The pyrimidine analog 5-bromodeoxyuridine (BUdR) competes with thymidine for incorporation into DNA. Substitution of BUdR for thymidine does not significantly affect cell viability but does block cell differentiation in many different lineages. BUdR substitution in a mouse myoblast line blocked myogenic differentiation and extinguished the expression of the myogenic determination gene MyoD1. Forced expression of MyoD1 from a transfected expression vector in a BUdR-substituted myoblast overcame the block to differentiation imposed by BUdR. Activation of BUdR-substituted muscle structural genes and apparently normal differentiation were observed in transfected myoblasts. This shows that BUdR blocks myogenesis at the level of a myogenic regulatory gene, possibly MyoD1, not by directly inhibiting the activation of muscle structural genes. It is consistent with the idea that BUdR selectively blocks a class of regulatory genes, each member of which is important for the development of a different cell lineage.  相似文献

The pea leafminer,Liriomyza huidobrensis (Blanchard), recently introduced into Israel, has proven to be extremely proliferous and difficult to control. It is a much more serious pest of vegetable and flower crops than the previously introducedLiriomyza trifolii (Burgess). Photosynthesis is reduced and cosmetic damage is incurred when adult flies stipple plant leaves with feeding punctures and larvae mine the leaves. Electroencephalography and space-occupying lesions in small animals

EEGs have been recorded from 207 dogs and cats with presumed intra-cranial space-occupying lesions; twenty-seven cases were confirmed postmortem. The clinical presenting signs of the series are considered, and the abnormal characters of the EEGs are analysed and discussed. The place of the EEG in differential diagnosis and its possible future value in association with brain surgery are discussed.
Résumé. On a enregistré des électroencéphalogrammes de 207 chiens et chats suspects de lésions intracraniennes expansives; ce diagnostic a pu être confirmé par l'examen nécropsique. On décrit les manifestations cliniques majeures dans cette série et on considère les anomalies de l'E.E.G., dont on analyse les particularités et dont on envisage l'importance diagnostique sous Tangle de la chirurgie du cerveau.
Zusammenfassung. Es wurden EEGs von 207 Katzen und Hunden mit vermuteten raumbean-spruchenden Läsionen im Schädelraum aufgenommen; 27 Fälle da von wurden durch Autopsie bestätigt. Die klinischen Erscheinungsbilder der Serie werden betrachtet und der anomale Gharakter der EEGs wird analysiert und besprochen. Laboratory study of SO2 dry deposition on limestone and marble: Effects of humidity and surface variables

The dry deposition of gaseous air pollutants on stone and other materials is influenced by atmospheric processes and the chemical characteristics of the deposited gas species and of the specific receptor material. Previous studies have shown that relative humidity, surface moisture, and acid buffering capability of the receptor surface are very important factors. To better quantify this behavior, a special recirculating wind tunnel/environmental chamber was constructed, in which wind speed, turbulence, air temperature, relative humidity, and concentrations of several pollutants (SO₂, O₃, nitrogen oxides) can be held constant. An airfoil sample holder holds up to eight stone samples (3.8 cm in diameter and 1 cm thick) in nearly identical exposure conditions. SO₂ deposition on limestone was found to increase exponentially with increasing relative humidity (RH). Marble behaves similarly, but with a much lower deposition rate. Trends indicate there is little deposition below 20% RH on clean limestone and below 60% RH on clean marble. This large difference is due to the limestone's greater porosity, surface roughness, and effective surface area. These results indicate surface variables generally limit SO₂ deposition below about 70% RH on limestone and below at least 95% RH on marble. Aerodynamic variables generally limit deposition at higher relative humidity or when the surface is wet.  相似文献   

Herbivores promote habitat specialization by trees in Amazonian forests

In an edaphically heterogeneous area in the Peruvian Amazon, clay soils and nutrient-poor white sands each harbor distinctive plant communities. To determine whether a trade-off between growth and antiherbivore defense enforces habitat specialization on these two soil types, we conducted a reciprocal transplant study of seedlings of 20 species from six genera of phylogenetically independent pairs of edaphic specialist trees and manipulated the presence of herbivores. Clay specialist species grew significantly faster than white-sand specialists in both soil types when protected from herbivores. However, when unprotected, white-sand specialists dominated in white-sand forests and clay specialists dominated in clay forests. Therefore, habitat specialization in this system results from an interaction of herbivore pressure with soil type.  相似文献   

The role of technology in enhancing low resource agriculture in Africa

Traditional forms of farming, herding, and fishing are remarkably adapted to African conditions but these traditional approaches are being overtaken by modern pressures, particularly population growth. According to a report published by the Office of Technology Assessment (OTA), a nonpartisan analytical support agency of the U. S. Congress, one promising way to help African farmers and herders would be for development assistance organizations to focus more attention on the various forms of low-resource agriculture that predominate in Africa. In keeping with OTA's mission and primary audience, “Enhancing Agriculture in Africa: A Role for U. S. Development Assistance” (1988) is a policy-oriented synthesis of available technical information. The report provides Congress with a range of options that, if pursued, could help Africans enhance agriculture, increase their food security, and improve their lives. This paper is drawn from the larger OTA report, and it focuses on the role technology might play in enhancing low-resource agriculture. Readers should see the full assessment (OTA, 1988) for more information on policy considerations; the specific technologies mentioned; or a complete list of advisory panel members, workshops and participants, and commissioned papers. OTA's report comes at a critical time: for a variety of reasons—ranging from changing values to increased budget constraints—U. S. foreign assistance policy is undergoing a fundamental reevaluation. This review of the potential of low-resource agriculture, and options the United States might pursue to enhance this approach, was intended to aid in this reevaluation.  相似文献

A technique was developed for studying protein-DNA recognition that can be applied to any purified protein, partially purified protein, or cloned gene. From oligonucleotides in which particular positions are of random sequence, that subset to which a given protein binds is amplified by the polymerase chain reaction and sequenced as a pool. These selected and amplified binding site (SAAB) "imprints" provide a characteristic set of preferred sequences for protein binding. With this technique, it was shown that homo- and heterooligomers of the helix-loop-helix proteins MyoD and E2A recognize a common consensus sequence, CA--TG, but otherwise bind to flanking and internal positions with different sequence preferences that suggest half-site recognition. These findings suggest that different combinations of dimeric proteins can have different binding sequence preferences.  相似文献   

Immunization of cattle against hypodermatosis (Hypoderma lineatum (deVill.) and H. bovis (L.)) using H. lineatum antigens

The feasibility of immunizing calves against Hypoderma lineatum and H. bovis with a crude H. lineatum larval extract or culture-derived antigen was investigated. Larval survival was followed through pupation. Grub appearance in the backs of both of the immunized groups was found to be 50% of that in the control groups. First appearance of H. bovis was significantly retarded in both immunized groups; however, H. lineatum was unaffected. Duration of H. bovis dropping was also shorter in animals receiving crude extract than in controls while duration of dropping of H. lineatum was not influenced by immunization. Survival of H. lineatum larvae to pupation was significantly reduced in animals immunized with crude larval extract, but not in those receiving culture-derived antigen. H. bovis survival was reduced by both treatments.  相似文献   

Feline immunodeficiency virus diagnosis after vaccination

Prior to the widespread use of vaccination for the control of feline immunodeficiency virus (FIV) infection, diagnosis was made by the detection of antibodies against FIV. A number of commercial animal side tests perform quite well for this determination, with positive predictive values between 91 and 100% and negative predictive values between 96 and 100%. Furthermore, results of these tests could be confirmed by western blot analysis of FIV test-positive sera. Currently, a killed whole virus FIV vaccine has been made available to practitioners. Vaccinated cats seroconvert by ELISA and western blot, making presently available diagnostic tests, which rely on antibody detection, useless in cats after vaccination. The advisory panels of the American Association of Feline Practitioners and Academy of Feline Medicine both recommend testing for feline leukemia virus antigen and FIV antibody before vaccination.  相似文献
