【目的】建立新疆冷水鱼中孔雀石绿(MG)和隐色孔雀石绿(LMG)残留量的超高效液相色谱-串联质谱(UPLC-MS/MS)更为简便、高效的检测方法。【方法】样品经1%冰乙酸-乙腈溶液提取,氨基及SCX(强阳离子交换)固相小柱净化,C18 色谱柱分离,电喷雾串联四极杆质谱的多反应监测扫描模式(MRM)进行检测,并采用内标法定量。【结果】MG和LMG在5.0~100.0 μg/L线性范围内具有良好的线性关系,相关系数分别为:0.999 7(MG)和0.999 2 (LMG)。测定低限(LOQ)为2.0 μg/kg。以空白样品为基体做加标回收,测得MG的回收率在88%~105%,LMG的回收率在85%~110%,相对标准偏差(n=6)均为2.2%~15.3%。【结论】该方法样品前处理简便,回收率高,稳定性好。适用于新疆冷水鱼中MG和LMG残留量的日常检测。  相似文献

The current study investigated dietary effects of soy genistein (SG) and citrus hesperidin (CH) on microbial spoilage and antioxidant quality of chicken meat. Broilers were divided to six treatment groups: control (C), SG and CH, supplemented with 5 mg of SG and 20 mg of CH per kg of feed, respectively, and SGCH1, SGCH2 and SGCH3, supplemented with 5, 10 or 20 mg of SG + CH (1:4) per kg of feed respectively. Both SG and CH reduced (< .01) the microbial load of spoilage organisms in breast meat at 0 and 15 days of refrigeration. Hepatic and muscle total antioxidant and superoxide dismutase activities, and crude protein contents were generally improved (< .05), whereas hepatic malondialdehyde concentration and muscle fat was decreased (< .01) by the dietary treatments. In conclusion, soy genistein and CH supplementation could improve meat composition, antioxidant and microbial quality of broilers meat.  相似文献

ABSTRACT

1. The genetic diversity and population structure were studied for eight local chicken breeds, including Anjiyan (AN), Hetian Black (HH), Hetian Ma (HM), Aheqi (AH), Baicheng You (BC), Hejing (HJ), Tashkurghan (TS) and Ruoqiang (RQ), in the Southern Xinjiang region of China, using 20 microsatellite markers.

2. Total 336 alleles were obtained from all chicken breeds, with a mean of 16.8 alleles per locus. The polymorphism information content ranged from 0.444 to 0.911, with a mean of 0.729 and almost all of the loci showed significant deviation from Hardy-Weinberg standards. The observed and expected heterozygosity of the eight breeds ranged from 0.5 to 0.677 and from 0.656 to 0.774, with the lowest observed in the AN and the highest in BC breed. The average breed genetic diversity was 0.655 for AN and 0.766 for BC chickens.

3. According to the neighbour-joining (NJ) method, three main clusters were identified in the NJ phylogenetic tree with AN and RQ breeds in one clade, HH and HM breeds in the second clade and TS, HJ, AH and BC breeds in the third clade.

4. Based on STRUCTURE analysis, the most likely cluster number of all breeds was K = 4, whereby HH and HM breeds formed one cluster and AH, BC, HJ and TS formed another, and RQ, AN chicken formed their own distinct cluster. These results indicated that HH and HM breeds had similar genetic background, as did the breeds of AH, BC, HJ and TS. RQ, AN breed had unique genetic backgrounds, distinct from the other breeds. Genetic introgression was detected from AN to HH and HM.

5. The results of the current study can be used as baseline genetic information to implement effective conservation programs and to make better use of these local chicken breeds, especially for the AN, RQ and TS breeds.  相似文献

目的】采用分子生物学方法对新疆感官异常的番茄酱和杏酱中细菌进行快速鉴定。【方法】采用梯度稀释法在需氧和厌氧条件下,对感官异常的番茄酱和杏酱中的腐败细菌进行培养,通过16S rDNA序列分析并结合菌株系统发育树对分离菌株进行鉴定。【结果】分离共获得9株细菌菌株,其中番茄酱中的5株菌分别为:甲基营养性芽孢灰枣杆菌、枯草芽孢灰枣杆菌、地衣芽孢灰枣杆菌、沃氏葡萄球菌、粪肠球菌;杏酱中的4株菌株分别为巴氏葡萄球菌、粪肠球菌、沃氏葡萄球菌、类芽孢灰枣杆菌,鉴定结果与生化方法一致。【结论】该方法准确快速,可为番茄酱、杏酱生产企业有效防控产品污染保障质量安全提供基础数据。  相似文献

试验采用二因子随机区组设计,研究Ovocrack对产蛋鸡生产性能及蛋品质的影响。结果表明,在本试验条件下,不同营养水平日粮添加不同水平的Ovocrack射产蛋鸡生产性能未产生显甚影响;但添加Ovocrack的各处理蛋黄颜色和褐壳蛋率比未添加Ovocrack的各处理分别提高了1.4~2.0个罗氏比色单位和12%~15%,而且提高的幅度随其添加量的增加而显著上升。综合各项指标,建议产蛋鸡常规营养水平日粮中添加500g/t Ovocrack较合适。  相似文献
