Arbuscular mycorrhizal fungi (AMF) are key components of ecosystems through their influence on plant communities and ecosystem processes. A major source of information regarding the importance of AMF species richness on process rates are mesocosm experiments using different levels of diversity of AMF as provided by single-species cultures of AMF. Since AMF inocula are generally made available in the form of non-sterile pot culture material, it is possible that AMF symbiosis-associated microbiota are at least partially responsible for some effects hitherto directly attributed to the AMF mycelium. Here, we provide evidence that microbiota associated with single-species cultures of AMF (after long-term pot culture enrichment of 7–8 years) can strongly affect the ecosystem process of soil aggregation. This effect occurred in an AMF isolate specific manner, but in the absence of live and active AMF mycelium. We additionally documented large differences in microbiota communities associated with the different AMF inocula (using PLFA analyses), suggesting that these differences were at least partly responsible for the observed changes in soil aggregation. This result points to AMF–microbiota interactions as a largely unexplored mechanism underlying soil aggregation (and potentially other ecosystem processes). We suggest that a reinterpretation of previous experiments using greenhouse-derived AMF cultures may be necessary, and the need to consider AMF symbiosis-associated microbiota in mechanistic studies of AMF and mycorrhizae in general is emphasized.  相似文献

The effects on bruising and muscle pH of mixing cows and steers at loading, and 96 hours before loading, for transport to slaughter were examined. A second experiment investigated the effect on bruising of mixing two strange groups of steers at loading, 24 and 96 hours before loading. All cattle were hornless Bos indicus crossbreds from the same property in north Queensland. The bruise score, bruise trim weight and muscle pH of mixed cows and steers did not differ significantly from that of those transported separately. Mixing of two strange groups of steers did not significantly affect bruising. Time of mixing had no noteworthy effect on bruising. Cows bruised significantly (P less than 0.01) more than steers. These results have important implications for low cost beef management.  相似文献

Fecal samples collected from cattle at processing during a 1-year period were tested for verotoxins (VT1, VT2), Escherichia coli O157:H7, and Salmonella. Verotoxins were detected in 42.6% (95% CI, 39.8% to 45.4%), E. coli O157:H7 in 7.5% (95% CI, 6.1% to 9.1%), and Salmonella in 0.08% (95% CI, 0.004% to 0.5%) of the fecal samples. In yearling cattle, the median within-lot prevalence (percentage of positive samples within a lot) was 40% (range, 0% to 100%) for verotoxins and 0% for E. coli O157:H7 (range, 0% to 100%) and Salmonella (range, 0% to 17%). One or more fecal samples were positive for verotoxins in 80.4% (95% CI, 72.8% to 86.4%) of the lots of yearling cattle, whereas E. coli O157:H7 were detected in 33.6% (95% CI, 26.0% to 42.0%) of the lots. In cull cows, the median within-lot prevalence was 50% (range, 0% to 100%) for verotoxins and 0% (range, 0% to 100%) for E. coli O157:H7 and Salmonella (range, 0% to 0%). Verotoxins were detected in one or more fecal samples from 78.0% (95% CI, 70.4% to 84.2%) of the lots of cull cows, whereas E. coli O157:H7 were detected in only 6.0% (95% CI, 3.0% to 11.4%) of the lots of cull cows. The prevalence of verotoxins in fecal samples was lower in yearling cattle than in cull cows, whereas the prevalence of E. coli O157:H7 in fecal samples was higher in yearling cattle than in cull cows. The prevalence of E. coli O157:H7 in fecal samples was highest in the summer months. Rumen fill, body condition score, sex, type of cattle (dairy, beef), and distance travelled to the plant were not associated with the fecal prevalence of verotoxins or E. coli O157:H7. The prevalence of verotoxins in fecal samples of cull cows was associated with the source of the cattle. It was highest in cows from the auction market (52%) and farm/ranch (47%) and lowest in cows from the feedlot (31%). In rumen samples, the prevalence of verotoxins was 6.4% (95% CI, 4.2% to 9.4%), and it was 0.8% (95% CI, 0.2% to 2.3%) for E. coli O157:H7, and 0.3% (95% CI, 0.007% to 1.5%) for Salmonella.  相似文献

Escherichia coli K12 strains TB1(pCG5), with the genes for Shiga-like toxin IIv from an edema disease isolate of E. coli and TB1(pCG5-1), with the toxin genes inactivated by transposon mutagenesis, were used to test the hypothesis that Shiga-like toxin IIv was the same as edema disease principle. Ammonium sulfate precipitated culture supernatants from the pair of E. coli K12 strains and from a wild edema disease isolate of E. coli (E145) were tested for their ability to induce signs and lesions of edema disease in intravenously inoculated weaned pigs. Similar preparations from E. coli which produce Shiga-like toxins I and II were also tested. Preparations from E. coli TB1 (pCG5) and E145 contained high levels of Shiga-like toxin IIv and induced signs and lesions similar to those seen in edema disease, whereas preparations from E. coli TB1 (pCG5-1) failed to induce signs or lesions of edema disease. All Shiga-like toxin preparations produced delayed neurological signs, fibrinoid necrosis of arterioles and hemorrhages in the cerebellum of pigs. High doses of Shiga-like toxin IIv were associated with superficial necrosis of the colonic epithelium and vasculitis. Shiga-like toxins I and II resulted in kidney lesions but no enteric pathology. Shiga-like toxin II preparations had the lowest median lethal dose for pigs, Shiga-like toxin IIv was intermediate and Shiga-like toxin I was the least toxic.  相似文献

Knowledge of how forest management influences soil microbial community interactions is necessary for complete understanding of forest ecology. In this study, soil microbial communities, vegetation characteristics and soil physical and chemical properties were examined across a rectangular 4.57 × 36.58 m sample grid spanning adjacent coniferous forest and clearcut areas. Based on analysis of soil extracted phospholipid fatty acids, total microbial biomass, fungi and Gram-negative bacteria were found to be significantly reduced in soil of the clearcut area relative to the forest. Concurrent with changes in microbial communities, soil macroaggregate stability was reduced in the clearcut area, while no significant differences in soil pH and organic matter content were found. Variography indicated that the range at which spatial autocorrelation between samples was evident (patch size) was greater for all microbial groups analyzed in the clearcut area. Overall, less spatial structure could be resolved in the forest. Variance decomposition using principal coordinates of neighbor matrices spatial variables indicated that soil aggregate stability and vegetation characteristics accounted for significant microbial community spatial variation in analyses that included the entire plot. When clearcut and forest areas were analyzed separately, different environmental variables (pH in the forest area and soil organic matter in the clearcut) were found to account for variation in soil microbial communities, but little of this variation could be ascribed to spatial interactions. Most microbial variation explained by different components of microbial communities occurred at spatial scales other than those analyzed. Fungi accounted for over 50% of the variation in bacteria of the forest area but less than 11% in the clearcut. Conversely, AMF accounted for significant variation in clearcut area, but not forest, bacteria. These results indicate broadly disparate controls on soil microbial community composition in the two systems. We present multiple lines of evidence pointing toward shifts in fungi functional groups as a salient mechanism responsible for qualitative, quantitative and spatial distribution differences in soil microbial communities.  相似文献

This study examines the results of bacterial culture from 159 endometrial biopsy samples from 97 commercial dairy cows and correlations between bacteriological and histological findings. Bacteria were isolated from approximately 80% of biopsies taken at day 26 and day 40 postpartum. Eleven percent of biopsies were positive for both aerobic and anaerobic culture. Streptococci, Escherichia coli and Actinomyces pyogenes were the most common isolates. Isolation of A. pyogenes from a biopsy at day 26 was positively correlated with isolation of anaerobic bacteria and segmented cell inflammation in the same biopsy, and with subsequent isolation of A. pyogenes at day 40. There was a strong association between isolation of A. pyogenes and anaerobes at day 26 with increased uterine lesions at day 40. Isolation of alpha hemolytic streptococci (AHS) was negatively correlated with isolation of A. pyogenes and with inflammation. Actinomyces pyogenes and AHS showed opposite associations with mononuclear cell inflammation and lymphocytic foci.  相似文献

It is estimated that between 20 400 and 23 850 × 106 1 of spoiled cutting fluids are discarded in the U.S. each year. Most undergo some form of biological waste treatment, yet there is little information regarding the effectiveness of such treatment on these fluids. In this study, sixteen commercially available synthetic and semisynthetic cutting fluid concentrates were tested for their susceptibility to biological degradation. The BOD5S of each compound was determined as an indication of biological oxidation potential. Inocula of both sewage and river origin were used, with statistically identical results. It was found that biological utilization ranged from 0 to 31% of the total oxygen demand, as measured by COD. Studies with triazine and morpholine preservatives showed no relationship between effectiveness of preservation and degradability.  相似文献
