柑橘溃疡病(Xanthomonas citri subsp. citri,Xcc)是严重阻碍柑橘产业发展的世界性检疫病害,防控困难,选育抗病品种为防治溃疡病的唯一有效办法。枸橼C-05是柑橘属独特的抗溃疡病种质,为抗性机理和抗病育种提供了种质基础。为了进一步评价枸橼C-05的抗性,另外收集了13份枸橼种质,对其进行溃疡病的离体和活体抗性评价。叶片形态学观察表明,14份枸橼种质存在较大的形态学差异。采用离体与活体注射接种不同浓度的柑橘溃疡病菌,根据叶片的感病症状、病斑周围黄色晕圈大小等典型溃疡病症状,综合评价枸橼类种质的抗病性。结果表明:枸橼种质之间对溃疡病的抗性存在极大差异,溃疡病的抗性评价综合病情指数显示枸橼C-05、园香橼、矮果香橼、美国枸橼、普通枸橼和印度大果综合病情指数较小,表现为极抗病或抗病,其他枸橼种质表现不同程度的感病性;枸橼种质的抗病或感病性与其叶片形态特征无关。枸橼C-05和5个抗病枸橼种质对柑橘溃疡病具有针对性的抗性,这对进一步研究抗病机理和抗病育种具有重要意义。 相似文献

Wheat aphid, Sitobion avenae (fabricius), is one of the most important wheat pests and has been reported to be resistant to commonly used insecticides in China. To determine the resistance mechanism, the resistant and susceptible strains were developed in laboratory and comparably studied. A bioassay revealed that the resistant strain showed high resistance to pirimicarb (RR: 161.8), moderate reistance to omethoate (32.5) and monocrotophos (33.5), and low resistance to deltamethrin (6.3) and thiodicarb (5.5). A biochemistry analysis showed that both strains had similar glutathione-S-transferase (GST) activity, but the resistant strain had 3.8-fold higher esterase activity, and its AChE was insensitive to this treatment. The I50 increased by 25.8-, 10.7-, and 10.4-folds for pirimicarb, omethoate, and monocrotophos, respectively, demonstrating that GST had not been involved in the resistance of S. avenae. The enhanced esterase contributed to low level resistance to all the insecticides tested, whereas higher resistance to pirimicarb, omethoate, and monocrotophos mainly depended on AChE insensitivity. However, the AChE of the resistant strain was still sensitive to thiodicarb (1.7-fold). Thus, thiodicarb could be used as substitute for control of the resistant S. avenae in this case. Furthermore, the two different AChE genes cloned from different resistant and susceptible individuals were also compared. Two mutations, L436(336)S in Sa.Ace1 and W516(435)R in Sa.Ace2, were found consistently associated with the insensitivity of AChE. 旨在分析猪丁型冠状病毒(porcine deltacoronavirus,PDCoV)在悬浮培养的猪肾细胞LLC-PK1上的增殖特性,为PDCoV灭活疫苗的规模化生产提供细胞材料。采用逐步降血清法优化LLC-PK1细胞悬浮培养工艺;利用有限稀释法筛选PDCoV适应性细胞株;利用间接免疫荧光法鉴定PDCoV对LLC-PK1细胞的感染性;分别对PDCoV接种LLC-PK1悬浮细胞的初始密度、MOI、收毒时间、TPCK胰酶浓度等参数进行优化,确定最佳悬浮培养条件。成功筛选出可高效增殖PDCoV的单克隆悬浮细胞株LLC-PK1Sa,且利用其增殖的PDCoV可特异性的感染LLC-PK1细胞;PDCoV按MOI为10-3接种于密度为2×106 cells·mL-1的LLC-PK1Sa细胞,当TPCK胰酶终浓度达到7.5 μg·mL-1时,接毒后48 h收获的病毒液滴度最高。本研究首次实现了PDCoV在LLC-PK1Sa悬浮细胞中的高效增殖,并对悬浮培养条件进行了初步优化,可为PDCoV灭活疫苗的规模化生产提供理论参考。 相似文献

Context
Although various tracking methods have been used in many ecosystems to investigate seed dispersal and seedling recruitment, it is still difficult to measure seed dispersal patterns due to methodological challenges in tracking seed movement away from parent trees and in finding the locations of seedlings.Aim
Here, we aimed to develop a new approach to track seed dispersal by animals in the field.Methods
Our approach involves soaking seeds directly into 15N-urea solutions with different dosages to enrich them isotopically. This new method is expected to create a reliable differentiation between the enriched seeds and natural ones and consequently between the corresponding seedlings.Results
We showed that acorns of Quercus variabilis and Cyclobalanopsis glauca and seeds of Pinus koraiensis soaked in 15N-urea solutions were successfully enriched. We did not find that δ 15N value of seeds was a linear function of 15N-urea soaking duration. However, with high urea dosage and extending soaking duration, the δ 15N values in seedlings of Q. variabilis and P. koraiensis were higher than those of normal plants. As expected, 15N isotope was significantly diluted in the growing seedlings germinated from 15N-enriched seeds. Using the 15N-urea soaking method, we successfully located five seedlings of Q. variabilis germinated from the enriched acorns in the field.Discussion
The 15N-urea soaking technique is powerful in tracking seed dispersal and seedling recruitment. This new method solves some of the problems inherent in traditional methods for tracking secondary seed dispersal and could further improve the study of seed dispersal ecology. 相似文献8.

对两种一年生禾草喷施增产菌的效果进行了研究。结果表明,叶面喷雾可促进燕麦的生殖生长,籽实提前成熟2 ̄5d,籽实产量比对照提高21.7%,纯增收325.50元,产投比为6.1:1.0,高粱鲜草产量比对照提高29.0%,纯增收586.50元,产投比为10.1:1.0,拌种+叶面喷雾对燕麦分蘖有促进作用,可明显提高鲜草产量,纯增收最高可达253.50元,产投比最高可达4.3:1.0。 相似文献

以安徽省2000—2005年的中籼稻区域试验和联合鉴定试验的424个品种(组合)为材料,对产量性状作了相关分析及主成份分析。分析结果表明:产量和每穗总粒数、每穗实粒数、结实率、千粒重均呈极显著正相关,与有效穗呈显著正相关,每穗总粒数、每穗实粒数、结实率与有效穗呈极显著负相关,千粒重与有效穗呈负相关。所以在中籼品种(组合)的高产育种中,应以选每穗总粒数、每穗实粒数为主攻目标,即要选育穗大粒多型品种,不必过于追求有效穗和千粒重。穗实粒数、产量与全生育期存在一定的正相关关系,但不显著,即生育期短的品种(组合)也可以实现高产。主成份分析也表明穗实粒数、穗总粒数是高产育种主攻的关键。并在分析基础上提出了中籼稻品种(组合)高产育种策略建议,为中籼新品种(组合)的选育提供参考。 相似文献
