为探究体细胞克隆荷斯坦奶牛出生后死亡的原因,对新生后死亡的克隆荷斯坦公牛和自然繁殖的荷斯坦公犊的主要组织器官进行比较。通过解剖和石蜡切片-HE(hematoxylin-eosin staining)染色技术,对主要的组织器官结构进行观察和分析。结果表明,新生后死亡的克隆荷斯坦公牛肺部结构清晰;肝脏的肝细胞明显肿大,出现脂肪轻度变性;肾小管上皮细胞出现变性;心肌的肌纤维间空隙增大;骨骼肌纤维间隙明显,空泡变性,这可能导致该克隆牛犊肌肉无力并功能不全;淋巴结皮髓质界限不分明,淋巴小结细胞较稀疏,生发中心不明显,淋巴窦细胞较少;脾脏内红细胞较少,这说明该克隆牛的造血功能可能不完善;胸腺的皮质部分和髓质部分界限不明显,嗜酸性胸腺小体不易辨认,可能发育不全。该克隆公牛免疫器官出现不同程度的发育不全现象较严重,这有可能是其出生后死亡率高的主要原因。 相似文献

The purpose of this study was to evaluate the usefulness of ultrasound-guided fine-needle aspiration biopsy and core biopsy in the diagnosis of infiltrative gastrointestinal diseases. Six dogs and seven cats with clinical signs of gastrointestinal disease underwent ultrasonography and intestinal lesions were identified. One or more ultrasound-guided fine-needle aspiration biopsy and/or core biopsy procedures were performed in each patient. Each patient also underwent one of the following additional procedures for comparison of results: 1) surgery (n=4), 2) endoscopy (n=2), 3) post mortem exam (n=3), and, 4) for lymphoma diagnosed with ultrasound-guided procedures, response to chemotherapy (n=4). Correct diagnoses were obtained in nine of the 13 patients, incorrect diagnoses in two of the 13 patients, and inconclusive diagnoses in two of the thirteen patients. Of the 16 total ultrasound-guided procedures performed, ten were confirmed as correct, three as incorrect, and three were non-diagnostic. Intestinal lesions with bowel wall thickness greater than 2.0 cm had a higher percentage of correct diagnoses than lesions of lesser wall thickness. Gastric lesions had a higher percentage of correct diagnoses than small and large intestinal lesions. Malignant lesions had a higher percentage of correct diagnoses than benign lesions. There were no complications. Ergot, caused by Claviceps africana , has emerged as a serious threat to sorghum hybrid seed production worldwide. In the absence of gene-for-gene-based qualitative resistance in commercial cultivars, varieties with high pollen production that can escape ergot infection are preferred. Recent demonstration of differences in ergot susceptibility among male-sterile lines has indicated the presence of partial resistance. Using chitin-specific fluorescin-isothiocyanate-conjugated wheat germ agglutin and callose-specific aniline blue, this study investigated the process of sorghum ovary colonization by C. africana . Conidia germinated within 24 h after inoculation (a.i.); the pathogen was established in the ovary by 79 h a.i., and at least half of the ovary was converted into sphacelial tissue by 120 h a.i. Changes in fungal cell wall chitin content and strategic callose deposition in the host tissue were associated with penetration and invasion of the ovary. The rate of ovary colonization differed in three male-sterile lines that also differed in ergot susceptibility. This work demonstrates a possible histological basis for partial resistance in male-sterile sorghum lines that could lay the foundation for variety improvement through further breeding and selection. 相似文献

Toxoplasma gondii is a zoonotic protozoan pathogen that infects many endothermic vertebrates, including humans; the domestic cat and other felids serve as the definitive host. Macropodids are considered highly susceptible to toxoplasmosis. Here, we describe the clinical, pathologic, and immunohistochemical findings of an outbreak of systemic toxoplasmosis in a mob of 11 red kangaroos (Macropus rufus), with high morbidity (73%) and mortality (100%) rates. Affected animals had either severe and rapidly deteriorating clinical conditions or sudden death, which was correlated with widespread necrotizing lesions in multiple organs and intralesional T. gondii organisms identified via MIC3-specific immunohistochemistry and confirmed by REP529-specific rtPCR. Quantification of parasites demonstrated the highest parasite density in pulmonary parenchyma compared with other tissues. 为探究高水温对尼罗罗非鱼无乳链球菌病暴发的影响,以体质量为(62.0±3.33)g的尼罗罗非鱼为研究对象,设置生长最适宜组(29°C)和高温应激组(33°C)2个处理水平,暂养7 d后分别进行人工感染实验统计累计死亡率,并于攻毒后0、12、24、48、96和120 h采集血液和组织样本,开展高温应激下无乳链球菌感染对尼罗罗非鱼血清生化指标和组织病理影响的研究。结果显示,水温33°C高温应激组尼罗罗非鱼累计死亡率显著高于29°C组;谷丙转氨酶(ALT)活性在感染96 h之后33°C组的值显著高于29°C组;谷草转氨酶(AST)活性在感染48 h之后33°C组的值显著高于29°C组;钾(K+)和钠(Na+)含量感染120 h时29°C组的值和攻毒前都没有显著性差异,而33°C组分别显著高于和低于攻毒前;肌酐(CREA)含量在感染12 h以后33°C组的值都显著高于29°C组;白蛋白/球蛋白(A/G)的值在120 h时33°C组的值显著低于29°C组;血清碱性磷酸酶(AKP)活性29°C组随时间延长而升高,33°C组则先升高后降低,但33°C组的达峰时间更短;超氧化物歧化酶(SOD)活性的变化趋势都是先升高后降低再升高;组织病理学观察显示高温应激使肝细胞排列紊乱,索状结构不清,脾脏和肾脏轻微充血;感染12 h后脾脏均严重充血,感染24 h后脾脏结构均被破坏,呈淀粉样变性,坏死;感染48 h后肾小球均发生萎缩,肾小管上皮细胞变性、坏死;33°C组鱼的肝脏感染96 h后严重脂肪变性和细胞内玻璃样变性,而29°C组鱼的肝脏在感染120 h后才发生相似的病理变化。研究表明高温应激抑制了鱼体免疫系统,降低了尼罗罗非鱼对无乳链球菌的抵抗力,脾脏对无乳链球菌感染响应最快,高温应激使病原菌感染对肝脏损伤更为迅速,感染后期受损更严重。 相似文献
