Summary The segregation of RFLP and RAPD markers was compared in two oilseed rape (Brassica napus L.) breeding populations from the cross Topas x R4, the latter being a low linolenic mutation line. A total progeny of 68 F2 and 40 microspore derived plants were studied with 25 markers. The results indicated a significant excess of Topas alleles at five RAPD loci in the microspore derived population. This suggests that genomic regions which probably affect microspore culture ability do not have identical distribution in the two population types.  相似文献

Somatic hybridization, a process of combining protoplasts from different plants, can provide new sources of disease resistances for plants. In the case of wild and cultivatedSolanum species, the hybrids express resistances from each partner in the fusion and can often be crossed with cultivars to improve agronomic characteristics of the tubers. Restriction fragment length polymorphism (RFLP) analyses can provide a means for determining that the plants being investigated are actually hybrids as well as a means for following the introgression of DNA into progeny lines. These points are addressed in this paper with specific reference to somatic hybrids betweenSolanum brevidens and potato.  相似文献

本研究通过PCR扩增和六种限制性内切酶(AluⅠ,HinfⅠ,MboⅠ,RsaⅠ,HaeⅢ和PvuⅡ)酶切,对国内害虫防治上常用的几种昆虫病原线虫,包括斯氏属S.car-pocapsae,S.feltiae和S.glaseri以及异小杆属H.bacteriophora,H.zealandica,H.indicus和H.megidis等8个品系rDNA-ITS进行分析。建立起可以区分各线虫种的标准RFLP图谱。该方法快速简便,稳定可靠,需要的样品量少。可以用于新鲜的,或冻存的样品,甚至分析单条的线虫,不仅可进行昆虫病原线虫的快速分类鉴定。而且进一步可以应用于线虫田间释放的辅助监测。实际田间感染率的测定和线虫毒力的比较。  相似文献

 以PRSV株系特异性引物对PRSV的PRSV126(PRSV日本分离物)、Ys、Vb和Sm等株系进行RT-PCR方法鉴定,引物PR21/PR22能把Ys从Vb和Sm中鉴定出来,PR300/PR301则能把Vb从Ys、Sm和PRSV126中鉴定出来;用限制性内切酶Hae Ⅱ、Sau3A I和Hinf I对PRSV的PRSV126、Ys、Vb和Sm等株系进行单酶切RT-PCR-RFLP分析,Hinf I能把PRSV126与Ys、Vb和Sm鉴别开来,Sau3A I能把Ys与Vb和Sm鉴别开来,Hae Ⅱ则能把Ys与PRSV126、Vb和Sm鉴别开来;以P1/P2为引物,对Vb、Ys和Sm株系进行RT-PCR-RFLP-SSCP分析,结果能一次把三者较好地区别开来。  相似文献

Coating of rice seeds with fungicide Thiram improved the seed germination capability over a long period of time (11 weeks) under low temperature conditions (4 and 8°C), which simulated the sowing of rice seeds in the winter season (the farmer's slack season). To analyze the effect of Thiram on the community structure of microorganisms on the rice seed surface, culture-dependent and culture-independent (PCR-RFLP) methods were applied. PCR-RFLP patterns of 16S rDNA showed that the bacterial communities on the rice seed surface were different between coated and uncoated treatments under 8°C conditions, but that they were very similar under 4°C conditions. PCR-RFLP patterns of 18S rDNA revealed the remarkable effect of Thiram on the fungal community structure under both 4 and 8°C conditions. Although the fungal communities were quite different between coated and uncoated seeds at the beginning of incubation, the fungal communities on the coated seed surface became similar to those of uncoated seeds along with the duration of the incubation period. As the dominance percentage of Fusarium spp. among the isolates increased with the duration of the incubation period for both coated and uncoated seeds, Fusarium was considered to be a responsible for the poor germination of rice seeds that were sown in the winter season.  相似文献

以[Mo17(rf_3rf_3)CMS-唐徐×HZ_1(Rf_3Rf_3)N]×Mo17(rf_3rf_3)N回交群体作为基因定位群体。采用RFLP和RAPD分子标记技术,定位了Rf_3/rf_3基因。RFLP分析表明,Rf_3/rf_3基因位于第二染色体长臂上的UMC36A和UMC49分子标记之间,其遗传距离分别为12.7cM和4.8cM。采用BSA法,筛选了340个10mer随机引物,找到与Rf_3基因紧密连锁的分子标记RAPD Eo8-1.2,将Rf_3/rf_3基因的标记距离缩小到2.7cM,为Rf_3/rf_3基因辅助选择提供了有价值的分子标记。  相似文献

We report the tagging of a brown planthopper (BPH) resistance gene (Bph–1) in rice using RAPD and RFLP markers. The Korean rice variety ‘Gayabyeo’ has dominant duplicate genes including Bph–1 conferring resistance to biotype 1 of BPH. Bulked segregant RAPD analysis was employed for rapid identification of DNA markers linked to resistance genes. For tagging these two genes, an F2F3 population from a ‘Gayabyeo’ × ‘Nagdongbyeo’ cross was developed and evaluated for BPH resistance. Three bulked DNAs from two groups of homozygous BPH resistant (each for Bph–1 and the other unknown gene) and homozygous susceptible F2 plants were analyzed by RAPD using 140 random oligomers. One primer, OPD–7 yielded a 700-bp fragment that was present in Gayabyeo and resistant F2 plants (homozygous for Bph-1 locus) but absent in Nagdongbyeo and susceptible F2 plants. Cosegregation of this marker with Bph-1 was verified using an F2 population segregating for Bph-1. Chromosomal regions surrounding the Bph-1 were examined with additional RFLP and microsatellite markers on chromosome 12 to define the location of the RAPD marker and Bph-1. Use of this RAPD marker could facilitate early selection of resistant lines for BPH. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

 分析了来自前苏联簇毛麦及其抗病衍生系的抗白粉病基因对不同生理小种的抗性反应。用120个随机引物对6D/6V代换系Pm930640进行RAPD分析,检测到5个引物OPAN03、OPAI01、OPAL03、OPAD07和OPAG15,分别在大约1 700、700、750、480和580 bp处有区别于小麦亲本的多态性条带。对Chancellor×Pm930640 F2群体进行OPAN03、OPAI01和OPAL03等3个RAPD标记与抗白粉病基因的连锁分析,表明这些标记同簇毛麦的抗白粉病基因是连锁的。对大部分分别含有Pm1-Pm20的已知抗病基因、含有簇毛麦抗病基因及其相关亲本的29个小麦品系进行RAPD标记分析。结果表明,这些标记不仅可以鉴定簇毛麦的抗病基因,而且可以判断其遗传背景。OPAL03750仅出现在含有前苏联簇毛麦6VS染色体的抗病材料中,可作为区别于Pm21的分子标记。RFLP标记的结果也表明两个不同簇毛麦的6VS染色体有明显的多态性。  相似文献

研究了利用玉米85个 DNA 限制性片段长度多态性(RFLP)标记在玉米自交系7922与5003杂交 F2群体中的分离。结果表明,8.33%的标记显著或极显著地偏离了预期的孟德尔比例而表现偏分离,发现了2个偏分离的染色体热点,即第1染色体上的 umc128-umc107和第3染色体上的 asg24-asg48,其偏分离形成的主要原因是存在配子体选择。  相似文献

利用RFLP标记水稻亲本遗传差异及其杂种优势中的利用   总被引:2,自引:0,他引:2  
本研究以17个杂交水稻亲本、3个新株型株系和24个光壳稻、爪哇稻品种为DNA样品来源,通过RFLP(限制性片段长度多态性,Restriction Fragment Length Polymorphism,简称RFLP)标记技术,研究光壳稻和爪哇稻及其与温带粳稻之间的关系,探索RFLP标记水稻亲本遗传差异在杂交稻育种中利用的可能性。研究结果表明:42个探针共产生69个不同的限制性片段,其中20个(占47.6%)探针显示47个(占68.1%)多态性片段,至少在2个基因型间存在差异。每个具有多态性片段分别以1和0记录存在与否。由RFLP数据计算的Nei‘s遗传距离,创建聚类树状图。聚类分析结果表明,灿稻和粳稻容易被分开,温带粳稻又容易与光壳稻、爪哇稻分开,但光壳稻和爪哇稻混合聚在一起,光壳稻与温带粳稻之间的遗传距离要比爪哇稻与温带粳稻之间的遗传距离大。根据聚类图发现温带粳稻亚群内杂种优势较弱,亚群间即生态群间的杂种优势较强,群间即灿、粳亚种间杂种优势更强。利用光壳稻、爪哇稻选育不同生态群方向的恢复系和不育系,已配组育成了强优势的杂交稻组合。  相似文献
