The pharmacokinetics of oxolinic acid and oxytetracycline were examined in kuruma shrimp (Penaeus japonicus) after intra-sinus (10 and 25 mg/kg, respectively) and oral (50 mg/kg) administration. The shrimp were kept in tanks with recirculated artificial seawater at a salinity of 22–23 ppt. The water temperature was maintained at 25±0.6 °C. The hemolymph concentrations of both drugs after intra-sinus dosing were best described by a two-compartment open model. The distribution and elimination half-lives (t1/2 and t1/2β) were found to be 0.59 and 33.2 h for oxolinic acid and 0.45 and 24.7 h for oxytetracycline, respectively. The apparent volume of distribution at a steady state (Vss) and total body clearance (CLb) were estimated to be 1309 ml/kg and 28.8 ml/kg/h for oxolinic acid and 748 ml/kg and 22.7 ml/kg/h, respectively. The hemolymph concentration–time curves after oral administration did not fit by the nonlinear least squares method using one- and two-compartment model with first-order absorption in either of the drugs. The peak hemolymph concentration (Cmax), the time to peak hemolymph concentration (tmax) and the elimination half-life were found to be 17.8 μg/ml, 7 h and 34.3 h for oxolinic acid and 24.3 μg/ml, 10 h and 33.6 h for oxytetracycline, respectively. The bioavailability (F) after oral administration was 32.9% for oxolinic acid and 43.2% for oxytetracycline. The hemolymph protein binding in vivo was determined to be 36.7±8.5% for oxolinic acid and 22.9±4.8% for oxytetracycline.  相似文献

The imponderable now is time. Certainly, it is in short supply if we hope to improve the world's prospects for food production. The requisite scientific skills that can contribute to greater world agricultural production exist in a uniquely concentrated form in the U.S. universities. Incentives for increased university involvement in technical assistance created by the International Development and Food Assistance Act of 1975 will not become effective until a mechanism which guarantees adequate and long-term funding is established. A continuing dialogue to define the nature and scope of needed reforms is necessary if maximum involvement of U.S. scientists in agricultural technical assistance is to be realized. Such involvement is vital in meeting the food needs of the developing world.  相似文献

We investigated fetal development and the estimation of fetal age of 127 Hokkaido sika deer fetuses, categorizing them into three groups according to the nutritional condition of populations. The order and time of the appearance of ossification centers were clarified, and fetal age was determined based on bone length and the appearance of ossification centers. Then we observed the differences in fetal growth among three populations, and discussed the effect of poor nutrition on the fetal growth. The results suggest that fetal diaphysial length of the femur was affected very little by nutritional conditions, whereas conception dates were delayed and fetal weight was restricted as the nutritional condition became poorer. Although it is impossible to know the exact accurate fetal age in wild populations, it was possible to create a standard to estimate fetal age more precisely by the method described in this study. Both the bone length and the appearance of ossification centers are reliable indices to estimate fetal age precisely in measurements available from fetuses of unknown age, and can be applied to estimate the fetal age of other populations of sika deer, whereas estimation of fetal age based on weight is prone to great errors.  相似文献

Cytochrome P450 (CYP) is important for metabolism of not only xenobiotics such as drugs, but also endogenous compounds including arachidonic acids. CYP4A11, CYP4F3v2, CYP4F11, and CYP4F45 have been identified in cynomolgus macaque, an animal species widely used for investigation of drug metabolism due to its evolutionary closeness to human. However, their metabolic functions have not been investigated. In this study, proteins were heterologously expressed in Escherichia coli and characterized by metabolic assays using arachidonic acids as substrates that are metabolized by CYP4 isoforms in human. The results showed that all four CYPs metabolized arachidonic acids. Therefore, cynomolgus macaque CYP4A11, CYP4F3v2, CYP4F11, and CYP4F45 are functional enzymes.  相似文献

In cynomolgus macaques (Macaca fascicularis), widely used in drug metabolism studies, CYP2C9, CYP2C76, CYP2D6, CYP3A4, and CYP3A5, important drug‐metabolizing enzymes, are abundantly expressed in liver and metabolize cytochrome P450 substrates. CYP2C9 (c.334A>C), CYP2C76 (c.449TG>A), CYP2D6 (c.891A>G), CYP3A4 (IVS3 + 1G>del), and CYP3A5 (c.625A>T) substantially influence metabolic activity of enzymes, and thus are important variants in drug metabolism studies. In this study, a real‐time PCR method was developed for genotyping these variants. The validity of the methods was verified by genotyping two wild type, two heterozygous, and two homozygous DNAs and was used to genotype 41 cynomolgus macaques (from Cambodia, Indonesia, the Philippines, or Vietnam) for the five variants, along with another important variant CYP2C19 (c.308C>T). The CYP2C9 and CYP2C19 variants were found only in Cambodian and Vietnamese animals, while the CYP2C76 and CYP2D6 variants were found only in Indonesian and Philippine animals. The CYP3A4 and CYP3A5 variants were not found in any of the animals analyzed. Mauritian animals, genotyped using next‐generation sequencing data for comparison, possessed the CYP2C19 and CYP2D6 variants, but not the other variants. These results indicated differences in prevalence of these important variants among animal groups. Therefore, the genotyping tool developed is useful for drug metabolism studies using cynomolgus macaques.  相似文献

We examined the use of external measurements and relative fat deposition of adult feral raccoons (Procyon lotor) to develop relative indices of body fat deposition in post-growth feral raccoons. From March 2006 to March 2010, 288 adult raccoon carcasses (110 males, 178 females) collected in Kanagawa Prefecture, Japan, which were determined to be 24 months old, were subjected to external measurements of body weight (BW), girth measurement (GM), and body mass index (BMI). To assess relative body fat deposition, we visually classified abdominal subcutaneous fat into three grades (Visible Fat Index [VFI]: I-III). Significant differences in the means of BW (both sexes:P<0.01), GM (females: P<0.05, males: P<0.01), and BMI (both sexes: P<0.01) were detected between seasons. Notably, the means of BW, GM, and BMI (all, both sexes: P<0.01) differed significantly between VFI grades. However, by discriminant analysis with BW, GM, and BMI as independent variables, we obtained a significant discriminant function (both sexes: P<0.01) for distinguishing VFI I from higher VFI grades, but no significant equation was obtained for distinguishing between VFI II and VFI III. Based on the obtained structure matrix of discriminant analysis, BMI was the most valuable component for the discrimination of VFI grades. Thus, we conclude that BMI is a suitable complementary index for assessing relative body fat deposition of adult feral raccoons in Kanagawa Prefecture and may be generalizable to populations in other areas.  相似文献
