The role of ribulose 1,5-bisphosphate carboxylase-oxygenase (Rubisco) in the regulation of photosynthesis in the field was assessed by following changes in the amount and activity of Rubisco and in the amounts of chlorophyll and total nitrogen in a willow (Salix cv. Aquatica gigantea) canopy during two growing seasons. An additional aim of this study was to determine how nitrogen is partitioned in the canopy and whether Rubisco and chlorophyll serve as sources of nitrogen. During both growing seasons, the total activity and the amount of Rubisco were high in young, sun-exposed leaves and decreased as the leaves aged and became shaded. In 1985, the specific activity was low (on average 0.8 micromol mm(-1) mg(-1) Rubisco protein) compared with the values obtained in 1986 (on average 1.8 micromol min(-1) mg(-1) Rubisco protein). These differences in the specific activity of Rubisco between the two years might be associated with changes in partitioning of nitrogen. During the cool summer of 1985, about 20% of the total nitrogen was partitioned to Rubisco compared with about 13% during the warm summer of 1986. Rubisco comprised a high proportion (about 60%) of the total soluble protein throughout the 1985 growing season, whereas during the 1986 growing season, the proportion of Rubisco in the total soluble protein fraction was lower and decreased markedly as the leaves became shaded. Chlorophyll did not serve as a source of nitrogen in the canopy. On the contrary, the proportion of total nitrogen in chlorophyll increased with time, which indicates acclimation of the thylakoid membranes to decreasing irradiance. The degree of activation of Rubisco in samples collected in 1986 was high in young, sun-exposed leaves, which suggests that in these leaves, Rubisco may limit the rate of CO(2) uptake. In the shaded leaves of the lower canopy, the degree of activation of Rubisco was low, which indicates that in these leaves, photosynthesis is limited by factors other than Rubisco.  相似文献

Context

‘Conserving Nature’s stage’ has been advanced as an important conservation principle because of known links between biodiversity and abiotic environmental diversity, especially in sensitive high-latitude environments and at the landscape scale. However these links have not been examined across gradients of human impact on the landscape.

Objectives

To (1) analyze the relationships between land-use intensity and both landscape-scale biodiversity and geodiversity, and (2) assess the contributions of geodiversity, climate and spatial variables to explaining vascular plant species richness in landscapes of low, moderate and high human impact.

Methods

We used generalized additive models (GAMs) to analyze relationships between land-use intensity and both geodiversity (geological, geomorphological and hydrological richness) and plant species richness in 6191 1-km2 grid squares across Finland. We used linear regression-based variation partitioning (VP) to assess contributions of climate, geodiversity and spatial variable groups to accounting for spatial variation in species richness.

Results

In GAMs, geodiversity correlated negatively, and plant species richness positively, with land-use intensity. Both relationships were non-linear. In VP, geodiversity best accounted for species richness in areas of moderate to high human impact. These overall contributions were mainly due to variation explained jointly with climate, which dominated the models. Independent geodiversity contributions were highest in pristine environments, but low throughout.

Conclusions

Human action increases biodiversity but may reduce geodiversity, at landscape scale in high-latitude environments. Better understanding of the connections between biodiversity and abiotic environment along changing land-use gradients is essential in developing sustainable measures to conserve biodiversity under global change.

Summary

Timing of harvest is an essential factor affecting the quantity, quality and storability of onion yield. This study aimed to establish when no further yield increase is to be expected and to determine the relationship between maturity stage and yield development. Experiments on 4-6 harvests at 9-14 d intervals were conducted at several locations in southern Finland in 1996-1999. Generally little, if any, yield increase was recorded after plants had reached 100% maturity, but in some cases bulb growth continued after complete fall-down of leaves. On the other hand, harvesting before 100% maturity resulted in a yield loss of 0-45% of final yield. Weight loss, and thus energy consumption during drying, was still reduced after complete leaf fall-down. Therefore, it may be concluded that delaying harvest up to 100% maturity, or even longer, ensures that highest yield and lowest drying costs.  相似文献

Ultraviolet resonance Raman spectroscopy (UVRRS) was used to study the formation of aromatic and/or lignin-carbohydrate (LC) structures at the reducing end groups of O-alkyl-substituted cellulose under conditions simulating the initial phase of kraft pulping. The derivatives studied were methyl cellulose (MC) with degree of substitution (DS) of 1.64–1.95, carboxymethyl cellulose (CMC) with DS ∼0.6, and a lignin model compound, creosol. The total alkali concentrations in the treatments were 0.1 M and 0.5 M and the sulfidities were 1%, 10%, and 30%. HS ions and creosol are both strong nucleophiles and they compete for the hot-alkali-generated unsaturated electrophilic reaction sites in the reducing end groups of the polysaccharides. The results indicated that conditions similar to those in the initial phase of conventional kraft cooking (high OH and low HS ion concentrations) increased the aromatic nature of the end groups and conditions similar to those in the initial phase of super batch cooking (low OH and high HS ion concentrations) partly inhibited the formation of aromatic and LC end groups.  相似文献

Scots pine battens were heat-treated at 100–240º C under saturated steam. Cross-sections of heat-treated battens were analysed using ATR and reflection FTIR microscopies. A typical absorption band of fats and waxes at 1740 cm-1 was detected on the sapwood edges in the temperature range of 100–160º C, indicating that fats and waxes moved along the axial parenchyma cells to the surface of the sapwood during the heat treatment. At the elevated temperatures (above 180° C) fats and waxes disappeared from the sapwood surface and were no longer detected with FTIR spectroscopy. Resin acids were detected at temperatures between 100 and 180º C in the middle of the battens. IR spectra of these spots showed a characteristic absorption band of resin acids at 1697 cm-1. At 200º C resin acids were not detected in the middle of the battens; however, resin acids were detected at distances of 500 and 600 mm from the midpoint of the battens and on the edges of battens. At temperatures above 200° C, resin acids had disappeared from the wood.  相似文献

An elemental chlorine-free (ECF)-bleached soft-wood kraft pulp was treated first with a carboxymethyl cellulose (CMC, degree of substitution 0.3, 1% on pulp) and then with alkyltrimethylammonium bromides (alkyl chain lengths of C10 to C16). Surprisingly, the treatment with dodecyltrimethylammonium bromide (C12TAB) markedly increased the internal and tensile strengths of the handsheets prepared from the modified pulp. Environmental scanning electron microscopy (ESEM) showed that these properties could be partly explained by the “rope/gum-like” bridges that were formed between the fibers.  相似文献

OBJECTIVE: To study gelatinolytic matrix metalloproteinases (MMPs) in tracheobronchial lavage fluid (TBLF) obtained from clinically normal calves and calves with Pasteurella multocida infection. SAMPLE POPULATION: Samples of TBLF obtained from 11 calves with clinical signs of respiratory tract disease and growth of P multocida and Mycoplasma spp during culture of TBLF and samples of TBLF from 6 clinically normal calves with no bacterial growth during culture of TBLF. PROCEDURE: MMPs in TBLF were analyzed by use of gelatin zymography. Gelatinases were identified on the basis of molecular weights and inhibition by EDTA. RESULTS: The main gelatinolytic MMPs detected were the proform (90 to 110 kd) and active form (75 to 85 kd) of MMP-9 (gelatinase B) and the proform (67 to 75 kd) and active form (< 65 kd) of MMP-2 (gelatinase A). Increased amounts of active MMP-2 and MMP-9 were detected in TBLF of calves with respiratory tract disease, compared with amounts of active MMP-2 and MMP-9 in TBLF of clinically normal calves. Concurrent infection with Mycoplasma bovirhinis in calves with pneumonia attributable to P multocida was associated with higher concentrations of MMP-9. CONCLUSIONS AND CLINICAL RELEVANCE: The host response to P multocida includes increases in MMP-2 and MMP-9 concentrations in TBLF. Greater amounts of MMPs detected in calves with concurrent M bovirhinis and P multocida infection indicates synergism between these organisms.  相似文献

Increased recycling of organic wastes has raised concern about the quality of compost end products. In addition to the limit values for heavy metals and impurities including weeds and pathogens, the quality criteria for compost products should also include criteria for maturity. There is a tremendous number of maturity assays, developed earlier by several authors, and recommended to be used to evaluate maturity of composts. Because no such single test alone reliably demonstrates the complex properties occurring during maturization of compost, we developed a fast and easy-to-use two-phase test scheme for the assessment of maturity. In the first phase the degradation phase e.g. stability of compost samples is evaluated by using a carbon dioxide evolution test and/or determination of the NO3-N/NH4-N ratio by simple test strips. In the second phase, the toxicity of the compost is evaluated by a plant growth test, germination tests and/or the Flash bioluminescence test. Eleven plants composting sewage sludge, source-separated biowaste, manure or a combination of these raw materials were sampled after 1-3 weeks of composting and when the compost was considered “ready for use”. Chemical and physical analyses were considered useful as additional information when evaluating maturity especially when the results were not conclusively clear. This fast and easy-to-use test scheme was designed especially for the composting plant operators and official laboratories responsible for evaluating compost quality.  相似文献
