为了探讨添加红枣提取物对促进虹鳟(Oncorhynchus mykiss)机体健康的可能性,选取体长为7.2~8.4 cm的健康虹鳟,随机分成4个试验组,分别投喂红枣提取物添加量为0%(对照组)、0.25%、0.50%和1.00%的等氮等脂饲料,饲养56 d,养殖结束后测定12项血清生化指标和头肾的6个免疫相关基因mRNA表达量。结果显示:与对照组相比较,血清中肌酐(CREA)和尿素氮(BUN)含量三个红枣提取物组均出现极显著下降;谷丙转氨酶(GPT)活性在0.50%和1.00%组极显著下降,谷草转氨酶(GOT)活性也在0.50%和1.00%组出现了一定程度下降,但差异不显著;酸性磷酸酶(ACP)和碱性磷酸酶(AKP)活性无显著变化;总蛋白(TP)和白介素6(IL-6)含量在0.25%组出现显著增加;溶菌酶(LZM)活性三个红枣提取物组均显著升高;超氧化物歧化酶(SOD)活性在0.50%组极显著增加;补体4(C4)和肿瘤坏死因子(TNF-α)含量在0.25%和0.50%组均极显著增加。在头肾中,与对照组相比,SOD、白介素1β(IL-1β)和补体3(C3)基因表达量在0.50%和1.00%组均为显著或极显著上调;Factor H基因表达量在0.50%组显著上调;过氧化氢酶(CAT)和LZM基因表达量在0.50%和1.00%组仅出现差异不显著上调。上述结果表明,在虹鳟饲料中添加一定量的红枣提取物,可以起到促进肾功能,保护肝脏和提高其非特异性免疫功能的作用。  相似文献

Using cDNA from a CRFK cell line as a template, PCR amplification was performed with the Ub1S and poly(dT) primers to isolate feline ubiquitin genes. Sequencing of the 495 bp PCR fragment revealed that the putative amino acids induced by this fragment gave a fusion protein consisting of a ubiquitin polypeptide (76 amino acids) and an extension protein of ribosomal proteins L40 (52 amino acids). The putative amino acid sequence of ubiquitin was identical to those of humans, rats and pigs.The recombinant glutathione S-transferase (GST)–feline ubiquitin fusion proteins were produced in Escherichia coli and purified. The fusion proteins had a molecular weight of about 42 kDa and were detected by immunoblot assay with rabbit anti-ubiquitin antiserum.The mRNAs from heat-shocked and non-heat-shocked cells were subjected to RT-PCR (Ub1S and poly(dT) primers) analysis. The molecular weights of the ubiquitinated proteins in heat-shocked CFRK cells were between 18 kDa and 24 kDa by immunoblot assay.These results suggested that there were more ubiquinated proteins in the heat-shocked CRFK cells than in the pre-heat-shocked cells.  相似文献

猪肾脏中氯丙嗪和异丙嗪残留检测方法的建立   总被引:6,自引:0,他引:6  
建立了猪肾脏中氯丙嗪和异丙嗪残留量检测的高效液相色谱法。猪肾脏中残留的氯丙嗪和异丙嗪,用乙腈提取,再依次用酸化乙腈和正己烷净化,氮气吹干,甲醇溶解后,用Waters高效液相色谱系统,HypersilC18柱,UV检测器,用V(乙腈)∶V(水)∶V(0.5mol·L-1乙酸铵)(50∶49∶1)为流动相,流速1.2mL/min,波长254nm测定,内标法定量。结果表明,氯丙嗪和异丙嗪在猪肾脏中的最低检测限均为10μg/kg,组织中添加量为10μg/kg时,氯丙嗪和异丙嗪的回收率分别为83%和84%,批间变异系数均小于20%。该方法样品处理简单,可同时检测氯丙嗪和异丙嗪在猪肾脏中的残留量。  相似文献

内毒素介导动物急性肾功能衰竭机理的试验研究   总被引:2,自引:0,他引:2  
为了阐明内毒素介导急性肾功能衰竭的机理,观察了内毒素性肾功能衰竭家兔血液凝固性变化和肾脏的病理形态学变化。与对照组家兔比较,模型组家兔血小板计数和血浆纤维蛋白原含量极显著降低(P<0 01),出血时间、凝血时间、凝血酶原时间和白陶土部分凝血活酶生成时间显著或极显著延长(P<0 05,P<0 01)。另一方面,内毒素注射在全部模型组家兔导致了弥漫性血管内凝血,71.4%的肾小球中发现了微血栓,肾小球纤维素密度指数达60 6%。此外,96.2%的肾小管上皮细胞变性坏死,84.1%的肾小管中有管型形成。对照组家兔均未发现这些病理变化。上述试验结果揭示,内毒素引发了弥漫性血管内凝血,而肾小球毛细血管内广泛形成的微血栓阻断了肾小球毛细血管血流,从而导致肾小球滤过率降低,这是内毒素介导急性肾功能衰竭的首要机理。此外,肾小管上皮细胞的变性、坏死可损害其重吸收和分泌功能,管型形成可阻塞肾小管管腔并增大肾球囊内的压力,这些也都是在内毒素介导急性肾功能衰竭发生发展过程中起重要作用的因素。  相似文献

Irisin, a novel peptide, was initially been shown to be expressed explicitly in the muscle tissues. We studied the presence of irisin immunoreactivity in porcupine adrenal glands and kidneys. Immunocytochemistry showed that irisin was localised both in the adrenal cortex and adrenal medulla. In organs, irisin immunoreactivity was found in the tubular and collecting system of the nephron. The functional role of irisin in the adrenal gland and kidney has not been precisely yet. However, irisin might have a paracrine and autocrine function as do other locally produced peptides.  相似文献

To study the effect of luteolin on blood indexes,liver and kidney in mice with acute mercury poisoning,28 mice were randomly divided into four groups:Control group (intraperitoneal injection 0.9% saline),luteolin group (lavage 100 mg/kg luteolin),mercuric chloride group (intraperitoneal injection 4 mg/kg mercury chloride) and mercury chloride+luteolin group (intraperitoneal injection 4 mg/kg mercuric chloride,lavage 100 mg/kg luteolin).The activities of ALT in serum,AST,CREA and BUN contents,blood WBC,RBC,HGB content and GSH and MDA contents of liver tissue were detected.Morphological changes of liver and kidney tissues were observed.The results showed that compared with the control group,the activities of ALT and AST of mercuric chloride group were extremely significantly increased (P < 0.01),serum CREA,BUN,blood WBC and liver tissue MDA contents were significantly increased (P < 0.05) while blood RBC,HGB and liver tissue GSH contents were significantly decreased (P < 0.05).Liver,kidney pathological changes were obviously.Compared with mercuric chloride group,the activities of ALT,AST in serum,CREA,BUN,blood WBC and liver tissue MDA contents of mercuric chloride+luteolin group were significantly decreased (P < 0.05),while blood RBC,HGB and liver tissue GSH contents were significantly elevated (P < 0.05).Liver,kidney pathological changes were attenuated obviously.The poisoning were characterized by inflammation and the occurrence of anemia when acute mercury poisoning occurred,liver and kidney showed different degrees of injury in mice.Luteolin could reduce the toxic effects of acute mercury poisoning on blood,liver and kidney.  相似文献

A variety of laboratory tests are available to assist the clinician in the assessment of kidney health and function. The majority of widely used tests are indicative of altered renal function, not becoming abnormal until function is significantly compromised. These include serum urea and creatinine concentrations, serum electrolyte concentrations, urine specific gravity (USG), fractional electrolyte excretion ratios and symmetric dimethylarginine (SDMA) concentrations. Some of these parameters may also be affected by nonrenal disease. The results of a further group of tests can indicate renal damage, but do not reflect renal function. These include urine tubular enzyme concentrations, urine protein concentrations, urine neutrophil gelatinase-associated lipocalin (NGAL) concentrations and cytological abnormalities.  相似文献

Contrast enhanced ultrasound (CEUS) is useful to evaluate tissue perfusion in the kidney. In veterinary medicine, sedation or anesthesia may be required in uncooperative or panting patients. The aim of this study was to evaluate and compare the normal kidney perfusion patterns in conscious and anesthetized dogs using CEUS. Eight healthy beagles were used in this study. Scanning was performed in conscious dogs using manual restraint (conscious group), or under general anesthesia using tiletamine-zolazepam and medetomidine (TZM group) or medetomidine (M group). The contrast agent (Sonovue®) was administered as an IV bolus. The peak intensity (PI), time to peak enhancement from injection (TTP0) and the time to peak enhancement from the initial rise (TTPup), upslope, downslope and area under the curve (AUC) were analyzed. Compared to the cortical values in the conscious group, TTP0 was significantly delayed in the TZM group, and upslope, TTP0 and TTPup were significantly different in the M group. The AUCs in the TZM and M groups were not different from those in the conscious group. The upslope of renal medullary perfusion was significantly decreased in the TZM and M groups. TTP0 and TTPup were also significantly delayed in these groups. The AUC of the medulla was significantly decreased in the M group. Therefore, TZM is useful as an anesthetic protocol when performing CEUS, and the obtained data may serve as reference values in the evaluation of renal perfusion using CEUS in dogs under anesthesia.  相似文献

旨在评价不同剂量伊曲康唑-环糊精制剂通过皮下注射途径对健康比格犬肝肾功能及血常规的影响。选用体重±10 kg的健康纯种比格犬32只,按试验要求随机分为4组,每组8只,建立稳定的生活制度。分别按照0.25 mg/kg、0.75 mg/kg和1.25 mg/kg剂量,皮下注射伊曲康唑制剂,3日1次,连续7次,对照组注射生理盐水。于试验过程中第0、5、10、15、19天采集相关样品,对犬临床表现、血常规指标、血液生化指标、尿常规指标及肝肾功能进行全面的评价。结果表明,给药组与对照组相比,除白细胞相关指标外,其余指标都在正常范围内。给药组在试验过程中均出现白细胞升高现象,但在试验结束后5 d内均恢复正常。因此,伊曲康唑-环糊精制剂作为犬皮下注射剂型,具有良好的安全性和耐受性,操作方便,动物依从性好,适合兽医临床应用。  相似文献
