growth period. This suggests that crop N demand could be also linked with LAI extension. In this paper, we develop theory to combine these two concepts within a common framework. The aim of this paper is to determine whether generic relationships between N uptake, biomass accumulation, and LAI expansion could be identified that would be robust across both species and environment types. To that end, we used the framework to analyze data on a range of species, including C3 and C4 ones and mono- and di-cotyledonous crops. All crops were grown in either temperate or tropical and subtropical environments without limitations on N supply. The relationship between N uptake and biomass was more robust, across environment types, than the relationship of LAI with biomass. In general, C3 species had a higher N uptake per unit biomass than C4 species, whereas dicotyledonous species tended to have higher LAI per unit biomass than monocotyledonous ones. Species differences in N uptake per unit biomass were partly associated with differences in LAI and N-partitioning. Consequently the critical leaf-N uptake per unit LAI (specific leaf nitrogen, SLN) was relatively constant across species at 1.8–2.0 g m−2, a value that was close to published data on the critical SLN of new leaves at the top of the canopy. Our results indicate that critical N uptake curves as a function of biomass accumulation may provide a robust platform for simulating N uptake of a species. However, if crop simulation models are to capture the genotypic and environmental control of crop N dynamics in a physiologically functional manner, plant growth has to be considered as the sum of a metabolic (e.g. leaves) and a structural (e.g. stems) compartment, each with its own demand for metabolic and structural N.  相似文献

In continuously stocked swards or pastures the frequency at which individual tillers and individual leaves are defoliated by ruminant livestock, relative to leaf lifespan of the grass species within the sward, determines the proportion of each leaf defoliated before senescence, and hence the efficiency of harvesting of herbage. In this paper, sets of data obtained in a range of climatic conditions and with a range of grass species are used in order to document this relationship. It is shown that the frequency of defoliation of individual tillers or individual leaves is closely linked to the average stocking density used within a period of time for maintaining a steady state sward or pasture height, herbage mass or leaf area index. Consequently, any decrease in herbage growth rate should lead to a decrease in the efficiency of harvesting of herbage and then to a more than proportional decrease in total herbage consumption by ruminant livestock. These effects will be more important for grass species having short leaf lifespan than for species with long lifespan. In rotational stocking, the link between herbage growth rate and frequency of defoliation of leaves can be broken by controlling the grazing interval, so any decrease in herbage growth would not be systematically associated with a decrease in efficiency of harvesting of herbage. Rotational stocking should be more efficient than continuous stocking in low herbage production conditions, while in high herbage production systems rotational and continuous stocking would have similar efficiency. The implications of these conclusions for the management of swards and pastures to meet different objectives are discussed briefly.  相似文献

Post-flowering tillering responses of 'Ellett' and 'Grasslands Ruanui' perennial ryegrass ( Lolium perenne L.) cultivars after inflorescence production were studied at INRA, Lusignan in France, at near ambient light (absence of light competition, control treatment) and under shading (low-light intensity and low red–far red ratio). A dense sward containing inflorescence bearing plants of both perennial ryegrass cultivars at ambient light was cut to a height of 50 mm above ground level after which the light treatments were imposed. Forty-five days later, data were collected on the number of tillers per plant, the number of new tillers per original tiller and the dry weight of vegetative and reproductive tillers. Regardless of light treatment, cv. 'Ellett' had fewer tillers per plant, higher weight per tiller and a higher proportion of reproductive tillers than did cv. 'Grasslands Ruanui'. Number of tillers per plant and number of new tillers per tiller were consistently reduced under shade compared with the control treatment for both cultivars. When shaded, weight of reproductive tillers of cvs 'Ellett' and 'Grasslands Ruanui' were 40% and 43%, respectively, less than the control, whereas vegetative tiller weight did not change. In addition, differences between cultivars in the tillering strategy after flowering seem to be unaffected by the light environment. Thus, field observations regarding tillering of cvs 'Ellett' and 'Grasslands Ruanui' were consistent with current results suggesting different grazing managements are needed after flowering to optimize the replacement of tillers for both cultivars.  相似文献
