Cathelicidins抗菌肽在家禽天然免疫系统中发挥重要作用,本研究将已构建的含Cathelicidins抗菌肽成熟肽的阳性克隆菌株Rosetta(DE3)/pET30-CathL1S、pET30-CathL2S、pET30-CathL3S分别在37℃、1.0 mmol/L IPTG的条件下进行诱导表达。SDS-PAGE和Western blotting分析结果表明,Cathelicidins成熟肽片段均在大肠杆菌中以融合形式成功表达,表达产物的表观分子质量分别为7、8、7.5 ku。琼脂糖孔穴扩散法检测结果显示,表达产物对多种革兰氏阴性菌和阳性菌均具有很好的抑制活性,其对金黄色葡萄球菌(Staphylococcus aureus)、枯草芽孢杆菌(Bacillus subtilis)、藤黄微球菌(Micrococcus luteus)、大肠杆菌(Escherichia coli)、鸡白痢沙门氏菌C79-13 (Salmonella pullorum)、巴氏杆菌、鸭疫里默氏杆菌及绿脓杆菌等供试菌株均有较强的抑制作用,尤其对抗生素耐药菌株有明显的抑制作用。以雏鸡为实验模型研究重组抗菌肽对金黄色葡萄球菌的体内抗菌活性,结果表明,注射中、高剂量CathL-1抗菌肽试验组(5、10 mg/kg),在试验结束后取肝脏组织匀浆液涂布于TMP平板,未分离到葡萄球菌。各抗菌肽试验组平均增重均高于感染对照组,其中低剂量抗菌肽试验组(2.5 mg/kg)体内抑菌效果虽与普通抗生素相当,但其增重效果尤为明显。  相似文献

Cathelicidins抗菌肽是含高度保守cathelin结构域的一大类抗菌肽,在家禽天然免疫系统中发挥重要作用。通过PCR方法,从含乌骨鸡Cathelicidins抗菌肽基因(CathL\|1,\|2,\|3)cDNA完整序列的质粒pMD\|CathL中分别扩增Cathelicidins\|1,\|2,\|3成熟肽的编码序列,将其分别克隆至原核表达载体pET\|30a(+)中,构建其重组表达质粒pET30\|CathL1S,pET30\|CathL2S,pET30\|CathL3S,经测序验证的重组质粒转化大肠杆菌Rosetta(DE3)菌株。阳性克隆菌株在37℃,10 mmol·L-1 IPTG的条件下进行诱导表达。SDS\|PAGE和Western\|blot分析表明,Cathelicidins成熟肽片段均在大肠杆菌中以融合形式成功表达,表达产物的表观分子量分别为70,80,75 kD。琼脂糖孔穴扩散法检测显示表达产物对多种革兰氏阴性菌和阳性菌均具有很好的抑制活性,其对金黄色葡萄球菌(Staphylococcus aureus)、枯草芽孢杆菌(Bacillus subtilis)、藤黄微球菌(Micrococcus luteus)、大肠杆菌(Escherichia coli)、鸡白痢沙门氏菌C79\|13 (Salmonella pullorum)、巴氏杆菌、鸭疫里默氏杆菌、绿脓杆菌等供试菌株均有较强的抑制作用,尤其对抗生素耐药菌株有明显的抑制作用。  相似文献

基于棘腹蛙(Paa boulengeri)皮肤转录组数据库,釆用RT-PCR方法从棘腹蛙皮肤组织中克隆到了编码Cathelicidin-Pb前体的核苷酸序列,其完整ORF长度为441 bp,编码146个氨基酸残基。通过信号肽预测和结构域查找比对,发现Cathelicidin-Pb前体包含N端20个氨基酸残基的信号肽、101个氨基酸残基的中间间隔区(cathelin结构域)和25个氨基酸残基的成熟抗菌肽。进一步研究发现,在适度高温范围内,皮肤组织中Cathelicidin-Pb的表达随着生长环境温度的升高而逐渐增加,但是当环境温度超过27℃后,其表达量急剧降低;Cathelicidin-Pb主要在皮肤组织中表达,在肌肉组织和血液组织的表达量较低。这些结果为棘腹蛙养殖过程中疾病控制及Cathelicidin-Pb抗菌肽的后续开发提供了重要的线索。  相似文献

抗菌肽是近年来发现的广泛存在于自然界中的一类阳离子抗菌活性肽,多数抗菌肽具有分子小、带正电、两亲性、抗菌谱广等共同特点。防御素和Cathelicidins是哺乳动物体内的两大主要抗菌肽家族,在哺乳动物的机体防御中起着重要的作用。猪源抗菌肽的一般性质、基因及其表达、在宿主防御中的作用、作用机理及研究前景将在文中予以概述。  相似文献

ABSTRACT: Cathelicidins are a major family of antimicrobial peptides present in vertebrate animals with potent microbicidal and immunomodulatory activities. Four cathelicidins, namely fowlicidins 1 to 3 and cathelicidin B1, have been identified in chickens. As a first step to understand their role in early innate host defense of chickens, we examined the tissue and developmental expression patterns of all four cathelicidins. Real-time PCR revealed an abundant expression of four cathelicidins throughout the gastrointestinal, respiratory, and urogenital tracts as well as in all primary and secondary immune organs of chickens. Fowlicidins 1 to 3 exhibited a similar tissue expression pattern with the highest expression in the bone marrow and lung, while cathelicidin B1 was synthesized most abundantly in the bursa of Fabricius. Additionally, a tissue-specific regulatory pattern was evident for all four cathelicidins during the first 28 days after hatching. The expression of fowlicidins 1 to 3 showed an age-dependent increase both in the cecal tonsil and lung, whereas all four cathelicidins were peaked in the bursa on day 4 after hatching, with a gradual decline by day 28. An abrupt augmentation in the expression of fowlicidins 1 to 3 was also observed in the cecum on day 28, while the highest expression of cathelicidin B1 was seen in both the lung and cecal tonsil on day 14. Collectively, the presence of cathelicidins in a broad range of tissues and their largely enhanced expression during development are suggestive of their potential important role in early host defense and disease resistance of chickens.  相似文献

The antimicrobial activity of linear, cationic alpha-helical peptides from cattle (BMAP28), sheep (SMAP28 and SMAP29), and pigs (PMAP23) were assessed to determine if activity was selective for Pasteurella multocida from a particular animal species or broad-spectrum against all P. multocida tested. The antimicrobial activities of synthetic peptides were determined for P. multocida isolated from cattle (10 isolates), sheep (10 isolates), and pigs (10 isolates) in a broth microdilution assay. All thirty isolates of P. multocida were susceptible to BMAP28 (MICs and MBCs, 1.0-1.9 microM); SMAP28 and SMAP29 (MICs and MBCs, 0.2-0.7 microM); and PMAP23 (MICs and MBCs, 4.3 to > or = 6.8 microM). Overall, the results of this study suggest that synthesized cathelicidins from cattle, sheep, and pigs had broad-spectrum antimicrobial activity against all P. multocida.  相似文献
