Two tests are used on a regular basis to detect Mycobacterium avium subsp. paratuberculosis (Map): ELISA and fecal culture. Fecal culture is considered more sensitive and specific but is costly and requires 3-4 months for results. Pooling of fecal samples of individual animals may reduce the high costs of fecal culture. The objective of the study was to investigate the diagnostic validity and costs for pooling of fecal samples in dairy farms relative to culture or an ELISA on individual samples to determine the cow- or herd-status for Map. Fifty fecal and blood samples per herd were collected in 12 Chilean dairy herds. The sensitivity of pooling was estimated given the pool-size, amount of shedding in the pool and the prevalence in the herd. The sensitivity of the pools relative to individual fecal culture was 46% (95% CI 29-63%) and 48% (28-68%) for pools of 5 and 10 cows, respectively. The sensitivity of the pools was lower in pools with low shedders (26 and 24% for pools of 5 and 10, respectively) than in pools with moderate or heavy shedders (>75% sensitivity). Pools of 10 cows are the better option to determine or monitor the herd status. A whole-herd ELISA is the least expensive way to determine the status of individual cows but has a lower Se and Sp than individual culture.  相似文献

The aim of this study was to evaluate daily weight gain (DWG), total dry matter (DM) intake, rumen degradability of forage, and urinary excretion of mimosine metabolites by hair sheep in a silvopastoral system with high densities of Leucaena leucocephala. A completely randomized design was carried out with two treatments: treatment 1 (T1) silvopastoral system with leucaena at a density of 35,000 plants/ha and treatment 2 (T2), leucaena at a density of 55,000 plants/ha. Leucaena was associated with tropical grasses Panicum maximum and Cynodon nlemfluensis. Twenty-four male Pelibuey lambs of 23.2?±?3.4?kg live weight (LW) were used (12 lambs per treatment). Results showed differences (P?<?0.05) in DWG of T1 (106.41?±?11.66?g?1 sheep?1) with respect to that of T2 (81.33?±?11.81?g?1 sheep). Voluntary intake was higher in lambs from T1 (83.81?±?04.07?g DM/kg LW0.75) with respect to that from T2 (71.67?±?8.12?g DM/kg LW0.75). There was a difference in color of urine between sheep of T1 and T2, the latter giving positive results for the presence of metabolites derived from mimosine (3-4 dihydroxypyridine and 2-3 dihydroxy pyridone). Rumen degradability of DM of L. leucocephala was higher (P?<?0.05) compared to that of P. maximum and C. nlemfluensis (72.94?±?0.40 vs. 67.06?±?1.50 and 63.25?±?1.51?%, respectively). It is concluded that grazing at high densities of L. leucocephala affects daily weight gain of hair sheep, possibly due to ingestion of high amounts of mimosine which may exert an adverse effect on voluntary intake.  相似文献

Leaf blight disease of water hyacinth was observed and collected from different geographical areas of Thailand. The disease is caused by a fungal pathogen that was identified as Myrothecium roridum by using its morphological characteristics. The most effective fungal strains were evaluated for pathogenicity on water hyacinth under greenhouse and natural conditions. Myrothecium roridum isolate, Kamphaeng Sean Campus (KKFC) 448, was found to be the most virulent. Different fungal formulations were evaluated for their level of control of water hyacinth. The results showed that spore suspensions with 10% palm oil or 1% Tween 20 caused a higher level of disease severity, compared to spores applied in water alone. The host range of KKFC 448 was evaluated by using 77 plant species that belong to 40 plant families. The fungus did not cause disease on 74 economically important plants but did produce disease signs on water hyacinth and two other aquatic weeds, duckweed and water lettuce. Leaf blight occurs on water hyacinth leaves after being treated with crude extracts of M. roridum and it was indicated that secondary metabolites were released from the fungal mycelia. Myrothecium roridum that was grown on boiled paddy rice produced β‐1,4‐exoglucanase, β‐1,4‐endoglucanase, β‐glucosidase, xylanase and pectinase more than Mroridum that was grown on potato dextrose agar. The results indicated that M. roridum is a pathogen of water hyacinth and the fungus is capable of producing different enzymatic activities on potato dextrose agar and boiled paddy rice, which might be important for infection.  相似文献

The aim of the study was to evaluate the effect of graded levels of Enterolobium cyclocarpum pods in the ration on feed intake and digestibility by Pelibuey lambs. Five dietary treatments were imposed where ground pods replaced concentrate diet at 0, 20, 30, 40 and 50 % of dry matter (DM), respectively. The concentrate portion was composed of ground sorghum, soybean meal, cane molasses and minerals. Five entire Pelibuey lambs with initial bodyweight 34?±?2 kg were allocated in the treatments in a 5?×?5 Latin square design. Values of dry matter intake (DMI) and dry matter (DMD) and organic matter (OMD) digestibility were measured and metabolisable energy intake (MEI) estimated. Rumen degradation constants for E. cyclocarpum were also measured. There were no differences (P?>?0.05) in average DMI (86.6 g/kg0.75) and OMI (81.2 g/kg0.75) among treatments. As the level of incorporation of E. cyclocarpum pods increased, voluntary DMI and OMI increased, whereas apparent DMD and OMD decreased linearly. Average digestible DM (65 g/kg0.75) and OM (61 g/kg0.75) intakes were similar (P?>?0.05) among treatments. Similarly, MEI (0.976 MJ ME kg0.75/day) was not different (P?>?0.05) among treatments. The potential rumen degradation (A?+?B) of ground pods of E. cyclocarpum was 866.4 g/kg DM. Ground pods of E. cyclocarpum can be employed for lamb feeding up to 50 % of the ration, without affecting DMI, DM apparent digestibility and MEI.  相似文献
