AIM: To observe effects of homocysteine and antagonized effects of taurine on electronic leakage and free radical production in myocardial mitochondria. METHODS: Myocardial mitochondria of rat heart was isolated, and was broken by supersonic wave to prepare submitochondria. Recombinant of succinic acid cytochrome c reductase was prepared with mitochondria of porcine heart. They were co-incubated with homocysteine and/or taurine with various concentration. The H2O2 and O2- were determined by chemiluminescence methods. The taurine transporter of heart mitochondria and its propert, and effects of homocysteine on its function were studied with glass filter. RESULTS: Homocysteine stimulated oxygen free radical production in heart mitochondria, submitochondria, and succinic acid cytochrome c in a concentration-dependent manner. Although taurine itself did not affect oxygen free radical production, taurine did inhibit oxygen free radical production in mitochondria, submitochondria and succinic acid cytochrome c in a concentration-dependent manner. Taurine transporters of Na+-dependent were existed in mitochondria membrane. Homocysteine inhibited taurine transtport in mitochondria in a concentration-dependent manner. CONCLUSIONS: Taurine inhibited electronic leakage and oxygen free radical production induced by homocysteine in electron transport chain. There were taurine transporters in mitochondria membrane, and transport functions of taurine transporter were inhibited by homocysteine. 相似文献

高效液相色谱法测定海洋生物中牛磺酸的含量 总被引:1,自引:0,他引:1
[目的]建立牛磺酸的高效液相色谱分析方法,并通过该方法测定不同海洋生物中牛磺酸的含量。[方法]应用邻苯二甲醛(OPA)柱前衍生法,研究流动相比例、流速、衍生剂比例、衍生时间等因素对牛磺酸检测的影响。[结果]当牛磺酸∶OPA=1∶2,衍生时间2 min,流动相乙腈∶水=15∶85,流速为0.5 mL/min,检测波长为360 nm时,牛磺酸浓度为10~400μg/mL时具有良好的线性关系(R~20.99),平行样品的相对标准偏差为2.9%~7.4%。利用该方法检测不同海产品中牛磺酸的含量,所选择的海洋贝类、甲壳类、鱼类中均含有牛磺酸,其中海瓜子中含量最高,达11.2 mg/g。不同组织比较,鱼肉中牛磺酸含量较高,而鳃中较低。[结论]该研究结果丰富了海洋生物中牛磺酸含量的数据,为进一步研究牛磺酸在不同组织中的合成机制提供了参考。 相似文献

以毛蚶(Arco subcrenat)、紫贻贝(Mytilus edulis)、脉红螺(Rapana venosa)和四角蛤蜊(Mactra veneriformis)的软体部分为原料,经水煮法粗提牛磺酸和酶解法粗提糖胺聚糖,并比较了4种贝类中牛磺酸和糖胺聚糖的含量。结果表明,四角蛤蜊、毛蚶、脉红螺中的牛磺酸平均含量分别为2.708、3.532、3.662g/kg,以脉红螺中牛磺酸含量最为丰富;四角蛤蜊、紫贻贝、毛蚶中糖胺聚糖平均含量为8.27%、4.53%、3.87%,以四角蛤蜊的含量最高。 相似文献

探讨了僧帽牡蛎在低温贮藏保鲜过程中鲜度指标及游离氨基酸,牛磺酸等的变化。实验表明:在0-2℃及3-5℃的贮藏中挥发性盐氮呈平缓上升趋势,贮藏15天分别为15.2mg/100g和20.6mg/100g;而在6-8℃下,贮藏,挥发性盐基氮变化相对较快,15天后高达26.8mg/100g。在萃取液中,牛磺酸的含量最高,其在贮藏中略有下降,但基本上在357mg/100g-387mg/100g范围内波动;游 相似文献

AIM: To investigate the effects of taurine on lipopolysaccharide (LPS)-induced myocardial damage in rats. METHODS: Healthy male SD rats (n=30) were randomly divided into control group (CON), LPS model group (LPS) and taurine treatment group (TAU). The rats in CON group and LPS group were intravenously injected with normal saline, and the rats in TAU group were injected with taurine (100 mg/kg). After 2 h, the rats in LPS group and TAU group were intraperitoneally injected with LPS at 10 mg/kg, and the rats in CON group were injected with normal saline. Six hours after injection of LPS, the blood samples were collected for determination of superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, and tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6) levels. The myocardial tissues were processed for histological examination and the analysis of Western blot. RESULTS: Compared with CON group, LPS significantly reduced SOD activity in the serum and heme oxygenase 1 (HO-1) protein expression in the myocardial tissues, increased the serum content of MDA and levels of TNF-α and IL-6. LPS also significantly elevated the levels of TNF-α and IL-6, and up-regulated the cyclooxygenase-2 (COX-2) expression and phosphorylation of nuclear factor kappa B (NF-κB) in the myocardial tissues. Taurine pretreatment significantly elevated SOD activity and HO-1 protein expression level, decreased the levels of COX-2, TNF-α, IL-6 and phosphorylated NF-κB. Histological observation showed that taurine reduced inflammatory response in the myocardial tissue. CONCLUSION: Taurine attenuates LPS-induced myocardial damage in rats. The beneficial effects of taurine may be associated with its reduction of p-NF-κB/COX-2 signaling by activation of HO-1/CO. 相似文献

利用基于离子交换色谱原理氨基酸自动分析仪检测了一些常见食用菌中牛磺酸的含量。牛磺酸在磷酸丝氨酸及磷酸乙醇胺洗脱液中经色谱分析可得到清晰的对称峰。灵芝的牛磺酸含量为每100g干重含3.7~83umol。火菇在所检蘑菇中牛磺酸含量高,每100g湿重含(83.2±6.6)umol,与陆生植物相近。双孢蘑菇与香菇的牛磺酸含量相对较高,每100g湿重含量约49~55umol,牡蛎蘑菇及糙皮侧耳的鲜重含量分别为(8.9±0.712)umol、(3.7±0.3)umol,木耳及甘露地衣的干重含量分别为(19.5±2.1)umol、(9.3±1.5)umol。高牛磺酸、无胆固醇的蘑菇有望作为低胆甾醇类食物的替代品。 相似文献
