Current and past industrial pollution leaves many traces in the environment, in particular along rivers in industrial and urban areas. The isotopic analysis of the lead found in soils and tree rings offers a kind of environmental archive for presenting a portrait of the pollutant distribution in the environment in both spatial and temporal terms. This study is an attempt to identify and compare the source of contamination found in soils and tree rings located along two rivers affected by pollution over several years. Specifically, the focus is on the pattern of lead concentrations and lead isotopic signatures (206Pb/207Pb, 208Pb/206Pb, and 206Pb/204Pb) detected in soils and tree rings located on polluted floodplains. The concentration of Pb in overbank sediments does not rise with the increasing distance downstream from the point source (mining area), suggesting that significant fluvial transport of the pollutant particles over 80 km is involved. For the soil profiles, Pb concentration levels range between 12.32 and 149.13 mg/kg, with the highest concentrations found at the base of the profiles (>1 m). For the lead isotope ratios in the soil profiles, the values obtained range from 0.851 to 0.872 (206Pb/207Pb), 2.081 to 2.111 (208Pb/206Pb), and 0.547 to 0.562 (206Pb/204Pb). The tree ring analysis of red ash (Fraxinus pennsylvanica Marsh.) shows average lead concentrations of 0.63 μg/g, and the lead values of all the tree specimens range between 0.03 and 11.38 μg/g. Pb concentrations varied greatly between the specimens in selected sites and lead isotope ratios in the tree rings showed a strong variability in the time series, particularly from 1945 to 1970. The greater number of variations in the lead concentration rates and isotopic ratios suggest that many more events associated with pollution and contamination have in fact occurred in this area. The study demonstrates the utility of combining stable isotope analyses (soils and tree rings) to examine the source and dispersion of contaminant Pb in fluvial systems by providing reliable and robust indicators for the detection of environmental changes on a local and regional scale.  相似文献

The genetic variation in equine arteritis virus (EAV) GL protein encoding gene was investigated. Nucleic and deduced amino acid sequences from 7 different EAV isolates, including 4 eastern Canadian field isolates, were compared with those of the Bucyrus reference strain. Nucleotide sequence identities between these isolates and the Bucyrus reference strain ranged from 87.5% (Vienna isolate) to 93.9% (11958 isolate). Amino acid identities with the Bucyrus reference strain varied from 90.2% (Vienna isolate) to 95.1% (19933 isolate). A 2nd potential N-linked glycosylation site was found at position 81 in the GL protein of all EAV isolates. Three amino acid substitutions at residue position 90 (Glu-->Val), position 101 (Ala-->Val or Thr), and position 119 (Val-->Leu, Phe or Ser) were also found in all EAV isolates. Phylogenetic analysis showed that the North American EAV isolates, including the Canadian isolates, and the European prototype Vienna isolate could be classified in 2 distinct groups. Three putative sequential antigenic sites were predicted in EAV GL protein. The 1st antigenic site (TAQRFT) was located at positions 24 to 29, and the 2nd antigenic site (RYDEHTA) at positions 47 to 53. The 3rd antigenic site was predicted to be located at positions 78 to 84 and showed the less conserved amino acid sequence.  相似文献

Habitat alteration caused by forest harvesting seems to contribute to the decline of forest-dwelling caribou, an ecotype of woodland caribou (Rangifer tarandus caribou) inhabiting the boreal ecosystem. To serve as basework to the establishment of conservation measures for the species, we have studied the hierarchical habitat selection of forest-dwelling caribou in a boreal landscape of Québec strongly impacted by logging. Fifteen females were surveyed by GPS telemetry between April 2004 and March 2006. Home ranges showed a high proportion of 90–120 year-old forests, a low proportion of regenerating forests (20–40 years old) and a tendency to include a greater proportion of 6–20 year-old clearcuts in relation to their availability in the study area. At the home range scale, selection patterns differed between periods, possibly reflecting specific requirements linked to caribou life cycle. Caribou selected open lichen woodlands throughout the year while mature closed forests (≥50 years) were selected uniquely during summer. The 6–20 year-old clearcuts were avoided during calving, in summer and during the rutting period but were selected during spring. Our results indicate that mature forest and open lichen woodlands are highly selected forest cover types by caribou at both spatial scales. Although clearcuts were generally avoided at the home range scale, such avoidance was not observed at the larger scale, the search for 90–120 year-old forests being hampered by a uniform distribution of clearcuts. An a posteriori landscape analysis highlighted the spatial association between 6–20 year-old clearcuts and 90–120 year-old forests, an association that can be explained by the current regulations used in Québec. Our results underline the importance of pursuing research concerning the impact of such an exploitation regime on the long-term maintenance of the forest caribou in the boreal landscape.  相似文献

The extreme 5' end, the entire leader sequence of the Arvac vaccine strain, and 10 equine arteritis virus (EAV) isolates, including the ATCC Bucyrus reference strain and 5 Canadian field isolates, were determined and compared at the primary nucleotide and secondary structure levels. The leader sequence of eight EAV isolates, including the Bucyrus reference strain, and the leader sequence of the Arvac vaccine strain was determined to be 206 nt in length (not including the putative 5' cap structure-associated nucleotide) whereas those of the 86AB-A1 and 86NY-A1 isolates were found to be 205 and 207 nt in length, respectively. The sequence identity of the leader sequences, between the different isolates and the Bucyrus reference strain, ranged from 94.2 to 98.5%. Phylogenetic analysis and estimation of genetic distances, based on the leader nucleic acid sequences, showed that all EAV isolates/strains are likely to represent a large phylogenetically-related group. An AUG start codon found at position 14 in all EAV isolates/strains could initiate an open reading frame (ORF) that could produce a polypeptide of 37 amino acids, except for the 86NY-A1 isolate where the intraleader polypeptide would contain 54 amino acids. Computer-predicted RNA secondary structures were identified in the 11 EAV leader regions analyzed. All EAV isolates/strains showed 3 conserved stem-loops (designated A, B and C). An additional conserved stem-loop (D) was observed in 7 EAV isolates, including the Bucyrus reference strain. The leader region distal to stem-loop D did not contain conserved sequences or stem-loop structures common to the EAV isolates/strains.  相似文献

Forty-seven dairy heifers of approximately 10 d of age were assigned to a factorial experiment in which a supplement of folic acid (0 or 40 mg) administered weekly by i.m. injection and level of feed intake were the two factors studied. The heifers were weaned after 5 wk of experimentation. Following weaning, and until the end of the experiment, 11 wk later, they had ad libitum access to grass hay and concentrates at two different levels, ad libitum or restricted, to allow a body weight gain of 700 g/d. A supplement of folic acid (P less than .05) and ad libitum access to feed (P less than .05) increased the mean concentration of serum folates. Blood hemoglobin and packed cell volume were not affected by the level of feed intake. However, they were both increased (P less than .05) by the supplement of folic acid. Average daily gain was analyzed over three different periods: 0 to 5 wk (before weaning), 5 to 10 wk, and 10 to 16 wk. Average daily gain was increased by the supplement of folic acid during the second period (P less than .05) and by ad libitum access to feed during the last two periods (P less than .05). Ad libitum access to feed increased (P less than .05) weight of the liver, decreased the (P less than .05) concentrations of RNA and DNA, and increased (P less than .05) the ratios of protein/DNA and RNA/DNA. The supplement of folic acid decreased (P less than .05) weight of the liver and increased the ratio RNA/DNA (P less than .05). These effects of supplement of folic acid on growth performance and on hematological cells may reflect a lack of folic acid during the weeks after weaning.  相似文献

This article pertains to the fluvial dynamics of rivers in southern Québec, in particular to the aggradation and pedogenetic processes observed in floodplains affected by periodic floods. The frequency of flood events, notably along the Saint-François River and its main tributaries, leads to fine materials being frequently deposited on floodplains and affected development of alluvial soils. Particle size and physical and chemical analyses have led to a better understanding of the dynamics involved in the formation of floodplains and the development of soils related to this fluvial environment. Also, sedimentological analyses (layer texture and thickness, microstructure) combined with radiocarbon dating (14C) and isotopic methods (210Pb, 226Rd) enabled the determination of sedimentation rates along the floodplains. The radiocarbon dating results obtained from the organic layers buried in alluvial soils show relatively variable ages, i.e. between 2210 ± 60 and 30 ± 70 years BP. The data gathered reveal an active overbank deposition, which shows evidence of the high flood recurrence in many rivers of the basin. It is estimated that the annual floodplain aggradation ranges from 1.0 to 7.6 mm yr− 1 on average, which causes increases in floodplain elevations (vertical accretion). The sedimentation rates obtained using the constant rate supply dating model (210Pb) show average values that range from 2.1 to 10.7 mm yr− 1. Also, the presence of contaminated layers at the lower level (> 100 cm) of the floodplains suggested an active sedimentation rates along the rivers affected by floods.  相似文献

Numerous efforts have been invested in designing and configuring residual forest stands in Canadian boreal forest to preserve their overall biodiversity. Now that several landscapes have been partially logged, the next issue in forest management involves the planning of residual forest stand harvesting without compromising wildlife populations. Residual stands can be cut when adjacent regeneration reaches 3 m in height according to current regulations in several Canadian provinces (e.g., Québec, Ontario, Alberta, and British-Columbia). However, little is known on whether such regenerating habitat (RE-3m) can maintain wildlife communities similar to those found in unharvested mature forest (CO). We estimated the relative abundance of small mammals and forest birds in RE-3m and CO habitats and characterized landscape and stand structures. These variables were then compared between the two contrasting successional stages and were used to build habitat use models (HUMs) for 21 species. CO and RE-3m differed with regard to several landscape characteristics and stand structure variables as a result of logging. Snowshoe Hare, Northern Flicker, Alder Flycatcher, Ruby-crowned Kinglet, White-throated Sparrow and Magnolia Warbler were more abundant in RE-3m than CO, while Red-backed Vole, Brown Creeper and Golden-crowned Kinglet exhibited lower abundances in RE-3m. No significant differences in abundance were observed for the 12 other species. Species HUMs were highly significant and explained between 64.3 and 99.1% of the total variability in abundance. Following variance partitioning, stand structure variables accounted for most of the explained variability (54.2%) while landscape characteristics accounted for only 28.7%. No difference in species richness was observed but community evenness was greater in CO than RE-3m. Our results suggest that current regulations may threaten the maintenance of 3 out of 21 censused species for which abundances were significantly lower in regenerating 3 m tall stands. As stand structure explained a large amount of variability in abundance, it should be considered during timber harvest planning in both mature and regenerating stands. Until we know more on whether the current regulations are suitable for maintaining overall biodiversity, our results suggest that some mature forest stands should be maintained within managed landscapes for a complete logging rotation period.  相似文献

Interleukin 2 (IL-2) is a lymphokine produced by activated T helper lymphocytes which exerts immunoregulatory effects on a variety of immune cells, including T cells, activated B cells, natural killer cells, and lymphokine-activated killer cells. In this study, we cloned and determined the entire beluga whale (Delphinapterus leucas) and grey seal (Halichoerus grypus) IL-2-encoding cDNA sequences, and analysed their genetic relationships with those from several mammalian species obtained from the Genbank Database. The encoding nucleic acid sequences of beluga whale and grey seal IL-2 were 465 and 468 bp in length, respectively. The identity levels of IL-2 nucleic and deduced amino acid sequences from the beluga whale and grey seal with those from the other mammalian species, ranged from 59.9% to 89.5%, and 52.9% to 77.3%, and from 61.1% to 93.2%, and 58.7% to 88.4%, respectively. Phylogenetic analysis based on both nucleic and amino acid sequences showed that the beluga whale IL-2 was closely related to that of the ruminant species, which includes the bovine, while the grey seal IL-2 was closely related to that of the canine.  相似文献
