Effects of amikacin, gentamicin, kanamycin, and streptomycin on renal tissue superoxide dismutase, glutathione peroxidase, glutathione and malondialdehyde, serum creatinine, potassium, sodium, total protein, glucose, uric acid, and total bilirubin levels were investigated. Pyriproxyfen is a biorational insecticide that acts as a juvenile hormone (JH) analogue and disrupts insect development with an unknown molecular mode of action. Pyriproxyfen is one of the major insecticides used to control the whitefly Bemisia tabaci (Gennadius) and comply with integrated pest management (IPM) programmes, resulting in minimal effects on the environment, humans and beneficial organisms. During the last few years, resistance to pyriproxyfen has been observed in several locations in Israel, sometimes reaching a thousandfold or more. No information exists about the molecular basis underlying this resistance that may lead to understanding the mode of action of pyriproxyfen and developing molecular markers for rapid monitoring of resistance outbreaks. In this communication, a cDNA microarray from B. tabaci was used to monitor changes in gene expression in a resistant B. tabaci population. Based on statistical analysis, 111 expressed sequence tags (ESTs) were identified that were differentially upregulated in the resistant strain after pyriproxyfen treatment. Many of the upregulated ESTs observed in the present study belong to families usually associated with resistance and xenobiotic detoxification such as mitochondrial genes, P450s and oxidative stress, genes associated with protein, lipid and carbohydrate metabolism and others related to JH-associated processes in insects such as oocyte and egg development.  相似文献

ABSTRACT:   The effects of the different catching methods (gillnet, longline, harpoon) on sensory, chemical (pH, total volatile base nitrogen, K -value) and microbiological (total viable count [TVC]) changes in pike perch Sander lucioperca stored in ice were investigated. The same soaking time was used for both gillnet and longline fishing. The catching method had considerable influence on the freshness quality of pike perch. The acceptable shelf life was 15 days for pike perch caught by gillnet, and 22 days for longline and harpoon. The initial concentrations of inosine monophosphate (2.4 μmol/g) in pike perch caught by gillnet were significantly lower ( P  < 0.05) than longline (4.1 μmol/g), and especially by harpoon (16.7 μmol/g). However, the initial K -values for fish caught by harpoon were significantly ( P  < 0.05) lower (24.36%) than fish caught by longline and gillnet (57.69%, 64.41%, respectively). The average K , Ki, G and H -values at rejection day in terms of sensory assessment were approximately 90, 98, 156 and 40%, respectively, for all catching methods during ice storage. However, TVC reached 7.0 log cfu/g after approximately 11 days of storage for fish caught by gillnet, 19 days for fish caught by longline and 8 days for fish caught by harpoon. The result of this study suggests that the best catching method for preserving the freshness of pike perch is longline, based on the data obtained from the sensory and microbiological analysis.  相似文献

Understanding cancer pathogenesis requires knowledge of not only the specific contributory genetic mutations but also the cellular framework in which they arise and function. Here we explore the clonal evolution of a form of childhood precursor-B cell acute lymphoblastic leukemia that is characterized by a chromosomal translocation generating a TEL-AML1 fusion gene. We identify a cell compartment in leukemic children that can propagate leukemia when transplanted in mice. By studying a monochorionic twin pair, one preleukemic and one with frank leukemia, we establish the lineal relationship between these "cancer-propagating" cells and the preleukemic cell in which the TEL-AML1 fusion first arises or has functional impact. Analysis of TEL-AML1-transduced cord blood cells suggests that TEL-AML1 functions as a first-hit mutation by endowing this preleukemic cell with altered self-renewal and survival properties.  相似文献

Fourier transform infrared spectroscopy (FT-IR, 4000-600 cm(-)(1)) was used to discriminate between intact and sonication-injured Listeria monocytogenes ATCC 19114 and to distinguish this strain from other selected Listeria strains (L. innocua ATCC 51742, L. innocua ATCC 33090, and L. monocytogenes ATCC 7644). FT-IR vibrational overtone and combination bands from mid-IR active components of intact and injured bacterial cells produced distinctive "fingerprints" at wavenumbers between 1500 and 800 cm(-)(1). Spectral data were analyzed by principal component analysis. Clear segregations of different intact and injured strains of Listeria were observed, suggesting that FT-IR can detect biochemical differences between intact and injured bacterial cells. This technique may provide a tool for the rapid assessment of cell viability and thereby the control of foodborne pathogens.  相似文献

The effect of chlorine-induced bacterial injury on spectral features using Fourier transform infrared (FT-IR) absorbance spectroscopy was studied using a mixed bacterial culture of (1:1) ca. 500 CFU/mL each Escherichia coli ATCC 25922 and Pseudomonas aeruginosa ATCC 15442 in 0.9% saline. Bacterial cells were treated with 0, 0.3, or 1.0 ppm of initial free chlorine (21 degrees C, 1 h of contact time). Chlorine-injured and dead bacterial cells retained the ATR spectral properties of uninjured or live cells in the region of C-O-C stretching vibrations of polysaccharides, indicative of the cell wall peptidoglycan layer and lipopolysaccharide outer leaflet. This confirms the observations of others that extensive bacterial membrane damage is not a key factor in the inactivation of bacteria by chlorine. The bactericidal effect of chlorine caused changes in the spectral features of bacterial ester functional groups of lipids, structural proteins, and nucleic acids, with apparent denaturation reflected between 1800 and 1300 cm (-1) for injured bacterial cells. Three-dimensional principal component analysis (PCA) showed distinct segregation and clustering of chlorine-treated and untreated cells. Cells exposed to chlorine at 0.3 or 1.0 ppm could be distinguished from the untreated control 73 and 80% of the time, respectively, using soft independent modeling of class analogy (SIMCA) analysis. This study suggests that FT-IR spectroscopy may be applicable for detecting the presence of injured and viable but not culturable (VBNC) waterborne pathogens that are underestimated or not discernible using conventional microbial techniques.  相似文献

A risk analysis was undertaken in an attempt to improve improvised rump straps on donkeys carrying tourists at the World Heritage Site at Petra, Jordan. Tail-base lesions were identified in 63 of the 86 donkeys. Observations and questionnaires were used to collect data relating to the straps, donkey health and human attitudes. The worse lesions were associated with padded rather than unpadded straps, if tightly fitted. Padding could be a cause of, or a response to lesions, but results suggest that it did not effectively aid healing. Significantly worse lesions occurred with unclean than with clean straps and, contrary to many recommendations, cotton straps were associated with worse lesions than were synthetic straps. Since this was an exploratory study, findings should be considered to generate (not to test) hypotheses and any resulting interventions will require monitoring. Further possible risks are discussed, referring to medical and veterinary literature and applied expertise in working equines.  相似文献

To reduce reliance on scientific fertilizer due to rapid increase of fertilizer prices and environmental constraint, it necessary to improve crop productivity and soil fertility on sustainable basses. Utilization of “summer gap” through biochar and legumes have pleasant effects on improving crop productivity and soil fertility on long term basses. Two years’ field experiments were conducted on wheat and maize crops with “summer gap” utilization with legumes and biochar at research farm of agronomy, the University of Agriculture Peshawar during 2011–2013. Wheat-maize-wheat cropping system was followed with the adjustment of legumes in “summer gap” (land available after wheat harvest till maize sowing). Legumes i.?e. mung bean, cowpea and Sesbania with a fallow were adjusted in the “summer gap” with and without biochar application. Biochar was included at the rate of 0 and 50?t?ha-1 with four N levels of 0, 90, 120, 150 and 0, 60, 90, 120?kg?ha-1 to subsequent maize and wheat crops, respectively. In legumes’ experiment, biochar increased fresh and dry fodder yield in cowpea and Sesbania, grain and biological yields in mung bean. In maize experiments, biochar improved grain yield. Nitrogen application increased grain and biological yields. In wheat experiments with increasing nitrogen level enhanced biological and grain yields. It is concluded that use of biochar and legumes in “summer gap” improve overall farm productivity and soil fertility on sustainable basses.  相似文献

BACKGROUND: The presence of certain symbiotic microorganisms may be associated with insecticide resistance in insects. The authors compared the susceptibility of two isofemale lines, Rickettsia-plus and Rickettsia-free, of the sweet potato whitefly Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) to major insecticides from different chemical groups, including imidacloprid, acetamiprid, thiamethoxam, pyriproxyfen, spiromesifen and diafenthiuron. RESULTS: While the Rickettsia-plus and Rickettsia-free lines showed no differences in their susceptibility to imidacloprid and diafenthiuron, higher susceptibility of the Rickettsia-plus line to acetamiprid, thiamethoxam, spiromesifen and especially pyriproxyfen was observed. LC(90) values indicated that the Rickettsia-free line was 15-fold more resistant to pyriproxyfen than the Rickettsia-plus line. CONCLUSION: Findings indicate that the infection status of B. tabaci populations by Rickettsia is an important consideration that should be taken into account when performing resistance monitoring studies, and may help in understanding the dynamics of B. tabaci resistance, symbiont-pest associations in agricultural systems and the biological impact of Rickettsia on whitefly biology.  相似文献

Many agrochemical companies in Turkey purchase generic active ingredients (a.i.) and, by way of ‘me-too’ registration, sell pesticides under different brand names. Farmers and researchers often claim that these products are ineffective. In order to determine the a.i. content and efficacy of me-too registered abamectin-based products, we studied seven (in 2004) and ten (in 2005) products, which were chosen based on their availability and high market share. Differences in both efficacy and a.i. contents were observed in the products investigated in the study, but making speculative statements that could affect all me-too registered products based on our investigations of only 17 abamectin-based samples is not realistic. Timectin® was found to be statistically less effective onTetranychus cinnabarinus in both laboratory and greenhouse trials than all other tested products. Furthermore, Timectin was determined to have 69.8% less a.i. than in its product specifications. Our findings make it clear that grower complaints about the qualities of agricultural pesticides are based on actual chemical inadequacies of formulated pesticides. Analysis of the a.i. should be carried out as part of regular market inspection in order to determine whether agricultural pesticides comply with their product specifications. In this way growers will be provided with good quality but inexpensive products.  相似文献
