Conventional methods of DNA recovery from agarose gel generally require expensive equipment, extended elution times, or considerable handling of the sample after elution. We developed a simple protocol for a quick and effective recovery of DNA from agarose gels with good yield and quality. Using a Sephadex resin filled spin column, DNA fragments of 500 bp to 6 kb in an agarose gel slice were easily recovered by a 2 min centrifugation. The recovery efficiencies were over 40%-50% and the eluted DNA can be used directly for downstream application, such as polymerase chain reactions (PCR) and restriction enzyme digestion. This method could also be used to recover large DNA fragment (48 kb) without degradation. The use of Sephadex helps to remove small molecular impurities from agarose and it also reduces the chance of clogging the column filter caused by direct contact with agarose.  相似文献

The inhalation of grain dust by grain workers is responsible for a large number of pulmonary pathophysiologies. These problems may be acute or chronic and may be mediated by the chronic activation of the immune system. Constant inflammatory states in the lung may eventually lead to tissue damage and respiratory deficit. This study was designed to measure the changes in the relative number of inflammatory cells in peripheral blood, bronchoalveolar spaces, and lung interstitium that occur in response to intratracheally instilled airborne spring wheat dust in rats. It was found that 6h after instillation with dust, neutrophils were present in greater numbers in the blood and bronchoalveolar spaces than in lung interstitium. After 24h, there appeared to be a larger number of neutrophils in the lung interstitium in dust-instilled animals than in saline-instilled controls. These results indicate that intratracheal instillation of grain dust initiates an acute inflammatory reaction, and that there is an initial influx of neutrophils into the air spaces of the lung followed by transit of these cells into the lung interstitium.  相似文献

The influence of forepoles on the patterns of a single tunnel plastic collapse was discussed using Acutronic 661. A series of plane strain centrifuge model tests in over-consolidated clay were conducted. Tests were carried out in a circular cavity with stiff resin inclusions embedded around the tunnel vault. These inclusions simulated elements of a secant pipe or forepole arch system. An image processing system was used to measure displacements below surface level and displacement transducers at the surface level of the models. The aim of the experimental work was to study the ground response due to simulated tunnel construction and to analyse how the vault stiff inclusions affect tunnel stability. According to the principle of the upper bound theorem of plastic limit analysis, a collapse mechanism of the single circular tunnels with embedded forepoles in cohesive soil was established. An upper bound equation of stability ratio was obtained. The upper bound solutions of stability ratio under different soil cover above the tunnel, undrained shear strength at tunnel axis level and embedded forepoles range were discussed from the equation. In the end,the correctness of the upper bound solutions was proved by the results of centrifugal model tests.  相似文献

An effect of freeze-milling on processing properties of beta-glucan oat bran has been evaluated. A comparison with existing, patented methods of high molecular weight oat beta-glucan has been carried out. The new method employs raw material pre-treatment with freezing and milling in a hammer mill, resulting in significant reduction of particle size – 89% was between 80 and 50 μm in comparison with initial material when 79% were between 250 and 200 μm. Reduction of particle size also improved some process parameters of extraction technology – 30% improvement in fat removal during first stage of beta-glucan recovery was observed. The achieved pilot plant yield of product containing beta-glucan extraction was 64,03%, purity (ie. beta-glucan content), of achieved product was 84,4% and average molecular weight was about 69,500 g/mol.  相似文献

【目的】根据渠道地基的运行特点,对浸水条件下的膨胀土渠道地基进行离心模拟试验。【方法】将模型离心加速度增加到60g,然后浸水并保持水头8.5 cm,再用各种传感器测量浸水过程中模型的表面位移、内部土压力和吸力。【结果】在上覆水压力作用下,膨胀土先发生压缩,然后由于水分浸入发生轻微膨胀,并导致侧压力系数逐渐增大。试验后对模型含水量和干密度的测定表明,模型内形成了含水量梯度,干密度随深度逐渐增加而增大。【结论】模型变形和内部土压力变化是模型内部含水量变化的结果,同时也受上覆压力影响;基质吸力主要与含水量有关,受重力加速度变化的影响较小。  相似文献

在建立分离机立轴轴系计算模型的基础上,采用有限元分析软件ANSYS进行分析计算,得出碟式分离机立轴系统各阶临界转速,与实测结果相比,两者具有较好的一致性;对影响轴系临界转速的结构参数进行了计算分析比较,明确了主要零部件对各阶临界转速的影响,为该类机器的结构设计和动平衡提供依据。  相似文献
