Previous experiments demonstrated that treatment of longan trees with potassium chlorate (KClO3) induces “off season floral induction” (FI) even in the absence of the naturally required cool temperature [Manochai, P., Sruamsiri, P., Wiriya-alongkorn, W., Naphrom, D., Hegele, M., Bangerth, F., 2005. Year around off season flower induction in longan (Dimocarpus longan, Lour.) trees by KClO3 applications: potentials and problems. Sci. Hortic. 104, 379–390]. Potassium chlorate, however, cannot replace the presence of functional mature leaves and sufficient light intensity. Here we examined in more detail the effect of shade (about 10% of natural sunlight) on KClO3 affected hormone concentrations/transport of leaves and shoot apical buds (SAB) and their interactions with FI.  相似文献

Corn has several mechanisms that allow it to perceive the incoming water deficit and rapidly regulate its growth and physiology to cope with this stress. Among these regulated physiological responses, phytohormones, such as abscisic acid (ABA), gibberellin (GA3), indole acetic acid (IAA), and zeatin (ZT) have important roles. To evaluate changes in hormonal status of corn hybrids under different drought stress levels, a greenhouse study was conducted at the College of Agriculture, Shiraz University, Iran, during summer 2010. Treatments consisted of drought stress in five levels: 100% field capacity (F.C.) as control, 80, 60, 40, and 20% F.C. and three corn hybrids, including single cross 704 (SC704), Maxima, and double cross 370 (DC370). Results showed that drought stress significantly affected phytohormones concentration, ABA concentration was increased and GA3, IAA and ZT concentrations were decreased. Indeed, there was an initial increase (about 13.5% more than control) in IAA concentration. In severe drought condition, ABA concentration was eventually decreased (37.8% less than the first drought level, i.e., 80% F.C.). The ratios of GA3, IAA and ZT to ABA concentration were also decreased. Among hybrids, SC704 showed the highest phytohormone concentrations and DC370 had the highest ratios of GA3, IAA, and ZT to ABA concentrations. Overall, the results of this study suggest that although mild drought stress was associated with greater ABA concentration, it had negative impact on GA3, IAA, and ZT concentrations. Furthermore, SC704 hybrid appeared to be more resistant to drought stress than the other hybrids.  相似文献

Abstract

The leaves of intact soybean plants (Glycine max. L. Merrill) characteristically turn yellow and abscise during pod maturation, and this may limit seed growth. We have used soybean expiants (excised 10 cm stem sections with a leaf and midfill pod attached) to study interactive effects of nutrient and hormone supply on leaf senescence symptoms and seed yield. Whereas mineral nutrients or cytokinin (zeatin) alone delayed development of visible leaf senescence symptoms in explants, a combination of mineral nutrients with zeatin acted synergistically to maintain green coloration throughout pod maturation and reduced declines in levels of leafsoluble protein and chlorophyll. The combination of complete mineral nutrients with zeatin gave seed yields equivalent to those in intact plants. Withdrawal of zeatin or individual mineral nutrients (particularly N and S) from the nutrient solution enhanced leaf senescence symptoms and reduced seed yields.  相似文献

The object of this study was to induce embryogenesis and establish plant regeneration system for anther culture in loquat (Eriobotrya japonica L.). Cold pretreatment was a key factor, and supplement of 2,4-D in the media was absolutely necessary for induction of calluses from cultured loquat anthers. The best response of anthers to in vitro culture was obtained when a 48-h cold pretreatment was employed to flower buds at 4 °C in darkness. Genotype was a decisive factor for embryo differentiation. When anther-derived calluses of three loquat cultivars, i.e., cv. ‘Longquan1’, ‘Dawuxing’ and ‘Zaozhong6’, were transferred to embryo differentiation medium, embryos were induced only for cv. ‘Dawuxing’ on MS medium containing 3% sucrose, 0.23 μM ZT in combination with 0.05 μM NAA + 0.05 μM IBA or 0.11 μM NAA + 0.10 μM IBA, and the differentiation rates were 3.33% and 10.00%, respectively. The results of histological studies showed that embryos developed through typical globular, heart, torpedo and cotyledon stages after 4 weeks of culture. The treatment designed to mature the embryos on medium containing 3% of sucrose at 4 °C under darkness for 4 weeks was effective for subsequent embryo germination and plant conversion, which gave rise to 72.5% plant recovery. Cytological studies showed that 26 plantlets were haploids (n = 17) and the remaining 4 plantlets were diploids for the 30 regenerants tested.  相似文献

In vitro seeds germination and plantlet establishment of Labisia pumila were studied in this report. The seeds obtained from the mature fruits of L. pumila were sterilized and cultured on Murashige and Skoog (MS) solid media supplemented with 1–3 μM of 6-benzylaminopurine (BAP) and 3% (w/v) sucrose. The presence of BAP in the medium significantly affects seeds germination. High percentage of seeds germination (up to 90%) was successfully achieved after 2 weeks of culture on medium supplemented with 2 μM BAP. Up to 70% of explants produced shoots through direct regeneration from newly emerged epicotyls after 5 weeks of culture. The average of 8.1 ± 1.0 shoots per explant obtained on media treated with 2 μM BAP. Seedlings were further transferred to growth media fortified with different types of cytokinin. Result observed after 12 weeks showed that medium supplemented with 1 μM zeatin (ZEA) promote the highest growth with an average of 2.9 ± 1.0 cm shoot length and 7.7 ± 3.2 leaves per explant after 12 weeks. In addition, medium added with 2 μM BAP and supplemented with 3–4% (w/v) of sucrose promote the best growth i.e., 3.0 ± 0.6 shoots per explant, 2.27 ± 0.2 cm length and 4.3 ± 0.5 leaves per explant.  相似文献

Success in “super” rice breeding has been considered a great progress in rice production in China. This study aimed to test the hypothesis that an improved root system may contribute to better shoot growth and consequently to higher grain yield in “super” rice. Two “super” rice varieties Liangyoupeijiu (an indica hybrid) and Huaidao 9 (a japonica inbred) and two elite check varieties Yangdao 6 (an indica inbred) and Yangfujiang 8 (a japonica inbred) were field-grown at Yangzhou, China in 2006 and 2007. Root and shoot dry weight (DW) was significantly greater in “super” rice varieties than in check ones throughout the growth season in both years, so was the root length density. Root oxidation activity (ROA) and root zeatin (Z) zeatin riboside (ZR) content, in per plant basis, were significantly greater in “super” rice than check varieties before and at heading time. However, both ROA and root Z + ZR content, either in per plant basis or per unit root DW basis, were significantly lower in ‘super’ rice than in check varieties at the mid- and late grain filling stages. Grain yield of the two ‘super’ rice varieties, on average, was 10.2 t ha−1 in 2006 and 11.4 t ha−1 in 2007, and was 13% and 21% higher than that of check varieties, respectively. The high grain yield was mainly due to a larger sink size (total number of spikelets) as a result of a larger panicle. The percentage of filled grains of the two “super” rice varieties, on average, was 72.9% in 2006 and 79.0% in 2007, and was 19.4% and 12.9%, respectively, lower than that of the check varieties. The mean ROA and root Z + ZR content during the grain filling period significantly correlated with the percentage of filled grains. Collectively, the data suggest that an improved root and shoot growth, as showing a larger root and shoot biomass and greater root length density during the whole growing season and higher ROA and root Z + ZR content per plant at early and mid-growth stages, contributes to the large sink size and high grain yield in the “super” rice varieties. The data also suggest the yield of “super” rice varieties can be further increased by an increase in filled grains through enhancing root activity during grain filling.  相似文献

为研究干旱胁迫下玉米素对高羊茅(Festuca arundinacea)和多年生黑麦草(Lolium perenne)抗旱性的影响,试验在土壤含水量分别为90%、60%、40%、25%的干旱胁迫和土壤含水量始终为90%的充分浇水条件下,设4个玉米素浓度处理(0,10,50,100μmol/L),测定了两种草坪草的坪观质量、叶片相对含水量、叶片萎焉、电导率、脯氨酸、丙二醛(MDA)和地下生物量等指标变化。结果表明:与不施加玉米素处理相比,干旱胁迫下,施加玉米素能显著提高两种草坪草的草坪质量和相对含水量(P0.05),减轻草坪草叶片细胞电解质渗漏和萎蔫度(P0.05),降低脯氨酸和丙二醛(MDA)含量(P0.05),提高高羊茅和多年生黑麦草抗旱性。在干旱胁迫后期,高浓度玉米素处理(50μmol/L或100μmol/L)的高羊茅和多年生黑麦草各项指标均表现出更加显著的优势(P0.05),表明高浓度玉米素对草坪草抵御干旱胁迫的作用更大。  相似文献

Three tomato cultivars were used to examine the influence of the genetic background on the regeneration efficiency. White embryonic calli were formed within two weeks of culture. Shoots emerged either directly from the explant or indirectly from the embryonic callus. Multiple adventitious shoots were formed by clonal propagation of somatic embryos in the presence of 2 mg zeatin L-1. The meristematic end of the hypocotyl of the cultivar Pontaroza showed a high regeneration frequency (70.2%) compared with the cotyledonary leaf explant (35.3%). The plants grown in the green house and the regenerants obtained showed a similar peroxidase banding pattern. The combined analysis of variance indicated that the difference in shoot induction between cultivars was highly significant. Shoot induction frequency was 57.2%, 43.5%, and 35.5% for the cultivars UC-97, Pontaroza, and Zuishi, respectively. The regeneration frequency was 50%, 28%, and 20% for the cultivars UC-97, Pontaroza, and Zuishi, respectively. The observed differences in shoot induction between cultivars were due to the genetic difference between them.  相似文献
