Streptococcus suis is an important swine pathogen and a zoonotic agent. Differences in virulence have been noted among the 33 described serotypes, serotype 2 being considered the most virulent. In this study, we aimed at assessing the serotype distribution and the production of virulence-associated markers by strains recovered from diseased pigs in the United States (U.S.). Results showed that among the 100 strains evaluated, serotype 3 (20% of the isolates) and serotype 2 (17%) were the most prevalent. We then investigated the presence in these isolates of the genes sly, epf and mrp, encoding the virulence-associated markers suilysin (SLY), extracellular factor (EF) and muramidase-released (MRP) protein, respectively. The effective production of the markers by the strains was also verified. Results showed that the presence of the gene did not always correlate with actual expression of the respective protein. In the case of MRP, this was due, in most cases, to frameshift mutations at the 5′ end of the gene resulting in premature stop codons. The most prevalent phenotypes among U.S. strains were MRP+EFSLY (40%) and MRPEFSLY+ (35%). Serotype distribution greatly differed from that reported in several European countries, as did the production of virulence markers, particularly for serotype 2. On the other hand, our results for the U.S. S. suis isolates are similar to those reported for Canadian strains, suggesting a common status in North America.  相似文献

Farmers’ perceptions over time of an agroforestry technology can have an important impact on adoption and disadoption. Their perceptions, in turn, may be influenced by the type and scale of farm they own and the social networks they create. We examined the factors underlying producers’ perceptions of silvopasture systems at the time of adoption and perceptions following several years of experience, and the factors explaining discontinuance of systems in Argentina. We found that while most adopters indicated that other people influenced their decision about whether or not to adopt silvopasture, the type of person that influenced them (professionals vs. other farmers) did not affect adopters’ perceptions of the relative benefits and challenges of the system. However, farm scale and farm type did explain farmers’ perceptions to a good degree. Smaller-scale farmers were less likely to see costs and returns as benefits of the system, but more likely to see cash flow properties as important advantages. Farmers’ perceptions after experiencing the system were good predictors of likely discontinuance, but influential people, farm scales, farm type, and perceptions at the time of adoption were not.  相似文献

The development of simple predictors of sulfur (S) mineralization and its correlation with field-derived data may help improving corn S availability diagnosis. The objectives of this study were (1) to compare methods to estimate soil S mineralization, (2) to develop a model to predict soil S mineralization from S mineralization indexes and edaphic variables, and (3) to predict field-grown corn S uptake (Suptake) and apparent S mineralization (Smin-app) from different S mineralization indexes and edaphic-climatic variables. We evaluated 26 experimental sites where we measured edaphic variables as soil organic C (SOC), organic C in the particulate fraction (C-PF), S mineralization potential (Smin-10wk), S mineralized during a short-term (7 days) aerobic incubation + initial inorganic S (Smin-7d?+?Sinorg), and N mineralized during a short-term (7 days) anaerobic incubation (Nan). Additionally, 18 field experiments were carried out to quantify Suptake and Smin-app. The C-PF, Smin-7d?+?Sinorg, Nan, and SOC were variables significantly correlated with Smin-10wk (r?=?0.89, 0.89, 0.88, and 0.85, respectively). We developed a simple model to predict Smin-10wk from selected edaphic variables (Smin-10wk?=?0.038*Nan?+?0.106*SOC?+?0.74; Ra2 =?0.87). The Smin-10wk, C-PF, and Smin-7d?+?Sinorg showed a liner-plateau association with Suptake (R2?=?0.73, 0.53, and 0.48, respectively). We modified the method to estimate Smin-app to account for S losses (Smin-app (modified)) and developed a model to predict Smin-app (modified) from C-PF (Smin-app (modified)?=?4.65*C-PF?+?9.86; R2?=?0.62) or Smin-10wk (Smin-app (modified)?=?3.0*Smin-10wk?+?7.4; R2?=?0.54). Our results demonstrate that S mineralization indexes can be used to predict corn S availability under field conditions.  相似文献

Recent analyses of Streptococcus suis isolates using multilocus sequence typing (MLST) suggested the importance of sequence type (ST) 1 and ST27 complexes for animal hygiene and public health. In this study, to investigate whether pilus-associated genes in S. suis can be used as novel genetic markers for important clonal groups, we examined the correlation between STs and putative pilus-associated gene profiles in S. suis. Genomic searches using sequenced genomes and sequence data determined in several isolates revealed the presence of at least four distinct putative pilus gene clusters in S. suis (srtBCD, srtE, srtF, and srtG clusters). On the basis of the presence or absence of genes in the four clusters, 108 S. suis isolates from various origins were classified into 12 genotypes (genotypes A–L). Genotypes A and B, which possessed srtBCD plus srtF clusters and srtF plus srtG clusters, respectively, were the most common in isolates from diseased pigs and humans, and 29.9% and 59.8% of the isolates belonged to genotypes A and B, respectively. In contrast, only 4.8% and 28.6% of isolates from healthy carriers were genotypes A and B, respectively. MLST analysis showed the associations of genotypes A and B with ST1 and ST27 complexes, respectively. In addition, srtBCD and srtG clusters were preferentially distributed to ST1 and ST27 complex members, respectively. These results suggest that profiling of selected pilus-associated genes could be used as an easy screening method to monitor isolates important for S. suis infection.  相似文献

Sulfur (S) availability indicators are necessary for a rational use of fertilizers. Experiments were carried out to evaluate the sensitivity and stability of nitrogen (N):S ratio in spring red wheat. Yield responses were determined in three replications over five experimental sites. Sulfur application did not increase N concentration in plant, whereas the effect on total S concentration was varied. Results showed 25, 35, 80, and 85% correctly diagnosed samples at stages Z22, Z24, Z31, and Z39, respectively. In treatments without S limitations, a decrease in N:S ratio was observed as the crop cycle advanced. Nitrogen: S ratio differed according to the site; however, these variations tend to decrease as the crop cycle advance. In summary, for spring red wheat, N:S ratio in plant is a useful method for S deficiencies diagnostic from the end of tillering to flag leaf.  相似文献

A key aspect of any sulfur (S) availability indicator is its stability throughout the crop cycle. Nine experiments were conducted with the objective to evaluate the stability of the nitrogen (N):S ratio in aerial biomass (W) in spring wheat, and to define the critical curve of S dilution. As the crop cycle progressed, a decrease (P < 0.05) in total N:S was determined. This lack of stability was explained by a lower S dilution in relation to N (P < 0.05). A decrease (P < 0.05) in N accumulation rate in relation to S was obtained as the crop cycle progressed, which points out that S accumulation in relation to N is later. A first approach was determined to the definition of the S dilution critical curve (Sc) from tillering beginning to stem elongation end (Sc = 0.37 W?0.169; r2 = 0.71 and n = 24).  相似文献

In this study, an unencapsulated Streptococcus suis mutant was used to investigate the pleiotropic effects resulting from capsule loss. The capsule deficient mutant of S. suis acquired a biofilm-positive phenotype, which was associated with significantly increased cell surface hydrophobicity. Cell-associated fibrinogen-binding and chymotrypsin-like activities were decreased in the unencapsulated mutant. The mutant did not differ significantly from the encapsulated parent strain for minimal inhibitory concentrations to penicillin G, ampicillin, and tetracycline. However, while the encapsulated strain was highly resistant to the bactericidal action of penicillin G and ampicillin, the unencapsulated mutant was approximately 60-fold more sensitive. Compared with the parent strain, the unencapsulated mutant induced a much higher inflammatory response in monocyte-derived macrophages resulting in an increased secretion of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, and IL-8. The capsule appears to hinder important adhesins or hydrophobic molecules that mediate biofilm formation, as well as cell wall components capable of stimulating immune cells.  相似文献
