Canine inflammatory mammary cancer (IMC) has been proposed as a model for the study of human inflammatory breast cancer (IBC). The aims of this study were to compare the immunohistochemical expression of aromatase (Arom) and several hormone receptors [estrogen receptor α (ERα), estrogen receptor β (ERβ), progesterone receptor (PR) and androgen receptor (AR)], in 21 IMC cases vs 19 non‐IMC; and to study the possible effect of letrozole on canine IMC and human inflammatory breast cancer (IBC) in vitro using IPC‐366 and SUM‐149 cell lines. Significant elevations of the means of Arom Total Score (TS), ERβ TS and PR TS were found in the IMC group (p = 0.025, p = 0.038 and p = 0.037, respectively). Secondary IMC tumours expressed higher levels of Arom than primary IMC (p = 0.029). Non‐IMC PR‐ tumours contained higher levels of Arom than non‐IMC PR+ tumours (p = 0.007). After the addition of letrozole, the number of IMC and IBC cells dropped drastically. The overexpression of Arom found and the results obtained in vitro further support canine IMC as a model for the study of IBC and future approaches to the treatment of dogs with mammary cancer, and especially IMC, using Arom inhibitors.  相似文献

Free and total fluorescent compounds, browning index, and color formation were measured in milk-based powdered infant formulas (IF) during 2 years of storage at 20 and 37 degrees C. The excitation spectra from 415 nm emission show three peaks (ex lambda1 = 270 nm, lambda2 = 325/315 nm, lambda3 = 350 nm) and from 347 nm excitation two emission peaks (415 and 520 nm), and no wavelength shifts were observed. Temperature and time of storage exert in general no significant effect on the development of fluorescence emission intensity and browning index. However, an important increase in pentodilysine was recorded-probably because of the iron and ascorbic acid contents of the samples-as well as in browning index in adapted IF. In both IF a color increase (deltaE) throughout storage was observed, this increase being greater in samples stored at 37 degrees C than in those stored at 20 degrees C. The increase in color with time fitted a linear regression model. Color appeared to be an indicator of sufficient sensitivity to measure the effect of temperature or storage time.  相似文献

Anthropogenic conversion of primary forest to pasture for cattle production is still frequent in the Amazon Basin. Practices adopted by ranchers to restore productivity to degraded pasture have the potential to alter soil N availability and N gas losses from soils. We examined short-term (35 days) effects of tillage prior to pasture re-establishment on soil N availability, CO2, NO and N2O fluxes and microbial biomass C and N under degraded pasture at Nova Vida ranch, Rondônia, Brazilian Amazon. We collected soil samples and measured gas fluxes in tilled and control (non tilled pasture) 12 times at equally spaced intervals during October 2001 to quantify the effect of tillage. Maximum soil NH4+ and NO3 pools were 13.2 and 6.3 kg N ha−1 respectively after tillage compared to 0.24 and 6.3 kg N ha−1 in the control. Carbon dioxide flux ranged from 118 to 181 mg C–CO2 m2 h−1 in the control (non-tilled) and from 110 to 235 mg C–CO2 m2 h−1 when tilled. Microbial biomass C varied from 365 to 461 μg g−1 in the control and from 248 to 535 μg g−1 when tilled. The values for N2O fluxes ranged from 1.22 to 96.9 μg N m−2 h−1 in the tilled plots with a maximum 3 days after the second tilling. Variability in NO flux in the control and when tilled was consistent with previous measures of NO emissions from pasture at Nova Vida. When tilled, the NO/N2O ratio remained <1 after the first tilling suggesting that denitrification dominated N cycling. The effects of tilling on microbial parameters were less clear, except for a decrease in qCO2 and an increase in microbial biomass C/N immediately after tilling. Our results suggest that restoration of degraded pastures with tillage will lead to less C matter, at least initially. Further long-term research is needed.  相似文献

The effect of forest structure and health on the relative surface temperature captured by airborne thermal imagery was investigated in Norway Spruce-dominated stands in Southern Finland. Airborne thermal imagery, airborne scanning light detection and ranging (LiDAR) data and 92 field-measured sample plots were acquired at the area of interest. The surface temperature correlated most negatively with the logarithm of stem volume, Lorey’s height and the logarithm of basal area at a resolution of 254?m2 (9?m radius). LiDAR-derived metrics: the standard deviations of the canopy heights, canopy height (upper percentiles and maximum height) and canopy cover percentage were most strongly negatively correlated with the surface temperature. Although forest structure has an effect on the detected surface temperature, higher temperatures were detected in severely defoliated canopies and the difference was statistically significant. We also found that the surface temperature differences between the segmented canopy and the entire plot were greater in the defoliated plots, indicating that thermal images may also provide some additional information for classifying forests health status. Based on our results, the effects of forest structure on the surface temperature captured by airborne thermal imagery should be taken into account when developing forest health mapping applications using thermal imagery.  相似文献

1. Lake Rodó is a turbid system, a condition attributed to algal biomass. The proximal source of the eutrophication was stormwater discharges from an ill‐defined urban area. This paper describes an attempt to restore the water quality of Lake Rodó, the first time this has been done in Uruguay. In spring 1996 it was drained, sediments were removed and stream inputs were diverted. Groundwater was used to re‐fill the lake. Due to its high nutrient concentration a re‐circulation system was designed, pumping water from associated pools covered with free‐floating plants. 2. After the lake was refilled, the system was characterized by oxygen saturation or over‐saturation, neutral to basic pH, and high phosphorus, nitrogen and silicate concentrations. Ratios of total nitrogen (TN):total phosphorus (TP) and chlorophyll a (Chl a):TP indicated that phosphorus was the primary limiting nutrient during the period of groundwater supply. Once groundwater pumping had ceased, there was a decrease in TN:TP and Chl a:TP ratios, suggesting N‐limiting conditions prevailed in some periods. 3. Before restoration, the phytoplankton community was dominated year‐round by Planktothrix agardhii; since restoration the community has been more diverse. This change has favoured grazing by mesozooplankton, and the onset of clear‐water phases in spring. 4. Abundant populations of small omnivorous fish maintained a high predation pressure on zooplankton, restricting the abundance of large‐bodied herbivores, which, in turn, allowed an increase in phytoplanton biomass and a decrease in water transparency. Based on this observation, together with the phosphorus concentration and the low abundance of filamentous cyanobacteria compared with previous studies, we suggest that top‐down control has played a key role in increasing transparency in Lake Rodó. 5. A nutrient reduction programme, by the mechanical harvest of floating plants, and a removal of small omnivorous fishes and stocking strictly with piscivores, could be key factors in the achievement of a stable clear‐water phase. However, if blooms of Microcystis or other similar genera occur in summer, additional measures (e.g. reduction of the hydraulic residence time) will be needed to improve water transparency. The total arsenic, inorganic arsenic, lead, cadmium, and mercury contents of 18 algae food products currently on sale in Spain were determined. The suitability of the analytical methodologies for this type of matrix was confirmed by evaluating their analytical characteristics. The concentration ranges found for each contaminant, expressed in milligrams per kilogram of dry weight, were as follows: total arsenic, 2.3-141; inorganic arsenic, 0.15-88; lead, < 0.05-1.33; cadmium, 0.03-1.9; and mercury, 0.004-0.04. There is currently no legislation in Spain regarding contaminants in algae food products, but some of the samples analyzed revealed Cd and inorganic As levels higher than those permitted by legislation in other countries. Given the high concentrations of inorganic As found in Hizikia fusiforme, a daily consumption of 1.7 g of the product would reach the Provisional Tolerable Weekly Intake recommended by the WHO for an average body weight of 68 kg. A more comprehensive study of the contents and toxicological implications of the inorganic As present in the algae food products currently sold in Spain may be necessary, which might then be the basis for the introduction of specific sales restrictions.  相似文献

Bitter taste, an organoleptic characteristic of virgin olive oil, has been related to phenolic compound composition. The usual method to assess this attribute is by a sensorial panel of tasters, while in the laboratory; methods based on physicochemical properties have been assayed as K225, the most widely used one. However, a direct determination of bitterness in virgin olive oil is useful for quality-control purposes. The proposed method is supported by the observable spectral change undergone by the compounds responsible for bitterness as pH varied. This measurement was carried out directly in the oil, without prior isolation of bitter analytes. The difference of absorbance between alkaline and neutral medium showed a highly significant correlation (r = 0.988, p < 0.0001) with the conventional parameter (K225). The method was rapid, required a small sample, allowed direct determination of bitterness in virgin olive oil, and could be easily automated.  相似文献
