采用Nano–LC–ESI–MS/MS蛋白鉴定技术对湖南永州尖吻蝮蛇蛇毒蛋白组分进行质谱分析,应用常规体外试管法对尖吻蝮蛇蛇毒体外溶血值进行检测,采用改良寇式法对尖吻蝮蛇蛇毒LD_(50)进行测定。结果显示:尖吻蝮蛇蛇毒含有50种蛋白,蛋白相对分子质量集中在1.0×10~4~2.0×10~4(46.9%)、4.0×10~4~5.0×10~4(22.4%)、2.0×10~4~3.0×10~4(10.6%)和7.0×10~4~8.0×10~4(7.6%),其中代表性的高丰度蛋白有蛇毒金属蛋白酶A(11.7%)、蛇毒金属蛋白酶H1(9.8%)、蕲蛇类凝血酶–2(7.3%)、抗凝血酶A–A亚基(6.8%),低丰度蛋白(0.1%)有Ecto–5'–核苷酸酶、碱性磷脂酶A2 DAV–N6、生长分化因子11;蛇毒引起红细胞溶血的最低质量浓度为60.00μg/mL,尖吻蝮蛇蛇毒对KM小鼠的LD_(50)为7.1796mg/kg,攻毒小鼠出现呼吸急促、躁动不安、注射部位出现奇特瘙痒和大面积溃烂,至死亡。  相似文献

中药苏木(Sappan Lignum)来源于植物苏木(Caesalpinia sappan L.)的干燥心材,具有止血 抗炎等功效。采用琼脂糖平板法研究不同浓度的苏木水提物(0.03g/ml,0.06g/ml,0.09g/ml, 0.12g/ml,0.15g/ml)对蛇毒sPLA2水解活性的抑制作用,测量透明圈和孔的直径,根据酶活标准曲 线计算其抑制率。结果表明0.15g/ml 苏木水提物对五步蛇毒sPLA2水解活性有抑制作用。  相似文献

Abstract

Crayfish harvest was initiated at a cool water temperature (15°C) and warm water temperature (20°C) from culture ponds (0.1 ha) with a resident population of white river crayfish, Procambarus acutus acutus.Crayfish were harvested for 24 days for a total of 1,977 trap-days/ha (March 9-May 12, 2000 from the cool-harvest treatment and May 5-June 15, 2000 from the warm-harvest treatment). The mean total length (TL) and production of harvested white river crayfish (WRC) was similar between treatments. The population structures of WRC from the cool and warm periods of harvest were substantially different; large WRC (≤100 mm TL) from the cool-harvest period were 3.1 times more abundant than large WRC during the warm-harvest period. A similar number of small WRC were harvested from the two treatments; however, those small WRC harvested during the warm period was significantly longer than the small WRC during the cool-harvest period. The estimated large WRC biomass contribution to the total harvest from the cool-harvest period was 37% in comparison to 15% during the warm-harvest period. The estimated dollar value of the production during the cool-harvest period was 40% more than that from the warm-harvest period.  相似文献

福建蕲蛇粗毒经硫酸铵沉降处理后,进行DEAE-Sephadex A-50色谱分离,取其最高活性的第11峰进行G-75凝胶色谱分离后,经CM-Sephadex C-50离子交换色谱、HQ20离子交换色谱分离,获得一个有高凝血活性的组分,在SDS-PAGE电泳图谱上对应分子量为24100。与其他从蕲蛇中已分离的类凝血酶比较,该蛋白组分是一个新的类凝血酶。  相似文献

AIM:To investigate the effects of protein C activator (PCA) from Agkistrondon acutusvenom (AAV) on the tension of thoracic aorta rings isolated from the rats with sepsis. METHODS:The model of sepsis was established by intraperitoneal injection of lipopolysaccharide (LPS). SD rats were randomly divided to 6 groups (n=6): sham group, LPS group, PCA intervention group (LPS+PCA, PCA at doses of 0.1 mg/kg, 0.3 mg/kg and 0.6 mg/kg) and LPS+ polymyxin B (at dose of 0.2 mg/kg) group. Using perfusion experiment in vitro, the tension of the aortic rings was measured by biological signal analytical system. RESULTS:The values of MABP, HR, LVDP and ±dp/dtmax were significantly lower in LPS group than those in sham group and LPS+PCA groups. Compared with sham group, the relaxation response to acetylcholine (ACh) and the contractile response of aorta rings induced by phenylephrine (Phe) were significantly decreased in LPS group, which were increased significantly in PCA intervention group (especially at dose of 0.6 mg/kg) compared with LPS group. The dose-response curve of aorta contraction with denuded endothelium induced by Phe shifted down significantly in LPS group compared with sham group, and no significant difference between LPS group and PCA intervention group was observed. Also no statistical difference was found in non-endothelium dependent relaxation of aortic rings induced by sodium nitroprusside among the groups. Pretreatment of N-nitro-L-arginine methl ester and methylene blue increased the contraction amplitude of aortic rings induced by Phe. CONCLUSION: PCA from AAV effectively reverses the hypoergia of the vessels in rats with sepsis through protecting vascular endothelium, the mechanism of which may be mediated by inhibiting NO-GC-cGMP signal transduction pathway.  相似文献

【目的】采用分子生物学方法鉴定1个五步蛇(Deinagkistrodon acutus)源隐孢子虫分离株,了解中国蛇隐孢子虫感染的种类分布。【方法】利用巢式PCR方法,扩增隐孢子虫18S rRNA基因特异性片段,对PCR产物进行限制性片段长度多态性分析(PCR-RFLP)和同源性分析,构建其种系发育进化树。【结果】成功扩增出18S rRNA基因目的片段,PCR产物经SspⅠ酶消化后获得的酶切结果和先前报道的蛇隐孢子虫(Cryptosporidium serpentis)一致。获得的840 bp 18S rRNA基因序列与C.serpentis同源性达到99.8%。种系发育进化树分析表明,该分离株与C.serpentis位于同一分支,节点支持值达100%。【结论】本研究获得的五步蛇源隐孢子虫分离株为C.serpentis,说明C.serpentis有更宽的感染宿主范围。  相似文献

Based on a recently discovered property of chloroacetamide herbicides—the inhibition of the incorporation of oleic acid into sporopollenin of Scenedesmus acutus—a rapid quantitative test was developed for chloroacetamide-type herbicidal activity. In this test, algal cells are incubated for 3 h with [14C]oleic acid, saponified and the lipids (including non-saponifiable ones) extracted and discarded. The radioactivity incorporated into the residual non-lipid fraction is determined, and inhibition of this incorporation is used as a marker of chloroacetamide-type activity. Twenty-two agrochemical compounds were screened in this assay, which was found to be very sensitive, a 50% inhibition being reached with submicromolar herbicide concentrations. It is specific to chloroacetamides and related amides, since all these herbicides tested were potent inhibitors, while other herbicides were not. Highest inhibition was shown by cafenstrole followed by butachlor, fluthiamid, metazachlor, alachlor, dimethenamid, metolachlor and mefenacet. For these herbicides (with the exception of butachlor) sensitivity in the test was positively correlated (r=0·984) with their phytotoxic effect on the alga. © 1998 SCI  相似文献

用稻草、塑料薄膜、泥土、砖块制成人造蝮蛇、五步蛇的越冬蛇窝,窝内温度可保持在气温5℃以上,湿度50%左右,用麦乳精、葡萄糖配成的营养液作为越冬蛇营养补充的饲料,通过上述处理,越冬蝮蛇和五步蛇的成活率可以提高到92%以上,体重和体质也有很大提高,经济效益明显。  相似文献

Abstract

The effects of renovation on the production of white river crayfish, Procambarus acutus acutus were evaluated in ten 0.1-ha earthen ponds. The bottoms and levees of five ponds were reshaped with heavy equipment prior to planting sorghum-Sudan grass forage during the de-watered phase of the culture cycle. Five non-renovated ponds served as a control. The mean total length (TL) of the crayfish harvested from renovated ponds was significantly larger than those crayfish harvested from non-renovated ponds. The renovated pond harvest contained an estimated 43.2% carryover crayfish (>95 mm TL) in comparison to 7.6% in the harvest from non-renovated ponds. By weight, carryover crayfish constituted an estimated 69.3% of the harvest from renovated ponds, compared to 21.0% from non-renovated ponds. No difference was observed in the harvest between treatments; however, the low proportion of young-of-the-year crayfish in the renovated pond harvest may adversely impact future production.  相似文献
