For this paper, the plasma membrane (PM) H -ATPase gene has been cloned from Populus euphratica Oliv. through a homology based strategy. The isolated 3,210 bp cDNA contains a single 2,862 bp open reading frame (ORF) which encodes a putative H -ATPase protein of 953 amino acid residues, with a significant homology to plasma membrane H -ATPase of Primus persica, Phaseolus vulgaris, Sesbania rostrata and Daucus carota. The predicted protein has a molecular weight of 104,553 Da. The copy number analysis revealed multiple copies of the PM H -ATPase in the P. euphratica genome after digestion of their genomic DNA by the restriction enzymes EcoRI, NdeⅠ, FbaⅠand BglⅡ, and Southern blot. 相似文献

The electrochromism of polyaniline films, which is prepared by electropolymer-ization from aniline in acidic aqueous solution onto platinum,is investigated in 330-700nm through in-situ ellipsometric technique. The results indicate that the ellipsometric peak of the reduced state(-0. 2V) of the film appears at 390-400nm and the oxidized state(0. 55V) at 500-530nm. The elec-trochromic reaction from the reduced "yellow" state to the oxidized "green" state is suggested via intermediate state whose ellipsometric peak appears at 466-470nm. 相似文献

The aims of this study were to evaluate the effects of a ketamine/propofol anaesthetic protocol in lions (Panthera leo), and to compare it to two commonly used anaesthetic protocols. Seventeen adult lions were anaesthetised using three different protocols. Group XK (n=6) was anaesthetised with intramuscular (i.m.) injections of xylazine and ketamine. Group KD (n=5) was anaesthetised with an i.m. injection of ketamine, followed by an intravenous (i.v.) injection of ketamine and diazepam. Group KP (n=6) was anaesthetised with an i.m. injection of ketamine followed by an i.v. injection of propofol. There was a significant difference in heart rate (P<0.0002), which was lowest in group XK and highest in KD. Jaw tone was significantly lower in Group XK (P<0.05). No undesirable effects were noted following injection of the propofol. Propofol was a suitable and safe drug for maintenance of anaesthesia in adult lions. 相似文献

Following challenge with Trichosirongylus colubrifonizis, increased numbers of T-cells and immunoglobulin responses are seen in the intestine of sheep immunised by repeated infection with live worms. IL-6 mRNA expression in the small intestine from T. colubriformis-immunised and naive sheep was determined by in situ hybridisation, whereas CD4(+), IgA(+), IgG(+) cells in the gut were evaluated by immunohistochemistry. There was constitutive expression of IL-6 mRNA by cells in the naive gut, and the number of these cells was increased by parasite challenge. There were corresponding increases in numbers of CD4(+) and TCR gamma/delta(+) T-cells and IgG(+) B-cells. Our data are consistent with a role for IL-6, perhaps produced by CD4(+) and/or TCR gamma/delta(+) T-cells or B-cells, in B-cell terminal differentiation. Infiltration of B-cells, particularly IgG(+) B-cells, may reflect parasite immunity in the host. 相似文献

Anthocyanidins found in certain flowers have been shown to act as strong antioxidants in various systems, exhibiting multiple biological actions. The antioxidative effects of water extract and ethanolic extract of longan (Dimocarpus longan Lour.) flowers were evaluated by radical scavenging activity and compared to those of gallic acid, myricetin, and epigallocatechin gallate. In this study, the suppressive effects of longan flower extracts on nitric oxide and prostaglandin E2 production were investigated using a lipopolysaccharide-stimulated RAW 264.7 cell model. Abundant levels of phenolic compounds including flavonoids, condensed tannins, and proanthocyanidins were found in water or ethanolic extracts prepared from dried longan flowers. The antioxidative effect of longan flower extract was similar to the effect exhibited by pure antioxidants. Moreover, longan flower extract showed prominent inhibitory effects on prostaglandin E2 production. Significant concentration-dependent inhibition of nitric oxide production was detected when cells were cotreated with lipopolysaccharide and various concentrations of longan flower extracts. These inhibitory effects were further attributed to suppression of inducible nitric oxide synthase protein expression and not to reduced enzymatic activity. These results suggest that longan flower crude extracts, especially ethanolic extract, have antioxidant and anti-inflammatory effects, and the probable mechanism involves inhibition of inflammation by proanthocyanidins. Preliminary observations suggest that longan flower extract, especially alcoholic extract, could be another potential source of natural dietary antioxidant and anti-inflammatory agent. 相似文献

Using double-stranded RNA(dsRNA) technology and sequence-independent amplification(SIA), the molecular identification on infected Rehmannia glutinosa in the field with mosaic symptoms was performed and the whole-genome of the Rehmannia mosaic virus(ReMV) Shanxi isolate(ReMV-SX) was sequenced. Sequencing analysis showed that the virus that infected Rehmannia glutinosa was Rehmannia mosaic virus(ReMV). The full-length of the obtained ReMV-SX sequence(GenBank accession no. JX575184) was 6 395 nt, containing four open reading frames(ORFs). The sequence homology analysis of the complete nucleotide sequence showed that ReMV-SX was 93.8%-97.0% homologous to ReMV in Tobamovirus subgroup I, while only 49.8%-58.9% homologous to the isolates in subgroups II and III of the same genus. Phylogenetic analysis showed that ReMV-SX and ReMV-Henan formed a separate branch and had the closest genetic relationship. The results laid the foundation for ongoing researches in the taxonomic status and evolution of ReMV and for further investigating the pathogenic mechanism of ReMV infecting Rehmannia glutinosa. 相似文献
