为明确引起羊巴贝斯虫病的病原虫莫氏巴贝斯虫临潭株(Babesia motasi Lintan,BLT)及羊巴贝斯虫未定种新疆株(Babesia sp.Xinjiang,BXJ)的核型和亲缘关系,通过DNA大片段脉冲场凝胶电泳(PFGE)和三代高通量测序对2种虫体进行核型分析和系统发育分析。结果表明:2种巴贝斯虫都有4条染色体,但基因组的大小与核型分析不一致,莫氏巴贝斯虫临潭株基因组大小为11.1 Mb,4条染色体大小分别为6.0 Mb、3.0 Mb、1.1 Mb和1.0 Mb;羊巴贝斯虫未定种新疆株基因组大小为7.0 Mb,4条染色体分别为2.4 Mb、2.0 Mb、1.7 Mb和0.9 Mb;羊巴贝斯虫未定种新疆株与牛巴贝斯虫的亲缘性较近,而莫氏巴贝斯虫临潭株与双芽巴贝斯虫的亲缘关系较近。  相似文献

AIM: To investigate the promoting effect of maxadilan, which specifically activate the type I receptor for pituitary adenylate cyclase-activating polypeptide (PACAP), on the proliferation of human induced pluripotent stem cells (IPSCs). METHODS: PACAP type I (PAC1) receptor in IPSCs was detected by RT-PCR and Western blotting. maxadilan at various concentrations was added to the medium of IPSCs as experimental groups. The medium in control group was without maxadilan treatment. The effect of maxadilan on theproliferation of IPSCs was measured with Cell Counting Kit-8 (CCK-8). The changes of cell cycle caused by maxadilan in IPSCs were analyzed by flow cytometry. The analysis of karyotype was carried out in IPSCs treated with maxadilan. Proteins and gene expression levels of both Nanog and OCT4 in IPSCs treated with maxadilan were detected by real-time quantitative polymerase chain reaction (real-time-qPCR) and immunofluorescence. The gene expression levels of Nestin and PAX6 in both IPSCs treated with maxadilan and cells of embryonic body, which was birthed from IPSCs with maxadilan treatment, were detected by real-time qPCR. The ability of IPSCs treated with maxadilan differentiating into 3 embryonic layers was evaluated by analyzing the component of embryo using RT-PCR. RESULTS: The PAC1 receptor in IPSCs was identified by RT-PCR and Western blotting. Viability of the IPSCs with 100 nmol/L maxadilan treatment was increased by 16% compared with control group. The differences with statistical significance were found in the cell viability between 100 nmol/L maxadilan treatment group and control group (P<0.05). The average values of proliferation index (PI) in IPSCs with 100 nmol/L maxadilan treatment for 3 h, 6 h and 9 h were 47.23%, 59.70% and 55.67%,respectively, while that in control group was 37.00%. The differences with statistical significance were found in PI between 100 nmol/L maxadilan treatment for 3 h group, 6 h group, 9 h group and control group (P<0.05). Normal karyotype and unchanged pluripotent state in IPSCs treated with maxadilan were observed. Compared with control group, the gene expression levels of Nestin and PAX6 were not significantly different in both IPSCs and the cells of embryonic body birthed from IPSCs with maxadilan treatment. The ability of differentiation into 3 embryonic layers in IPSCs treated with 100 nmol/L maxadilan was found. CONCLUSION: PAC1 receptor presents in IPSCs. Maxadilan promotes the proliferation of IPSCs but does not affect their pluripotent state and karyotype.  相似文献

Sex determination of bovine embryos using H-Y antibodies   总被引:3,自引:0,他引:3  
6 days old bovine embryos (n = 126) were obtained from 8 superovulated cows or heifers by flushing the uteri and oviducts either non-surgically or after slaughter. Part of the embryos (n = 72) (morula stages) were placed in Ham's F-10 or PBS supplemented with 10% fetal calf serum (FCS) diluted 1:1 with supernatant from the H-Y antibody producing clone and cultured at 38 degrees C, in 5% CO2/95% air and 100% humidity. Control embryos (n = 54) were cultured in H-Y antibody free medium. After culture the embryos could be separated into a blastocyst--and a morula group. A subsequent colchemid and hypotonic treatment and fixation and Giemsa staining allowed a precise karyotyping, and thus sex determination for 36 H-Y antibody treated embryos and 22 control embryos. The limiting factor for proper karyotyping was lack of metaphases, incomplete methaphases or poor preparation. Among the H-Y antibody treated embryos we found 7 males and 15 females in the blastocyst and 14 males and 0 females in the morula group. A statistical analysis of these proportions led to the conclusion that the H-Y antibody had a significant influence on the sex ratio.  相似文献

A diagnosis of congenital stricture of the vestibulo-vaginal fold was reached in an 11-year-old mare without previous reproductive records, during a breeding soundness examination. The mare had an annular stricture of the vestibule–vaginal fold that did not allow palpation or vaginoscopic visual examination of the anterior portion of the vagina. Endoscopic evaluation of both anterior and posterior portions of the vagina revealed a normal mucosa without scar tissue. Ultrasound examination of the cervix, uterus and ovaries did not reveal any other abnormality. The mare had a normal karyotype – 64 XX. Vestibulo-vaginal stricture or stenosis is a congenital anomaly that occurs when there is an incomplete perforation of the hymen or occurs in association with hypoplasia of the genital canal that results in the formation of an annular fibrotic stenosis at the vestibule–vaginal junction. To our knowledge, this is the first report of vestibulo-vaginal stricture in a mare with confirmed normal karyotype, 64,XX.  相似文献

用核型分析方法对甘蔗祖亲种、含两物种血缘栽培种、含三物种血缘栽培种、含四物种血缘栽培种染色体组型的系统演化特征进行了研究.从祖荣代野生种向高代杂交栽培种(含两物种血缘以上)系统演化的基本特征是:染色体数目、异染色质含量相对增加,易位现界普遍存在,中部着丝点染色体所占比例逐渐减少,核型从对称向不对称方向演化。野生种和栽培种的染色体数目分别为2n=68-92和2n=90-120;染色体约对长度为1.56-6.38μm,相对长度为1.03-4.01;染色体主要为中部着丝点染色体,少数为亚中部及亚端部着丝点染色体,未见端部着丝点染色体;除春尼为1B核型外,其余6个品种(印度割手密、墨车里本、POJ2875、Co419、F134和ROC10)全为2B类型.分别以印度割手密和春尼基因组DNA为探针,对栽培种Co419进行染色体原位杂交,结果表明,Co419(2n=114)染色体组构成中,有6条来源于割手密种,12条来源于印度种,96条为热带种或种间重组染色体.   相似文献

本研究详细阐述了早熟、高产、高糖、抗逆性强的甘蔗品种粤糖06-233的选育过程,并就其特征特性、蔗茎产量、蔗糖分、抗性等性状表现进行综合分析。以粤糖94-128为母本、桂糖93-159为父本配制杂交组合,历经8代农艺性状、糖分和产量相关性状的选择,育成了早熟、高产高糖、抗逆性强的甘蔗新品种粤糖06-233。通过国家区试和生产试验结果表明,粤糖06-233蔗茎产量达119 932.56 kg/hm 2,含糖量达18 145.79 kg/hm 2,该品种对甘蔗黑穗病具有较强的抗性。同时,利用去壁低渗法对粤糖06-233进行染色体数目与核型分析,结果表明,该品种的染色体主要由中部(m)及近中部(sm)着丝点染色体组成,染色体数为2n=88,核型公式K(2n)=88=82m+6sm。  相似文献

Bovine embryos (n = 64) collected at day-6 from 10 superovulated and inseminated donors were evaluated upon recovery and again after incubation for approx. 12 hrs. According to cleavage rate they could be divided into a fast, an intermediate and a slow developing group, respectively. This grouping was correlated to embryonic sex determined by karyotyping (n = 29). From the fast developing group we found 7 males and 2 females (male ratios 78%), from the intermediate group 3 males and 8 females (male ratio 27%) and from the slow group 1 male and 8 females (male ratio 11%). From this preliminary study it is concluded that the rate of embryonic development may be used as a sex discriminator.  相似文献

The somatic chromosomes of standard indica diploid rice, IR 36, were squashed on glass slides and stained with Giemsa. The condensation patterns (CP) of prometaphase chromosomes were quantitatively analysed using CHIAS III software. The relative length and centromeric index (CI) were converted from CHIAS III to numerical data and calculated by EXCEL program. The ideogram based on CP of indica rice was established. There were 2 pairs of satellite chromosomes and the result was confirmed by fluorescence in situ hybridization using 45S rDNA as a probe.
