Nine isolates of Phtophthora nicotianae were isolated from infected pepper plants. Their pathogenicity was studied in Capsicum annuum in comparison with P. nicotianae isolates from tomato and tobacco. The pathogenicity test showed that pepper isolates of P. nicotianae are adapted to their host. Banding patterns obtained by RAPD analysis with six oligonucleotide primers revealed polymorphism that grouped the isolates independently of the plant host. The polygenic dendrogram showed that pepper isolates were more similar to tomato isolates than to tobacco isolates. The RAPD bands of 1300 and 1500 bp, detected with primers OPD-01 and OPD-10, respectively, appeared specific to the most pathogenic pepper isolates. The OPK-08-1950 seems specific to the isolates of P. nicotianae from tomato. These results suggest that host specified might occur in P. nicotianae and that may be due to interspecific hybridization events resulting in novel pathogenic behavior.  相似文献

ABSTRACT

The objective of this study is to evaluate the impacts of the direct use of natural rock phosphates on two forage legumes, Vicia sativa, and Medicago sativa. Crops were grown in pots containing two P deficient soils and fertilized with Tunisian rock phosphate (TRP) or chemical triple superphosphate (TSP). TRP amendment significantly increased shoot dry yield (+53% to +162%), P-content, protein content, and chlorophyll content mainly at the rate of 150 kg P ha?1. No significant effects were observed on mycorrhization and antioxidant enzyme activities. However, TSP negatively affected the frequency of root mycorrhization and stimulated antioxidant enzyme activities. TRP amendment induced an increase in fluorescein diacetate activity, alkaline phosphatase activity, and species richness. The stimulation involved mainly Actinobacteria, Proteobacteria, and Firmicutes with higher stimulation of Actinobacteria in the case of vetch. The present work indicated a better P-use efficiency of vetch. It suggested also that at least a part of the enhancement observed in plant-growth parameters following TRP fertilization is driven by changes in the structure of rhizosphere microbial communities. Nevertheless, the correlation between changes in microbial composition and phosphorus uses efficiency needs to be investigated further.  相似文献

Chickpea is an important pulse crop grown mainly in the arid and semi-arid regions. The effect of water deficiency on nodulation, biomass production, and competition for nodule occupancy was evaluated in three different soils with two chickpea cultivars, Amdoun I and Chetoui. Two watering regimes were considered; a control that was irrigated three times per week and a water-deficient treatment that was irrigated only one time a week. Results showed that water deficiency significantly decreased the nodule number and the shoot dry weight for both cultivars. Root-nodule bacteria were isolated and characterized by PCR-RFLP of 16S rDNA and nifD-K intergenic spacer. The results show that water deficiency affects the diversity of nodulating rhizobia. The nodulation by Mesorhizobium mediterraneum was reduced while inefficient nodulation by Ensifer meliloti was favoured. In both treatments, chickpea was preferentially nodulated by nifD-K type N6. Analysis for NaCl tolerance showed that most of rhizobia nodulating chickpea under water deficiency are NaCl tolerant. Inoculation with the selected salt-tolerant strain of M. mediterraneum LILM10 increased significantly nodule number and grain yield in the field.  相似文献

The combined effects of NaCl-salinity and phosphorus deficiency on biomass production, nutritional status, and photosynthetic activity were studied in Catapodium rigidum: an annual Poacea with fodder potential. Plants were grown in hydroponic conditions for 55 days under two phosphorus (P) supply: 5 (low) or 180?µmol KH2PO4 (sufficient), in the absence or presence of 100?mM NaCl. Obtained results demonstrated that both salinity and P deficiency treatments applied separately reduced substantially plant growth and CO2 assimilation rate with a more marked impact of salt stress. Salinity has no substantial effect on both shoot P concentrations and phosphorus acquisition efficiency independently of P availability. The highest decrease in plant growth (?91%) was observed in plants simultaneously submitted to both stresses suggesting an additive effect of the two stresses and that P deficiency increased the susceptibility of C. rigidum to salinity. This may be linked to a significant decrease in potassium acquisition (?95%), K/Na selectivity ratio (?73%), stomatal conductance (?66%), CO2 assimilation rate (?64%), and shoot water content (66%). Furthermore, plants cultivated under combined salinity and sufficient P supply displayed higher stomatal conductance, CO2 assimilation rate, K/Na selectivity ratio, and plant growth than plants cultivated under combined effects of salinity and P deficiency. These results suggest that adding P to saline soils could be an alternative for alleviating the negative effects of salinity and may ameliorate salinity tolerance.  相似文献

Faecal samples were collected from 89 dairy calves to determine the prevalence of rotavirus infection in Tunisia and the genomic diversity of bovine rotavirus strains. After screening of all faecal samples by enzyme-linked immunosorbent assay, rotavirus strains were analysed by RNA polyacrylamide gel electrophoresis and characterized antigenically by monoclonal antibodies to the VP6 subgroup. The VP7 genotype was determined by nested RT-PCR. Of the 89 calves tested, 27 (30%) were positive for rotavirus antigen. Four different long electrophoretypes were identified. All VP6 typeable strains carried the subgroup I specificity. G8 genotype was the most prevalent, but G6 and mixed strains G(6 + 8) were also detected.  相似文献

Purpose

Ichkeul National Park, NW Tunisia, is a UNESCO Biosphere Reserve. Garaet El Ichkeul Lake is known for its seasonal variability in water level and salinity. In recent decades, the waterbody has been affected by the construction of new hydraulic structures. To reduce the impacts of dams and to maintain the sustainability of the ecosystem, a sluice was built at the outlet of the lake, and it operated for the first time in 1996. This paper describes an investigation of recent sedimentation dynamics in Ichkeul Lake, determined by radiometric dating of sediment cores.

Materials and methods

A sediment core was collected with a UWITEC gravity corer at the deepest, central part of the lake in August 2009. Specific activities of unsupported lead-210 (210Pb) and caesium-137 (137Cs) were measured in the core, enabling calculation of recent sediment accumulation rates (SAR). Published radiometric data from nearby sediment cores, collected in 1997 and 1982, provide a comparison.

Results and discussion

The measured excess 210Pb inventory was 5300?±?500?Bq?m?2, leading to an estimation of constant flux of 165?±?16?Bq?m?2?yr?1, a value higher than the best estimate for local atmospheric fluxes (123?±?12?Bq?m?2?yr?1) and the flux estimated from the core collected in 1982 (48?Bq?m?2?yr?1). The 137Cs inventory was 3550?±?120?Bq?m?2, two times higher than the historical 137Cs atmospheric deposition in the area. The 137Cs profile displayed a distinct peak, but the 137Cs depth-distribution did not follow the pattern expected from atmospheric deposition. Application of the constant rate of supply (CRS) model, with the reference point method, produced a chronology and SAR values comparable to those found in previous work. The whole 137Cs profile was quantitatively reconstructed from the historical records of atmospheric deposition, using the system-time-averaged (STA) model.

Conclusions

The CRS and STA models provide consistent sediment accumulation results for the whole data set, considering the time resolution of the chronology (~6?years) and analytical uncertainties. Results from cores sampled in 1982, 1997 and 2009 reveal an increasing SAR trend, from ~0.25?g?cm?2?yr?1 in the early 1940s to ~0.67?g?cm?2?yr?1 at present. In the 13?years since installation of sluice gates at Tinja, SAR in the central Ichkeul Lake has not declined. Thus, if siltation continues at the present rate, shallowing of the lake will seriously affect the hydromorphology and ecology of this important lake.  相似文献

Based on the analysis of 11 morphometric variables of body (total length, body area and perimeter, myotome height and eye diameter) and yolk sac (length, height, area, perimeter and volume) of pike larvae, the aim of this study was to evaluate how larval size at hatching and growth of larvae hatched from single egg batches vary according to three hatching times: early, mid and late. Hatching time structures strongly pike larval morphometrics. Early hatched larvae have smaller body sizes at hatch, faster growth and higher yolk use efficiency than late hatched ones. Early hatched larvae seem to be premature and hatch at precocious developmental stage whereas late hatched individuals continue their growth within the egg shell and hatch at larger size but with lower reserves (yolk). A compensatory growth phase was observed for the early hatching pike larvae particularly during the first 5 days post hatch. Consequently, no significant difference in body parameters was recorded from day 10 post hatching whatever the hatching time. The higher growth accomplished by early hatched larvae may be related to a particular metabolic activity that converts more efficiently yolk into body tissues.  相似文献

Since 2006, verticillium wilt of olive induced by Verticillium dahliae has caused considerable economic losses in olive orchards in Tunisia. The genetic structure of V. dahliae isolates collected from different olive growing regions was investigated using virulence tests, vegetative compatibility grouping (VCG) and amplified fragment length polymorphism (AFLP) analyses. In total, 42 isolates of V. dahliae from diseased olive trees were tested. Cluster analysis and principal coordinate analysis revealed that geographic origin was the main factor determining the genetic structure of V. dahliae populations and both methods indicated a genetic separation between the central and coastal isolates. Isolates were divided into two major groups: the AFLP‐I group included all isolates from Sidi Bouzid, Kairouan, Kasserine and Sfax (centre of the country) and the AFLP‐II group included isolates from Monastir, Zaghouane, Sousse, Mahdia (coastal region), and two isolates from Sfax. Analysis of the molecular variance (amova ) indicated a significant level of genetic differentiation among (76%) and within (23%) the two populations. Analyses of both the defoliating (D) and non‐defoliating (ND) pathotypes and VCG markers indicated that most of the isolates belong to VCG 2A and 4B/ND pathotype. The disease severity was highly variable among the isolates tested (< 0·05) with no evidence of association between aggressiveness and geographical origin of the isolates. Overall, results of this study revealed a clear association between the genetic diversity of the isolates and their geographic origin, but not between genetic diversity and virulence patterns.  相似文献

This study aimed to investigate the potential effects of an oral treatment by a newly isolated probiotic Lactobacillus plantarum TN8 strain on trinitrobenzene sulphonic acid (TNBS)‐induced colitis in Wistar rats. Thus, 18 rats were divided into three groups (n = 6 per group): group 1 (control) – rats not receiving TNBS application; group 2 – rats receiving an intrarectal TNBS infusion (100 mg/kg TNBS dissolved in ethanol); and group 3 – rats treated with intragastrical TN8 strain once per day (for 5 days before TNBS induction). The performance and the effects of the probiotic treatment were evaluated using a series of histological, biophysical and biochemical analyses. The results have shown that the treatment with the L. plantarum TN8 strain improves the body weight and reduces the diarrhoea, colonic mucosal inflammation and colon shortening. TN8‐treated rats showed a significant decrease in the total cholesterol content from 1.86 (for group 2) to 1.32 mmol/l and in triglyceride (TG) content from 2.09 (for group 2) to 1.23 mmol/l. Furthermore, the findings revealed that the high‐density lipoprotein (HDL) cholesterol contents increased from 0.95 to 1.02 mmol/l. The histological studies have confirmed that the architecture of the liver and kidney tissues of the TN8‐treated rats were found to be improved. Overall, the results suggest that the L. plantarum TN8 presents promising perspectives for the development of safe and cost‐effective agents for the prevention or alleviation of several intestinal pathologies.  相似文献
