It has been postulated that osmotic effects on plant tissue are mediated by abscisic acid (ABA). Hybrid larch (Larix kaempferi (Lambert) Carr. x L. decidua Mill.) plantlets, transformed with the ABA-inducible wheat Em promoter associated with the Gus reporter gene, were axenically inoculated with two ectomycorrhizal fungi: Cenococcum geophilum Fr., considered tolerant to water stress, and Laccaria bicolor (Marie) Orton, considered less tolerant to drought. The mycorrhizal and non-mycorrhizal transgenic plantlets were subjected to osmotic stress by adding polyethylene glycol (PEG) to the culture medium. In the presence of PEG, L. bicolor and C. geophilum reduced shoot water potential and turgor potential, but increased host osmotic potential. Treatment of plantlets with PEG induced a significant increase in endogenous ABA concentrations. Laccaria bicolor and C. geophilum behaved similarly and significantly decreased the ABA response of plantlets to PEG treatment. Moreover, inoculation with either fungus regulated the ABA response of the plantlets even when the fungus was separated from the host by a cellophane sheet that prevented mycorrhiza formation. Although the wheat Em promoter was inducible in larch plantlets, it was not regulated by endogenous ABA. Induction of the wheat Em promoter in larch plantlets depended on organ type, with maximum induction in the root apex. Induction of the Em promoter was significantly decreased by mycorrhizal inoculation.  相似文献

Environmental problems many times could evolve when manure-containing pathogens are distributed into an open environment with no effort made to reduce the content of pathogens or limit their movement in the environment. Wind, surface flow, and subsurface flow can all carry enough pathogens to receiving waters to exceed water quality standards. This study was conducted to assess the microbiological quality of water associated with animal-based agriculture in the sub-basin of Pinhal River located in the rural area of Concordia, Santa Catarina, Brazil. Six sampling points representing different agricultural land uses (LU1—dairy cattle; LU2—without animals; LU3—dairy + pigs + poultry + crops; LU4—pigs + poultry + crops; LU5—dairy + pigs + poultry + crops + human; and LU6—dairy+pigs+crops) along the Pinhal River sub-basin (north to south) were sampled biweekly from August 2006 to December 2008. Concentrations of fecal coliforms and Escherichia coli varied significantly (p?≤?0.05) with land use (LU), but there was no interaction effect of LU, season, and time. Water samples from the catchment area of LU1 had the highest concentration of fecal coliforms (4,479?±?597 CFU ml?1) when compared with other catchment areas. Catchment area associated with LU2 (no animal) had the lowest concentrations of fecal coliforms (39.2?±?5.2 CFU ml?1). With the exception of LU2 (control site), all the maximum concentrations of E. coli exceeded the single maximum allowable concentration for E. coli (100 CFU ml?1). When LU1 was compared with other catchment areas (LU3, 50%; LU4, 67%; LU5, 58%; and LU6, 44%), it had the lowest counts (39%) of Salmonella sp. Our results suggest that spatial pattern of bacterial water quality is evident, which can be linked to the different land uses and associated practices (present or absent of animal activities). Therefore, varying responses associated with the different land uses would be critical in identifying the importance of different sources of bacteria in the catchment area and the mechanisms transferring them.  相似文献

Low-temperature fermentations (13 degrees C) are considered to improve wine aromatic profiles. However, because the risk of stuck and sluggish fermentations is high, these fermentations are not common. The aim of this paper was to analyze the effect of different preadaptation protocols in two commercial wine strains on the fermentation and some wine parameters. Preadaptation is understood to be the process between the rehydration of active dry yeast and the inoculation. In this study, it consisted of preparing a fermentation starter (addition of yeast grown at 25 degrees C) or inocula preadapted at low temperatures (as before, but grown at a fermentation temperature of 13 or 17 degrees C). These results were compared with those of rehydrated active dry yeast, and a commercial "cryotolerant" yeast was used as a reference. General fermentation kinetic parameters, yeast imposition, nitrogen consumption, and main wine products were analyzed. The results showed that the preadaptation of a yeast could improve the fermentation performance, although this improvement was strain-dependent. Low-temperature fermentations also had some general effects: reduction of acetic acid and fusel alcohol production and increased concentrations of glycerol. When the yeast performed better in fermentation because of preadaptation, nitrogen consumption was faster and the wine's "negative" attributes (acetic acid, fusel alcohols) were significantly reduced. Thus, in some strains, preadaptation could be an effective mechanism for improving low-temperature fermentation, which also significantly reduces detrimental wine attributes.  相似文献

This paper describes a modified hematoxylin staining technique that can be used to locate sites of A1 binding in freshwater plants and animals. This technique is fast, simple, and inexpensive to use. It is more reliable for organisms raised under controlled conditions, although it can be used on organisms isolated from the field. In the presence of Al, a purple stain appears which absorbs between 560 and 570 nm. This stain can be used as a live stain over the pH range of 3 to 9. The stain remains stable for at least 12 mo in alcohol-preserved specimens. Low concentrations of Fe (reddish-brown) and high concentrations of Pb (grey) and Cu (pink) can mask the reaction with Al (purple). Fluoride (10:1 molar ratio to Al), EDTA (1:1 ratio to Al), humic acid and PO4 (1:10 ratio to Al) can prevent A1 uptake or interfere with Al-hematein binding. Based on the technique described in this paper, sites of Al binding in aquatic plants and animals include: nucleus and cell wall of the green alga Mougeotia; cell wall of the aquatic moss Leptodictyum riparium; chloride cells, tip of the penis, and hind gut of the fairy shrimp, Branchinecta paludosa; and the anal papillae of the phantom midge, Chaoborus.  相似文献

The kinematics of the outer parts of three intermediate-luminosity elliptical galaxies were studied with the Planetary Nebula Spectrograph. The galaxies' velocity-dispersion profiles were found to decline with the radius, and dynamical modeling of the data indicates the presence of little if any dark matter in these galaxies' halos. This unexpected result conflicts with findings in other galaxy types and poses a challenge to current galaxy formation theories.  相似文献

The wild potato germplasm collection of the series Acaulia, maintained at the Centre for Genetic Resources, The Netherlands, consists of 314 accessions. In previous investigations, 15 potential duplication groups with a total of 36 accessions were identified based on AFLP analysis of a limited sample per accession. In the present study, the potential redundancies, plus one additional accession, were studied with increased sample sizes to examine intra- and inter-accession variation more accurately, with the aim to reduce the size of the collection. No variation was observed within two potential duplication groups, whereas only limited differentiation among accessions was detected within seven groups, resulting in a total of 15 redundant accessions (nearly 5% of the collection). A cluster analysis of all the accessions of the collection showed that these nine groups each had a distinct identity. It was decided to maintain the accessions of the remaining six groups as separate entries based on the large differentiation observed among accessions and the absence of a clear identity. An analysis of molecular variance in the set of 37 accessions showed that 91% of the observed variation could be found among accessions. This variance component appeared unaffected when the set was analysed without the 15 redundant accessions. The invested costs to identify redundancies in the series Acaulia by AFLP analysis are estimated at k 57.3, whereas the savings achieved by reduction of the collection are estimated at k 21.0 per generation. However, a cost-benefit analysis should not only focus on the short-term return of investments, but should also consider the value of newly obtained data and information. These include taxonomic information about accessions, optimised sampling strategies, optimised regeneration procedures, additional data for core collection formation and more efficient utilisation of germplasm.  相似文献

The purpose of the present study was to assess the rate of recovery of acidified lakes located near the town of Coniston following an abrupt reduction in atmospheric SO2 and metal emissions at the Coniston smelter which closed in 1972. The water chemistry of several lakes was studied over a period of 16 yr (1968–1984). In one extremely acidic lake close to the smelter, the pH increased from 4.05 in 1972 to 5.8 in 1984. Conductivity, as well as concentrations of SO4, Cu, Ni, Co, Mn, and Zn decreased by 60% to 90% in the lake water during the same period. In another initially less acidic lake nearby, the increase in pH was less dramatic, while the decrease in conductivity, SO4, and some metals was similar to that of the more acidic lake. Local SO4 deposition decreased approximately 75 % while Cu and Ni deposition decreased by 90% following closure of the Coniston smelter. These results indicate that even severely acidified lakes can improve within a few years following a substantial reduction in atmospheric S emissions; and that in some regions recovery can occur due to reductions in SO2 emissions even in the absence of concurrent NOx control.  相似文献

Heavy metals such as cadmium (Cd) enter the ecosystem chiefly as the result of human activities. Nowadays the heavy‐metal pollution of the soil is causing ever greater problems, exacerbated by the fact that the heavy metals accumulated in plants may, either directly or indirectly, find their way into animals and human beings. Maize is one of the world's most important crops, ranking third after wheat and rice, so the changes induced by one of the most toxic heavy metals, Cd, in maize plants is a cause of some concern. This review discusses not only the toxic symptoms caused by Cd stress, but also the tolerance mechanisms activated in the plants. Cadmium induces a number of physiological changes, such as growth inhibition, changes in the water and ion metabolism, the inhibition of photosynthesis, changes in enzyme activities, and the formation of free radicals. The synthesis and compartmentalization of phytochelatins is induced shortly after the initiation of Cd stress, while other defense mechanisms also play an important role.  相似文献

The sulphur hexafluoride (SF(6)) gas tracer method was used to measure methane (CH(4)) production of crossbred (3/4 Holstein x Zebu) dairy heifers fed two types of sugarcane (Saccharum officinarum L.; cultivar IAC-862480 (CC1) or cultivar IAC-873184 (CC2)) and supplemented with urea or concentrate. The study was performed at Embrapa Southeast Cattle, S?o Carlos, SP, Brazil, using a completely randomised design. Differences between treatments were significant for digestibility of dry matter, organic matter and energy. When animals were supplemented with urea differences between sugarcane cultivars did occur for NDF consumption, but not for daily methane production. This suggest that variation in chemical composition of sugarcane did not affect bovine ruminal CH(4) emissions. Concentrate inclusion in animal diet increased digestible organic matter intake, improving the nutrient intake by animals, but did not reduce CH(4) production expressed as a percentage of gross energy intake.  相似文献
