Bovine isoleucyl-tRNA synthetase (IARS) disorder, a major cause of weak calf syndrome, is caused by a homozygous missense (c.235G>C) mutation in the bovine IARS gene of Japanese Black (JB) cattle, which was identified in 2013. However, the extent to which the carrier rate has changed at Kagoshima prefecture, Japan, and whether the carrier status is associated with any clinical or reproductive problems, have yet to be ascertained. In this study, using a real-time polymerase chain reaction-based genotyping assay, we determined the carrier rate in a regional JB cow population at Kagoshima prefecture. Comparative analyses were performed on the metabolic profile test (MPT) results and reproductive performance data obtained for heterozygous carrier and homozygous wild-type cows. In 2009 and 2018, DNA samples were collected from 130 and 462 clinically healthy JB cows, respectively, in Kagoshima prefecture. MPT results and reproductive performance data were evaluated for 62 cows, comprising four heterozygous carriers and 58 wild-type cows. Genotyping revealed that the carrier rate was 6.9% in 2009 and 1.5% in 2018, the difference of which was statistically significant (P<0.005). There were no statistically significant differences between the carrier and wild-type cows with respect to either MPT results or reproductive performance, indicating that the carrier cows have necessary IARS activity to maintain minimal health and reproductive potential. 相似文献

Variation in influenza A viruses was examined by comparison of nucleotide sequences of the NS gene (890 bases) of 15 human viruses isolated over 53 years (1933 to 1985). Changes in the genes accumulate with time, and an evolutionary tree based on the maximum parsimony method can be constructed. The evolutionary rate is approximately 2 X 10(-3) substitution per site per year in the NS genes, which is about 10(6) times the evolutionary rate of germline genes in mammals. This uniform and rapid rate of evolution in the NS gene is a good molecular clock and is compatible with the hypothesis that positive selection is operating on the hemagglutinin (or perhaps some other viral genes) to preserve random mutations in the NS gene. 相似文献

ABSTRACT Production from small scale pond aquaculture in Bangladesh trebled in the decade to 1999 under the influence of extension projects of the Department of Fisheries. The impacts of completed aquaculture extension projects and practices of former participants have rarely been investigated so this was done for four past projects. The different extension approaches were: extension with credit and intensive or little supervision; demonstrations; group extension and informal extension by fry traders; and adaptive research-development. Generally extension recipients and non-participants had similar sized landholdings, but demonstration farmers had larger farms and ponds. Extension recipients mainly received information from project-related sources and rated its utility highly. Neighbours and control farmers mainly obtained information from mass media and other farmers, over 90% of these households also cultivated fish. Extension recipients generally followed better practices: for example stocking fewer fingerlings and achieving higher production than non-participants, but with wide variations. Training of fry traders appeared to reduce overstocking in general probably due to the advice of fry traders. Overall, 2–6 years after extension the recipients have continued to farm fish, have a fair understanding of good practice and achieve reasonable yields. Limited demonstration effects occurred. Extension through group approaches and informal agents seem to be cost effective for the many pond owners in Bangladesh. 相似文献

Comparative evaluation of anthropometric measures to predict cardiovascular risk factors in Tehranian adult women 总被引:1,自引:0,他引:1
OBJECTIVE: To compare the ability of waist circumference (WC), body mass index (BMI), waist-to-hip ratio (WHR) and waist-to-height ratio (WHtR) to predict cardiovascular risk factors in an urban adult population of Tehranian women. DESIGN: Population-based cross-sectional study. SETTING: Tehran, the capital of Iran. SUBJECTS: This study was conducted on 5073 women aged 18-74 years, participants of the Tehran Lipid and Glucose Study. Demographic data were collected. Anthropometric indices were measured according to standard protocols. Cut-off points of BMI, WC, WHR and WHtR were considered as 25 kg m(-2), 80 cm, 0.8 and 0.5, respectively. Blood pressure was measured and hypertension was defined based on the sixth report of the Joint National Committee on Prevention, Detection, Evaluation and Treatment of High Blood Pressure. Biochemical analyses were conducted on fasting blood samples. Diabetes was defined as fasting plasma glucose > or = 126 mg dl(-1) or 2-hour plasma glucose > or = 200 mg dl(-1) and dyslipidaemia based on the third report of the National Cholesterol Education Program Expert Panel. The presence of 'at least one risk factor' from the three major cardiovascular risk factors (hypertension, dyslipidaemia and diabetes) was also evaluated. RESULTS: Mean (+/-standard deviation) age of women was 39.9+/-14.6 years; mean BMI, WC, WHR and WHtR were 27.1+/-1.5 kg m(-2), 86.5+/-13.5 cm and 0.83+/-0.08 and 0.55+/-0.08, respectively. Of the four anthropometric measures, WC had the highest sensitivity and specificity to identify subjects with risk factors in both the 18-39 year and the 40-74 year age categories. WC was seen to have a higher percentage of correct prediction than BMI, WHR and WHtR. CONCLUSION: It is concluded that WC is the best screening measure for cardiovascular risk factors, compared with BMI, WHR and WHtR, in Tehranian adult women. 相似文献

Measurement of glycated hemoglobin percentages for use in the diagnosis and monitoring of diabetes mellitus in nonhuman primates 总被引:2,自引:0,他引:2
OBJECTIVE: To identify a technique for measurement of glycated hemoglobin percentage in blood samples obtained from various species of nonhuman primates (NHPs), to determine whether these percentages varied with respect to glycemic control, and to assess whether this physiologic variable provided a suitable test for diagnosing diabetes mellitus in NHPs. SAMPLE POPULATION: 166 blood samples collected from 121 NHPs comprising 22 species from the Haplorhine and Strepsirhine suborders and including nondiabetic, treated-diabetic, and diabetic animals in 23 zoologic institutions throughout the United States. PROCEDURE: Hemoglobin A1c percentage was measured in 154 samples by use of high-performance liquid chromatography. Total glycated hemoglobin percentage was measured in 159 samples by use of a boronate-affinity chromatographic assay. Glucose concentration was measured in 157 samples with an autochemical analyzer by use of a hexose kinase method. RESULTS: The boronate-affinity chromatographic technique for measurement of total glycated hemoglobin percentage was the most suitable method. Nondiabetic Haplorhines had percentages higher than those in nondiabetic Strepsirhines. In Haplorhines, diabetic animals had percentages higher than those in treated-diabetic animals, which had percentages higher than those in nondiabetic animals. In Strepsirhines, this pattern was less pronounced. CONCLUSIONS AND CLINICAL RELEVANCE: Measurement of total glycated hemoglobin percentage provides useful information for diagnosing diabetes mellitus in Haplorhines and, possibly, in Strepsirhines. Until reference ranges are established for each species, it is recommended that results for samples from NHPs without clinical signs of diabetes mellitus be compared with results of samples collected concomitantly from NHPs with clinical signs of this condition. 相似文献
